      • Role of hyperforin in diabetes and its associated hyperlipidemia in rats

        The aim of the present study was to evaluate the possible roles of hyperforin against hyperglycemia, hyperlipidemia and oxidative stress in streptozotocin-induced diabetic rats. Diabetes was induced by a single intraperitoneal injection of streptozotocin (65 mg/kg). Biochemical parameters were measured following hyperforin treatment (10 mg/kg, i.p.) for 7 days. Hyperforin treatment significantly reversed the elevations in plasma glucose, triglycerides, total cholesterol and LDL-cholesterol. Hyperforin also reversed the declines in plasma HDL-cholesterol and liver glycogen, but did not reverse the change in plasma insulin levels when compared to the diabetic control rats. Hyperforin treatment also reversed the oxidative stress induced by streptozotocin. Moreover, the effect of the hyperforin on peripheral glucose utilization in normal rats was evaluated by an oral glucose tolerance test (OGTT). Hyperforin treatment significantly increased (p < 0.05) the glucose tolerance compared to the vehicle in OGTT. The antihyperglycemic, antihyperlipidemic and antioxidant activities of hyperforin (10 mg/kg, i.p.) were comparable qualitatively to glibenclamide (1 mg/kg, p.o.). In conclusion, we report for the first time through an in vivo study that hyperforin is potentially valuable for the treatment of diabetes and its associated hyperlipidemia and oxidative stress by enhancing the glucose utilization by peripheral tissues such as muscle and adipose tissues.

      • Reliability analysis of an embedded system with multiple vacations and standby

        This investigation deals with reliability and sensitivity analysis of a repairable embedded system with standby wherein repairman takes multiple vacations. The hardware system consists of ‘M’ operating and ‘S’ standby components. The repairman can leave for multiple vacations of random length during its idle time. Whenever any operating unit fails, it is immediately replaced by a standby unit if available. Moreover, governing equations of an embedded system are constructed using appropriate birth-death rates. The vacation and repair time of repairman are exponentially distributed. The matrix method is used to find the steady-state probabilities of the number of failed components in the embedded system as well as other performance measures. Reliability indexes are presented. Further, numerical experiments are carried out for various system characteristics to examine the effects of different parameter. Using a special class of neuro-fuzzy systems i.e. Adaptive Network-based Fuzzy Interference Systems (ANFIS), we also approximate various performance measures. Finally, the conclusions and future research directions are provided.

        Cloning, expression, molecular modelling and docking analysis of steroidogenic acute regulatory protein (StAR) in Clarias batrachus

        The steroidogenic acute regulatory protein (StAR) plays a key role in transferring cholesterol across the inner mitochondrial membrane. In this study, the StAR gene was isolated from the gonads of Clarias batrachus. The gene has an open reading frame of 857 bp and encodes 285 amino acids with a predicted molecular weight of 32 kDa. The signalP analysis predicted that StAR would be a non-secreted protein that lacks a signal peptide. The subcellular localization demonstrated that the presence of the StAR protein was higher in mitochondria (41.8%), followed by the nuclear region (37.1%) and cytoplasm (11.1%). The StAR protein was found to interact highly with cyp11a1, followed by the cytochrome P450 family 11 proteins and the START5 domain. The homology modelling revealed that the protein has 4 helices and twisted U-shaped 10 beta sheets numbered from αA to αD and β1 to β10, respectively. Molecular modelling analysis showed that resveratrol and eurycomanone has high binding affinity with the StAR protein. The C. batrachus StAR transcript was found to be expressed exclusively in the gonads, kidney, and liver. These results overall lay a solid foundation for understanding the structure of StAR protein in fish. The identification of 3D structures and binding sites will help in designing a structure-based drug of StAR agonists for the treatment of impaired steroidogenesis.
