Two New Species of Placolecis (Lichenized Ascomycota) from China

Two new species of the lichen genus Placolecis are discovered in China, namely P. kunmingensis An. C. Yin & Li S. Wang and P. sublaevis An. C. Yin & Li S. Wang. The new combination P. loekoesiana (S.Y. Kondr., Farkas, J.J. Woo & Hur) An. C. Yin is proposed. Placolecis kunmingensis is characterized by having simple, spherical or ellipsoid, hyaline spores, and pear-shaped pycnidia; while P. sublaevis can be distinguished by its thallus forming larger aggregations with slightly flattened lobes at the thallus margin, and urn-shaped pycnidia. Descriptions, a phylogenetic tree and a key are provided for all the known Placolecis species in China.
Study of PD Location in Generators by PD Pulses Propagation

When a partial discharge takes place at the stator of a generator, the electrical pulse will propagate along the stator bars and the capacitor chains formed by the end part of the stator winds. On the first path, the pulse propagates as a travel wave at slow speed. On the second path, the pulse propagates at quick speed. Based on the data of the experiments on a real 50 MW steam generator, the author has found the pulses can propagate by magnetic field of the stator winding. It was studied that how to locating the partial discharge by signals coming from the different paths, including the features of signals on the two paths at time domain and frequency domain, the measurement frequency rang of the signals, the blind area, the advantage and disadvantage of this method.

The Glu/Asp<sup>7.32</sup> residue in extracellular loop 3 of the mammalian type-I gonadotropin-releasing hormone receptor (GnRHR) interacts with Arg<sup>8</sup> of GnRH-I, conferring preferential ligand selectivity for GnRH-I over GnRH-II. Previously, we demonstrated that the residues (Ser and Pro) flanking Glu/Asp<sup>7.32</sup> also play a role in the differential agonist selectivity of mammalian and non-mammalian GnRHRs. In this study, we examined the differential antagonist selectivity of wild type and mutant GnRHRs in which the Ser and Pro residues were changed. Cetrorelix, a GnRH-I antagonist, and Trptorelix-2, a GnRH-II antagonist, exhibited high selectivity for mammalian type-I and non-mammalian GnRHRs, respectively. The inhibitory activities of the antagonists were dependent on agonist concentration and subtype. Rat GnRHR in which the Ser-Glu-Pro (SEP) motif was changed to Pro-Glu-Val (PEV) or Pro-Glu-Ser (PES) had increased sensitivity to Trptorelix-2 but decreased sensitivity to Cetrorelix. Mutant bullfrog GnRHR-1 with the SEP motif had the reverse antagonist selectivity, with reduced sensitivity to Trptorelix-2 but increased sensitivity to Cetrorelix. These findings indicate that the residues flanking Glu<sup>7.32</sup> are important for antagonist as well as agonist selectivity.
Fast Switching Direct Torque Control Using a Single DC-link Current Sensor

This paper presents a fast switching direct torque control (FS-DTC) using only a single DC-link current sensor. In FS-DTC, six new active voltage complex space vectors (CSVs) are synthesized by the conventional active voltage space vectors (SVs). The corresponding sectors are rotated in the anticlockwise direction by 30 degrees. A selection table is defined to select the CSVs. Based on the "Different Phase Mode", the output sequence of the selected CSV is optimized. Accordingly, a reconstruction method is proposed to acquire the phase currents. The core of the FS-DTC is that all of the three phase currents can be reliably reconstructed during every two sampling periods, which is the result of the fast switching between different phases. The errors between the reconstructed and actual currents are strictly limited in one sampling period. The FS-DTC has the advantages of the standard DTC scheme such as simple structure, quick torque response and robustness. As can be seen in the analysis, the FS-DTC can be thought of as an equivalent standard DTC scheme with 86.6% of the maximum speed, 173.2% of the torque ripple, and 115% of the response time of the torque. Based on a dSPACE DS1103 controller, the FS-DTC is implemented in an induction machine drive system. The results verify the effectiveness of the FS-DTC.

<P>We synthesized a diosgenyl saponin bearing a unique disaccharide from the natural product β-hederin, together with twelve glycosylated derivatives and determined their cytotoxicity against five different human cancer cell lines. Most of them showed weak cytotoxicity, with the exception of compound <B>20</B>, diosgenyl α-<SMALL>L</SMALL>-rhamnopyranosyl-(1→2)-[α-<SMALL>L</SMALL>-arabinopyranosyl-(1→4)]-α-<SMALL>L</SMALL>-arabinopyranoside, which exhibited strong cytotoxicity against A549 cells. The cytotoxicity of <B>20</B> was associated with apoptotic cell death, which was characterized by morphological changes, chromatin condensation, DNA fragmentation, and phosphatidylserine externalization. Compound <B>20</B> induced apoptosis of A549 cells through a caspase-8-mediated extrinsic pathway and a caspase-9-mediated intrinsic pathway. In addition, phosphorylation of JNK increased but the phosphorylation of ERK decreased after treatment with <B>20</B>. Glutathione peroxidase (GPX) is one of the important members of the antioxidant enzyme family. It can catalyze the reduction of hydroperoxides with glutathione to protect cells against oxidative damage. Single-chain variable fragment (scFv) can be converted into seleniumcontaining single-chain variable fragment (Se-scFv) by chemical modification of the hydroxyl groups in scFv, thus Se-scFv possesses GPX activity and becomes a prodrug. To improve the expression of scFv and simplify its purification steps, Single-protein production (SPP) system was used to express scFv and chemical modification was used to synthesize Se-scFv. Therefore, we must construct a new scFv-WCD1-lessACA gene, which can express its mRNA not containing any ACA sequences and express its amino acid sequence of target protein (scFv) being same to scFv-WCD1. In this way, the scFv-WCD1-lessACA can be only expressed in SPP system and no other background proteins in the cells could be expressed. The expression results showed that high level of scFv-WCD1-lessACA synthesis was at least sustained for 96 h in the virtual absence of background protein synthesis. Then, selenocysteine (Sec) was incorporated into the scFv-WCD1-lessACA by chemical modification and resulted in Se-scFv-WCD1-lessACA. The enzymatic characteristics of Se-scFv-WCD1-lessACA were determined. GPX activity was 2,563 U/μmol,its binding constant for GSH was 0.687 ×105/mol. Moreover,Se-scFv-WCD1-lessACA was confirmed to have a strong antioxidant ability to protect mitochondria against oxidative damage induced by Vc/Fe2+ (mitochondrial damage model),suggesting that Se-scFv-WCD1-lessACA has potential application for protection of mitochondrial damage induced by reactive oxygen species (ROS).

The work proposes a new design tool that integrates design-around concepts with the algorithm for inventive problem solving (Russianacronym: ARIZ). ARIZ includes a complete procedure for analyzing problems and related resource, resolving conflicts and generatingsolutions. The combination of ARIZ and design-around concepts and understanding identified principles that govern patent infringementscan prevent patent infringements whenever designers innovate, greatly reducing the cost and time associated with the productdesign stage. The presented tool is developed from an engineering perspective rather than a legal perspective, and so can help designerseasily to prevent patent infringements and succeed in innovating by designing around. An example is used to demonstrate the proposed method.

Ethylene performs an important function in plant growth and development. 1-aminocyclopropane-1-carboxylate (ACC) synthase (ACS), the key enzyme involved in ethylene biosynthesis, has been the focus of most ethylene studies. Here, a cotton ACS gene referred to as Gossypium hirsutum ACS1 (GhACS1), was isolated. The full-length cDNA of GhACS1 encodes for a 476-amino acid protein which harbors seven conserved regions, 11 invariant amino acid residues, and the PLP binding active site, all of which characterize ACC synthases. Alignment analysis showed that GhACS1 shared a high degree of identity with other known ACC synthases from different species. Two introns were detected in the genomic DNA sequence, and the results of Southern blot analysis suggested that there might be a multi-gene family encoding for ACC synthase in cotton. From the phylogenetic tree constructed with 24 different kinds of ACC synthases, we determined that GhACS1 falls into group II, and was closely associated with the wound-inducible ACS of citrus. The analysis of the 5' flanking region of GhACS1 revealed a group of putative cis-acting elements. The results of expression analysis showed that GhACS1 displayed its transient expression nature after wounding, abscisic acid (ABA), and $CuCl_2$ treatments. These results indicate that GhACS1, which was transiently expressed in response to certain stimuli, may be involved in the production of ethylene for the transmission of stress signals.
