        Syndecan-4 regulates ADAMTS-5 activation and cartilage breakdown in osteoarthritis

        Aggrecan cleavage by a disintegrin and metalloproteinase with a thrombospondin type 1 motif, member 5 (ADAMTS-5) is crucial for the breakdown of cartilage matrix during osteoarthritis, a degenerative joint disease that leads to the progressive destruction of articular structures. The mechanisms of ADAMTS-5 activation and their links to the pathogenesis of osteoarthritis remain poorly understood, but syndecans have been shown to be involved in the activation of ADAMTS-4 (ref. 3). Here we show that syndecan-4 is specifically induced in type X collagen–producing chondrocytes both in human osteoarthritis and in murine models of the disease. The loss of syndecan-4 in genetically modified mice and intra-articular injections of syndecan-4–specific antibodies into wild-type mice protect from proteoglycan loss and thereby prevent osteoarthritic cartilage damage in a surgically induced model of osteoarthritis. The occurrence of less severe osteoarthritis-like cartilage destruction in both syndecan-4–deficient mice and syndecan-4–specific antibody–treated wild-type mice results from a marked decrease in ADAMTS-5 activity. Syndecan-4 controls the activation of ADAMTS-5 through direct interaction with the protease and through regulating mitogen-activated protein kinase (MAPK)-dependent synthesis of matrix metalloproteinase-3 (MMP-3). Our data suggest that strategies aimed at the inhibition of syndecan-4 will be of great value for the treatment of cartilage damage in osteoarthritis.

      • Integrating Temporal and Spatial Scales: Human Structural Network Motifs Across Age and Region of Interest Size

        <P>Human brain networks can be characterized at different temporal or spatial scales given by the age of the subject or the spatial resolution of the neuroimaging method. Integration of data across scales can only be successful if the combined networks show a similar architecture. One way to compare networks is to look at spatial features, based on fiber length, and topological features of individual nodes where outlier nodes form single node motifs whose frequency yields a fingerprint of the network. Here, we observe how characteristic single node motifs change over age (12–23 years) and network size (414, 813, and 1615 nodes) for diffusion tensor imaging structural connectivity in healthy human subjects. First, we find the number and diversity of motifs in a network to be strongly correlated. Second, comparing different scales, the number and diversity of motifs varied across the temporal (subject age) and spatial (network resolution) scale: certain motifs might only occur at one spatial scale or for a certain age range. Third, regions of interest which show one motif at a lower resolution may show a range of motifs at a higher resolution which may or may not include the original motif at the lower resolution. Therefore, both the type and localization of motifs differ for different spatial resolutions. Our results also indicate that spatial resolution has a higher effect on topological measures whereas spatial measures, based on fiber lengths, remain more comparable between resolutions. Therefore, spatial resolution is crucial when comparing characteristic node fingerprints given by topological and spatial network features. As node motifs are based on topological and spatial properties of brain connectivity networks, these conclusions are also relevant to other studies using connectome analysis.</P>

        In 1950s, until the end of the Korean War, economy of the Republic of Korea has been strongly oriented to agriculture. From 1960s to 1970s, four times successive Economic Development Plan has changed its economic aspects to the labor oriented export industry one. This change is connected with big problem. One of these problems which is important for the success is education in the field of technology. Then, College of Industrial Education at Chungnam National University is assigned being Koreas first, only and unique institution to educate the teachers for Technical High Schools, thus, to contribute to a prosperous development of the nation skilled manpower in a cardinal point. The main goal of this field study is to gather useful materials and informations, relevant and meaningful for the development of curricula measurments within the field of vocational technical education. This includes not only pre-vocational education in Technical High School but also the training of technical teachers at C.I.E.. After analysing the informations and results of this field study we can find quite a lot of possible recommendations for persons and institutions involved in vocational-technical education. But we will express only a certain amount of them, 1) Put higher emphasis on basic vocational qualifications ! 2) Provide realistic perspectives towards the world of work ! 3) Link theory and practice in vocational-technical education ! 4) Develop an active qualification strategy in schools ! 5) Develop thoroughly appropriate curricula ! 6) Reduce the number of students in classes ! 7) Adapt the equipment of schools towards educational goals ! 8) Integrate vocational practice into technical education !
