      • Effectiveness of keratinocyte growth factor-encoding stem cells and their exosome-rich conditioned medium in hair growth

        In modern society, baldness might be one of the hot social issues, beyond the individual"s seriously-depressing problem. Keratinocyte growth factor (KGF) is a key factor for the growth of germinal matrix and differentiation. Platelet rich plasma (PRP) was reported to play an important role in angiogenesis and activation of principal cells in the skin. We established human neural stem cells (NSC) encoding KGF gene (named NSC.KGF), and collected exosome-rich conditioned medium (ERCM) was collected after 24-hour hypoxic (2% O2) culture of NSC.KGF. The hair growth-enhancing activities of stem cells and ERCM were assessed, in comparison with PRP. After shaving the backside of male C57BL/6 mice, NSC (104 cells/200 μL), NSC.KGF (104 cells/200 μL), ERCM (200 μL) or PRP (200 μL) was subcutaneously injected once at 10 sites (20 μL each). Hairs were observed from 11-12 days after shaving in the mice treated with stem cells or ERCM, earlier than the hair growth in control animals. NSC.KGF and ERCM were superior to NSC and PRP in facilitating the hair regrowth. The enhancing effects of NSC.KGF and ERCM on the number and thickness of hair follicles were also confirmed by microscopic examination. As an underlying mechanism, the stem cells and ERCM facilitated hair cycle, increasing anagen phase while decreasing telogen phase. Since the withdrawal of application of 5α-reductase inhibitors rendered hair loss reoccurs in human trials, it is expected that the KGF-encoding stem cells and their ERCM could be good candidates for the long-term improvement of hair loss including alopecia.

        Effects of Acupuncture on 1-chloro-2, 4-dinitrochlorobenzene-induced Allergic Contact Dermatitis in Mice

        Allergic contact dermatitis (ACD) is a chronic inflammatory skin disease. Topical corticosteroids are the first-line therapy for ACD despite their significant adverse effects. Acupuncture has been widely used in the treatment of various skin diseases, but its underlying mechanism remains unrevealed. In this study, we investigated the characteristics of acupuncture treatment based on effectiveness and mechanism. BALB/c mice received 1-chloro-2,4-dinitrobenzene (DNCB) application to build AD-like model. Results showed that acupuncture was an effective treatment method in inhibiting inflammatory conditions, serum IgE levels, and expression of proinflammatory cytokine Th2 (IL-4, IL-6), and Th2 (IL-1b, TNF-a) mRNA compared with DNCB treatment. Acupuncture treatment also inhibited nuclear factor-kB p65, phosphorylation of IkBa, and phosphorylation of occludin proteins expression. Furthermore, it could improve the expression of epidermal growth factor in both mRNA and protein levels. These results suggest that acupuncture, as an alternative therapy treatment for its no significant side effects, was effective in alleviating ACD by reducing proinflammatory cytokines and changing proteins’ expression.

      • Accelerated Life Test Study on Thyristors of HVDC Converter Valve

        As the key component of HVDC converter valve system, thyristors have vital impact on the reliability of the entire HVDC transmission project. In order to effectively obtain the life model of thyristor, this paper uses the existing accelerated life test theory, combined with the operating conditions of the converter valve and the relevant characteristics of the thyristors, to deeply analyse the several stress levels that have the most serious impact on the life of the thyristors. An accelerated life test scheme that can effectively guide the life model of the thyristor, according to the test data collected by this test method, the thyristor junction temperature life model under the short-circuit current stress is established, and compared with the thyristor failure rate in the practical engineering, the results show the feasibility of the test scheme and the correctness of the life model.

        Experimental study and analysis on physicochemical properties of coal treated with clean fracturing fluid for coal seam water injection

        In the process of coal seam water injection, the components of different fracturing fluids have differenteffects on coal. To analyze the effect of water injection fracturing fluid on the physical and chemical propertiesof coal, we studied the relationship between guar-based fracturing fluid and cationic clean fracturingfluid (C-VES). Microscopic experiments show that guar-based fracturing fluid has a certain viscosityeffect on coal, while C-VES can adsorb and capture the primary coal dust in coal. FTIR analysis shows thatC-VES has a great change on the content of coal functional groups, and guar-based fracturing fluid haslittle effect on coal. Industrial analysis shows that C-VES can better reduce the ash content of coal andhas a certain impact on the thermal efficiency of coal than guar based fracturing fluid. Combustion characteristicexperiment shows that guar-based fracturing fluid does not affect the combustion effect of coal,and C-VES can expand the exothermic temperature range of coal but has little effect on the main combustionof coal. Molecular dynamics simulation results show that C-VES has the best wetting effect oncoal. This research provides theoretical guidance for dust prevention and improving the use and conversionefficiency of coal.

        Aqueous Extracts of Se-Enriched Auricularia auricular Exhibits Antioxidant Capacity and Attenuate Liver Damage in High-Fat Diet/Streptozotocin-Induced Diabetic Mice

        Oxidative stress triggered by hyperglycemia is thought to be a major factor in the development of liver disease during diabetes mellitus (DM). The aim of this study was to determine the antioxidant capacity of aqueous extracts of selenium-enriched Auricularia auricular (AESA) and further investigate the hepatic protection and potential mechanism of AESA in a mouse model of diabetes. An in vitro antioxidant assay confirmed that AESA exhibited better antioxidant capacity characterized by increased reducing power and scavenging capacity of free radicals, such as diphenyl picrylhydrazyl radical, hydroxyl radical, and superoxide radical. The diabetic model was induced by high-fat diet combined with a single injection of streptozotocin in C57BL/6 mice. Our results showed that AESA treatment improved diabetes-induced disorders of lipid metabolisms and alleviated liver damage in diabetic mice. Furthermore, the antioxidant enzyme activities of glutathione peroxidase and catalase in liver were increased and malondialdehyde level was decreased with AESA treatment compared with those in the DM group. In parallel, AESA significantly reduced the contents of tumor necrosis factor-alpha and interleukin-1beta in liver in comparison with the DM group. In addition, western blot results showed that the expression of receptor for advanced glycation end products (RAGE), phospho-c-Jun NH2-terminal kinase, phospho-extracellular signal-regulated kinase, and phospho-p38 kinase were remarkably decreased in AESA treatment group compared with DM group. Taken together, supplementation of AESA may effectively attenuate diabetic hepatopathology by exerting antioxidant function through the RAGE/mitogen-activated protein kinase pathway.

        DK-MGAR101, an extract of adventitious roots of mountain ginseng, improves blood circulation by inhibiting endothelial cell injury, platelet aggregation, and thrombus formation

        Background: Since ginsenosides exert an anti-thrombotic activity, blood flow-improving effects of DK-MGAR101, an extract of mountain ginseng adventitious roots (MGAR) containing various ginsenosides, were investigated in comparison with an extract of Korean Red Ginseng (ERG). Methods: In Sprague-Dawley rats orally administered with DK-MGAR101 or ERG, oxidative carotid arterial thrombosis was induced with FeCl<sub>3</sub> (35%), and their blood flow and occlusion time were measured. To elucidate underlying mechanisms, the cytoprotective activities on rat aortic endothelial cells (RAOECs) exposed to hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>) were confirmed. In addition, the inhibitory activities of DK-MGAR101 and ERG on agonist-induced platelet aggregation, thromboxane B<sub>2</sub> production, and ATP granule release from stimulated platelets as well as blood coagulation were analyzed. Results: DK-MGAR101 containing high concentrations of Rb1, Rg1, Rg3, Rg5, and Rk1 ginsenosides (55.07 mg/g) was more effective than ERG (ginsenosides 8.45 mg/g) in protecting RAOECs against H<sub>2</sub>O<sub>2</sub> cytotoxicity. DK-MGAR101 was superior to ERG not only in suppressing platelet aggregation, thromboxane B<sub>2</sub> production, and granule release, but also in delaying blood coagulation, FeCl<sub>3</sub>-induced arterial occlusion, and thrombus formation. Conclusions: The results indicate that DK-MGAR101 prevents blood vessel occlusion by suppressing platelet aggregation, thrombosis, and blood coagulation, in addition to endothelial cell injury.

        Two Typical Synoptic‑Scale Weather Patterns of Dust Events over the Tibetan Plateau

        Synoptic-scale weather systems play dominant roles in inducing high tropospheric dust over the Tibetan Plateau (TP). However, few studies have summarized the typical synoptic-scale weather patterns when high tropospheric dust occursover the TP, as well as the difference between the distribution and transport methods of dust under weather patterns. Basedon dust optical depth (DOD) from remote sensing data and reanalysis data during 2000 to 2019, two typical synoptic-scaleweather patterns (T1 and T2) in the middle troposphere in association with high DOD in spring over the TP were obtainedby using the self-organizing map (SOM) clustering method. The results show that the T1 features a deep trough over theAltai Mountains and the westerly wind increases over the TP. As a result, dust is transported from the Taklimakan Desertand Qaidam Basin to the upper troposphere and extends to the TP and northern China. T2 shows a low-pressure system overthe western TP and decreased westerly winds over the TP, resulting in dust from the Taklimakan Desert, Qaidam Basin, andwestern TP to downstream areas. T1 (T2) contributes more to DOD over the eastern (western) TP. Therefore, we believe thata small increase (decrease) in DOD in the western (eastern) part of the TP from 2000 to 2019 may be related to an increase(decrease) in the occurrence of the T2 (T1). This work may provide a new possibility for projecting dust transport and itsinfluence on tropospheric dust over the TP.
