Molecular networks underlying ovarian aging for clinical applications

Due to modern trends with postponing child-bearing and getting worse living environment in women, an ovarian aging increased pregnancy failure and other complications with menopause or premature ovarian failure. Although several theories have been suggested such as mitochondrial malfunction, DNA damage/repair/methylation, caloric restriction, studies regarding ovarian aging-related molecular mechanisms for development of therapeutic methods are insufficient so far. Our objective is to determine molecular pathways of ovarian aging that result in pregnancy failure and other complications in women health to develop treatment strategies. This study is consisted of two parts: in Phase I stage, we analyzed distinct gene expression profile between young and aged mouse ovaries, and in Phase II stage several preferentially expressed genes in both ovaries were selected and analyzed their physiological functions and involved molecular networks related to ovarian aging for development of diagnostic markers and therapeutic methods. Ovaries from 10 week and 11 month-old FVB/NJ female mice with synchronized estrus cycle were collected for this study. A half of each ovary was used for RNA preparation and the other half for histological analysis. Using the Illumina HiSeq 2000 System, preferentially expressed genes were identified. Functional annotation database-based gene-set enrichment analyses and Pathway Studio® were employed to evaluate aging-related molecular networks. These findings were confirmed through qRT-PCR and immunohistochemistry. To validate RNA-Seq data, we examined expression patterns of marker genes (Amh, Bmp15 and Nobox) that were wellknown to be decreased in ovarian aging process. In young or aged ovary, preferentially expressed 876 genes were identified and extracellular matrix (ECM; p<0.001) and chromatin/nucleosome-related (p<0.001) protein-coded genes have the majority in these genes by GOTERM analysis. Amongthem, we selected several candidate genes and confirmed their expression profiles by qRT-PCR and immunohistochemistry followed by molecular network analysis. Regarding molecular interactions in these genes, PathwayStudio® was employed to predict aging-involved molecular networks in mouse ovary. Here we report a couple of candidate molecular networks and medicines (chemicals) for targeting these preferentially expressed genes/proteins. Further analyses are scheduled to produce transgenic animal models and with human ovarian tissues/cell lines.

Susceptibility-weighted imaging (SWI) is well known for detecting the presence of hemorrhagic transformation, microbleeds and the susceptibility of vessel signs in acute ischemic stroke. But in some cases, it can provide the tissue perfusion state as well. We describe a case of a patient with hyperacute ischemic infarction that had a slightly hypodense, patchy lesion at the left thalamus on the initial SWI, with a left proximal posterior cerebral artery occlusion on a magnetic resonance (MR) angiography and delayed time-to-peak on an MR perfusion performed two hours after symptom onset. No obvious abnormal signals at any intensity were found on the initial diffusion-weighted imaging (DWI). On a follow-up MR image (MRI), an acute ischemic infarction was seen on DWI, which is the same location as the lesion on SWI. The hypointensity on the initial SWI reflects the susceptibility artifact caused by an increased deoxyhemoglobin in the affected tissue and vessels, which reflects the hypoperfusion state due to decreasing arterial flow. It precedes the signal change on DWI that reflects a cytotoxic edema. This case highlights that, in some hyperacute stages of ischemic stroke, hypointensity on an SWI may be a finding before the hyperintensity is seen on a DWI.
Degradation of Backsheets for Crystalline Photovoltaic Modules under Damp Heat Test

The reliability of crystalline silicon photovoltaic (PV) modules is determined not only by the efficiency loss of the silicon cells but also by the degradation of other components (ethyl vinyl acetate, ribbon, glass, and backsheet). Among these components, the backsheet provides electrical insulation and physical support, and acts as a barrier against moisture and weathering of the PV module. We investigated the degradation of different types of backsheets under damp heat conditions [95°C/85% relative humidity (R.H), 85°C/85% R.H]. During the damp heat treatment, tensile tests were carried out to investigate the mechanical degradation of the backsheets. The molecular weight of the polyethylene terephthalate (PET) in the backsheets was measured by gel permeation chromatography (GPC) to understand the intrinsic mechanism of degradation. After damp heat test, mechanical degradation of backsheet occurred significantly beyond passing the specified molecular weight region of PET film in backsheet materials. By finding the correlation between tensile strength and the molecular weight of the backsheets, our results make it possible to improve the reliability of backsheets.
Parallelism between Imperative Clauses and Interrogative Clauses in English

Inverted English imperative clauses such as Don’t you talk to everyone! share the same word order and scope interactions with interrogative clauses. This suggests that there seems to be parallelism between interrogative clauses and imperative clauses. With much emphasis focused on similarities between imperative clauses and interrogative clauses, this paper proposes that English imperative clauses undergo T-to-C movement as do English interrogative clauses. More specifically, when T and C in imperative clauses show the same matched feature with [IMP], they undergo T-to-C movement, which results in inverted imperative clauses (formal imperative clauses). On the other hand, if T and C in imperative clauses do not share the feature, there is no T-to-C movement, which yields non-inverted imperative clauses (‘economical’ imperative clauses). In other words, imperative clauses have two different types of syntactic structures whether they undergo T-to-C movement or not. We argue that two distinct structures of imperative clauses follow from the fact that T in imperatives has the mood feature [IMP] aside from C.

In recent years, discoloration defects, called as snail trail, have been observed at many crystalline photovoltaic modules after a period of time ranging from several months to several years after initial installation. It has been reported that this phenomenon doesn"t impact on the performance of photovoltaic modules, but it can be detected through simple visual inspection. The origin and detailed mechanism for the formation have not been identified. In this study, non-destructive analysis by Raman spectroscopy has been carried out to investigate the origin of this phenomenon. In parallel, destructive analysis by scanning electron microscopt and transmission electron microscopy was also performed in order to confirm the results from non-destructive method. Through the extensive analysis, it was found that the main cause for discoloration is the formations of Ag₂CO₃ and AgC₂H₃O₂. Detailed mechanism for the formation of these particles was indentified through systematic studies.
