Effects of Aqueous Extract of Schizandra Chinensis Fruit on Cadmium-Induced Change of Monoamine Neurotransmitters in Rats

        Zhao, Zheng Lin,Zhao, Guang Wen,Li, Li,Li, Meng Quan,Guan, Li Xin,Yang, Xu Dong,Li, Hou Zhong,Lin, Feng,Lee, Jong-Rok,Zhao, Rong Jie Korean Society of ToxicologyKorea Environmental Mu 2009 Toxicological Research Vol.26 No.1

The effects of aqueous extract of Schizandra Chinensis Fruit (AESC) on cadmium-induced changes of monoamine neurotransmitters in the different brain regions of adult rats were investigated. Male rats were received intraperitoneal (i.p.) administration of CdCl2 (0.6 mg/kg/d) for 21 days and sacrificed 7 days after the last administration. Concentrations of norepinephrine (NE), dopamine (DA) in striatum and serotonin (5-HT), 5-hydroxyindole acetic acid (5-HIAA) in cortex were measured by HPLC. There were significant decreases of NE, DA, 5-HT and 5-HIAA in Cd intoxicated rats (P < 0.05), while pretreatment with AESC (20 mg/kg/d or 60 mg/kg/d, p.o., 30 min before $CdCl_2$) greatly inhibited the decrease of monoamine transmitters, respectively (P < 0.05). Also, AESC significantly increased the reduction of glutathione contents and superoxide dismutase activities in cortex induced by $CdCl_2$. These results suggest that AESC ameliorates Cd-induced depletion of monoamine neurotransmitters in brain through its antioxidant activity.

      • Optimization of Reference Genes for Normalization of the Quantitative Polymerase Chain Reaction in Tissue Samples of Gastric Cancer

Zhao, Lian-Mei,Zheng, Zhao-Xu,Zhao, Xiwa,Shi, Juan,Bi, Jian-Jun,Pei, Wei,Feng, Qiang Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.14

For an exact comparison of mRNA transcription in different samples or tissues with real time quantitative reverse transcription-polymerase chain reaction (qRT-PCR), it is crucial to select a suitable internal reference gene. Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and beta-actin (ACTB) have been frequently considered as house-keeping genes to normalize for changes in specific gene expression. However, it has been reported that these genes are unsuitable references in some cases, because their transcription is significantly variable under particular experimental conditions and among tissues. The present study was aimed to investigate which reference genes are most suitable for the study of gastric cancer tissues using qRT-PCR. 50 pairs of gastric cancer and corresponding peritumoral tissues were obtained from patients with gastric cancer. Absolute qRT-PCR was employed to detect the expression of GAPDH, ACTB, RPII and 18sRNA in the gastric cancer samples. Comparing gastric cancer with corresponding peritumoral tissues, GAPDH, ACTB and RPII were obviously upregulated 6.49, 5.0 and 3.68 fold, respectively. Yet 18sRNA had no obvious expression change in gastric cancer tissues and the corresponding peritumoral tissues. The expression of GAPDH, ${\beta}$-actin, RPII and 18sRNA showed no obvious changes in normal gastric epithelial cells compared with gastric cancer cell lines. The carcinoembryonic antigen (CEA), a widely used clinical tumor marker, was used as a validation gene. Only when 18sRNA was used as the normalizing gene was CEA obviously elevated in gastric cancer tissues compared with peritumoral tissues. Our data show that 18sRNA is stably expressed in gastric cancer samples and corresponding peritumoral tissues. These observations confirm that there is no universal reference gene and underline the importance of specific optimization of potential reference genes for any experimental condition.

      • Feature fusion via hierarchical supervised local CCA for diagnosis of autism spectrum disorder

        Zhao, Feng,Qiao, Lishan,Shi, Feng,Yap, Pew-Thian,Shen, Dinggang Springer-Verlag 2017 Brain imaging and behavior Vol.11 No.4

Early diagnosis of autism spectrum disorder (ASD) is critical for timely medical intervention, for improving patient quality of life, and for reducing the financial burden borne by the society. A key issue in neuroimaging-based ASD diagnosis is the identification of discriminating features and then fusing them to produce accurate diagnosis. In this paper, we propose a novel framework for fusing complementary and discriminating features from different imaging modalities. Specifically, we integrate the Fisher discriminant criterion and local correlation information into the canonical correlation analysis (CCA) framework, giving a new feature fusion method, called Supervised Local CCA (SL-CCA), which caters specifically to local and global multimodal features. To alleviate the neighborhood selection problem associated with SL-CCA, we further propose a hierarchical SL-CCA (HSL-CCA), by performing SL-CCA with the gradually varying neighborhood sizes. Extensive experiments on the multimodal ABIDE database show that the proposed method achieves superior performance. In addition, based on feature weight analysis, we found that only a few specific brain regions play active roles in ASD diagnosis. These brain regions include the putamen, precuneus, and orbitofrontal cortex, which are highly associated with human emotional modulation and memory formation. These finding are consistent with the behavioral phenotype of ASD.

Effect of Circuit Parameters on Stability of Voltage-fed Buck-Boost Converter in Discontinuous Conduction Mode

Feng, Zhao-He,Gong, Ren-Xi,Wang, Qing-Yu The Korean Institute of Electrical Engineers 2014 Journal of Electrical Engineering & Technology Vol.9 No.4

The state transition matrix are obtained by solving state equations in terms of Laplace inverse transformation and Cayley-Hamilton theorem, and an establishment of a precise discrete-iterative mapping of the voltage-fed buck-boost converter operating in discontinuous conduction mode is made. On the basis of the mapping, the converter bifurcation diagrams and Lyapunov exponent diagrams with the input voltage, the resistance, the inductance and the capacitance as the bifurcation parameters are obtained, and the effect of the parameters on the system stability is deeply studied. The results obtained show that they have a great influence on the stability of the system, and the general trend is that the increase of either the voltage-fed coefficient, input voltage or the load resistance, or the decrease of the filtering inductance, capacitance will make the system stability become poorer, and that all the parameters have a critical value, and when they are greater or less than the values, the system will go through stable 1T orbits, stable 2T orbits, 4T orbits, 8T orbits and eventually approaches chaos.

Protective effect of Lactobacillus plantarum ATCC8014 on acrylamide-induced oxidative damage in rats

        Zhao Sijia,Zhao Xiaoduo,Liu Qingbo,Jiang Yujun,Li Yanhua,Feng Wenxiao,Xu Honghua,Shao Meili 한국응용생명화학회 2020 Applied Biological Chemistry (Appl Biol Chem) Vol.63 No.4

Acrylamide (AA), which is mainly found in fried foods, causes neurotoxicity, genetic toxicity, carcinogenic effects, and DNA damage. This study confirms that a strain of lactic acid bacteria (Lactobacillus plantarum ATCC8014) could alleviate the toxicity of rats by inhibiting the AA-induced oxidative damage. Forty-eight adult male SD rats were randomly divided into eight groups: control group, AA group (40 mg/kg), three different doses (1 × 107 CFU/ml, 1 × 108 CFU/ ml, 1 × 109 CFU/ml of Lactobacillus plantarum ATCC8014) of prevention groups and therapeutic groups, respectively. At the end of three-week experiment, AA treatment produced a significant reduction in the rate of weight gain along with the symptoms of hind limb splay and ataxia. Histological examinations revealed various degrees of injury in five tissues. Levels of superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH) in group AA rats were significantly decreased, but the level of lipid peroxidation (LPO) was significantly increased (p < 0.05). Both prevention and therapeutic groups with 1 × 109 CFU/ml of Lactobacillus plantarum ATCC8014 could effectively reduce the injury of AA to the body. However, reductions in both groups were not statistically significant.

      • Induction of MicroRNA-9 Mediates Cytotoxicity of Curcumin Against SKOV3 Ovarian Cancer Cells

        Zhao, Song-Feng,Zhang, Xiao,Zhang, Xiao-Jian,Shi, Xiu-Qin,Yu, Zu-Jiang,Kan, Quan-Cheng Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.8

Background: Curcumin, a phenolic compound extracted from the rhizomes of Curcuma longa, has shown cytotoxic effects against a variety of cancers. The aim of this study was to identify potential microRNA (miRNA) mediators of the anticancer effects of curcumin in ovarian cancer cells. Materials and Methods: SKOV3 ovarian cancer cells were treated with curcumin ($10-60{\mu}M$) and miR-9 expression, cell proliferation, and apoptosis were assessed. The effects of miR-9 depletion on curcumin-mediated growth suppression were also examined. Phosphorylation of Akt and forkhead box protein O1 (FOXO1) was measured in cells with miR-9 overexpression or curcumin treatment. Results: Curcumin caused a significant and dose-dependent increase of miR-9 expression in SKOV3 cells, while significantly impeding cell proliferation and stimulating apoptosis. Depletion of miR-9 significantly (p<0.05) attenuated the growth-suppressive effects of curcumin on SKOV3 cells, coupled with reduced percentages of apoptotic cells. In contrast, overexpression of miR-9 significantly enhanced the cleavage of caspase-3 and poly(ADP-ribose) polymerase and promoted apoptotic death in SKOV3 cells. Western blot analysis showed that both miR-9 overexpression and curcumin similarly caused a significant (p<0.05) decline in the phosphorylation of Akt and FOXO1, compared to untreated cells. Conclusions: The present study provided evidence that curcumin exerts its cytotoxic effects against SKOV3 ovarian cancer cells largely through upregulation of miR-9 and subsequent modulation of Akt/FOXO1 axis. Further studies are needed to identify direct targets of miR-9 that mediate the anticancer effects of curcumin in ovarian cancer cells.

Experimental and Simulation Study of PEMFC based on Ammonia Decomposition Gas as Fuel

Zhao, Jian Feng,Liang, Yi Fan,Liang, Qian Chaos,Li, Meng Jie,Hu, Jin Yi The Korean Electrochemical Society 2022 Journal of electrochemical science and technology Vol.13 No.1

Compared with hydrogen, ammonia has the advantages of high gravimetric hydrogen densities (17.8 wt.%), ease of storage and transportation as a chemical hydrogen storage medium, while its application in small-scale on-site hydrogen production scenarios is limited by the need for complex separation equipment during high purity hydrogen production. Therefore, the study of PEMFC, which can directly utilize ammonia decomposition gas, can greatly expand the application of fuel cells. In this paper, the output characteristics, fuel efficiency and the variation trend of hydrogen concentration and local current density in the anode channel of fuel cell with the output voltage of PEMFC fueled by ammonia decomposition gas were studied by experiment and simulation. The results indicate that the maximum output power of the hybrid fuel decreases by 9.6% compared with that of the pure hydrogen fuel at the same inlet hydrogen equivalent. When the molar concentration of hydrogen in the anode channel is less than 0.12, the output characteristics of PEMFC will be seriously affected. Employing ammonia decomposition gas as fuel, the efficiency corresponding to the maximum output power of PEMFC is approximately 47%, which is 10% lower than the maximum efficiency of pure hydrogen.
