      • Preparation of 5-fluorouracil-loaded Nanoparticles and Study of Interaction with Gastric Cancer Cells

        Aims: To prepare 5-fluorouracil (5-Fu) nanoparticles with higher encapsulation efficiency and drug loading, and then investigate interaction with the SGC-7901 gastric cancer cell line. Materials and Methods: Prescription was optimized by orthogonal experiments, the encapsulation efficiency and loading capacity were tested by high-performance liquid chromatography, and inhibition of proliferation by 5-Fu nanoparticles and 5-Fu given to cells for 24, 48 and 72 hours was investigated by methyl thiazolyl tetrazolium assay (MTT). In addition, 5-Fu nanoparticles were labeled by fluorescein isothiocyanate (FITC), and absorption into cells was tested by flow cytometry. Results: The optimal conditions for preparation were concentrations of 5-Fu of 5mg/ml, of $CaCl_2$ of 60 mg/ml and of chitosan of 2 mg/ml. With a stirring speed of 1200rpm, encapsulation efficiency of 5-Fu nanoparticles was $55.4{\pm}1.10%$ and loading capacity was $4.22{\pm}0.14%$; gastric cancer cells were significantly inhibited by 5-Fu nanoparticles in a time and concentration dependent manner, and compared to 5-Fu with slower drug release, in a certain concentration range, inhibition with 5-Fu nanoparticles was stronger. 5-Fu nanoparticles were absorbed by the cells in line with the concentration. Conclusions: 5-Fu nanoparticles can inhibit growth of gastric cancer cells in vitro to a greater extent than with 5-Fu with good adsorption characteristics, supporting feasibility as a carrier.

        Association of a novel antisense lncRNA TP73-AS1 polymorphisms and expression with colorectal cancer susceptibility and prognosis

        Background: TP73-AS1 is a novel antisense long noncoding RNA and plays an important role in cell proliferation and cancer development. However, the link between TP73-AS1 and colorectal cancer (CRC) has not yet been reported. Objective: To explore the association of genetic variants in TP73-AS1 and its expression with CRC susceptibility and prognosis. Methods: A case-control study (including 507 CRC cases and 503 controls) and bioinformatics analysis were conducted. Results: rs9800 polymorphism was significantly related to higher risk in CRC [adjusted odds ratio (AOR) = 1.33, 95% confidence interval (CI) = 1.02-1.75, P = 0.034 in heterozygote codominant model]. There was no difference between TP73-AS1 polymorphisms and different tumor node metastasis (TNM) stages in the adjusted model. Moreover, TP73-AS1 expression level was positively related to different TNM stages. After adjusted for age, gender and TNM, higher TP73-AS1 expression levels were related to shorter recurrence-free survival time [hazard ratio (HR) = 1.66, 95% CI = 1.02-2.71, P = 0.043]. Conclusion: TP73-AS1 polymorphisms and expression may be associated with susceptibility and prognosis of CRC.

        SEDs and Beaming Effect for Fermi Blazars

        In this work, based on our previous calculations of spectral energy distributions for a sample of Fermi blazars (Fan et al. 2015a), we calculated the radio loudness and performed correlation analyses. Our analysis results show that radio loudness is closely anti-correlated with synchrotron peak frequency and positively correlated with gamma-ray luminosity, suggesting that the gamma-ray emissions are strongly beamed.

        Decentralized Fault Tolerant Control of Modular Manipulators System Based on Adaptive Dynamic Programming

        This paper presented an original decentralized fault tolerant control approach for modular manipulators which based on adaptive dynamic programming (ADP) algorithm. First, the dynamic model of modular manipulators is established via joint torque feedback technique. Then, the fault tolerant controller is designed which composes of model-based compensation controller, observer-based fault tolerant controller and ADP-based optimal controller. According to ADP algorithm, the Hamiltonian-Jacobi-Bellman (HJB) equation can be tackled by critic neural network (NN). The closed-loop modular manipulators system is guaranteed asymptotic stable based on Lyapunov theory. Experiments are performed to verify the proposed method, and the results have guaranteed its effectiveness.

        Study on Leading-phase Operation Capability of a 770 MW Jumbo Hydro-generator based on Stability Analysis and End-Region Heat Analysis

        A generator-grid coupling calculation model is established to study the leading-phase operational capability of a 770 MW jumbo hydro-generator in a Chinese ultra-mega hydropower station. The static and dynamic stability of the generator are analyzed and calculated to obtain stability limits under leading-phase operating conditions. Three-dimensional (3D) time-varying nonlinear moving electromagnetic and temperature field models of the generator end-region are also established and used to determine the magnetic field, loss, and temperature of the end-region under the leading-phase operating condition. The simulation results agree with data measured from the actual 770 MW hydro-generator. This paper provides reliable reference data for the leading-phase operation of a jumbo hydro-generator, which will help to improve in the design and manufacture of future hydro-generators.

        Effect of Shifting the Pole-shoe and Damper-bar Centerlines on the No-load Voltage Waveform of a Tubular Hydro-generator

        This study optimises the no-load voltage waveform of tubular hydro-generators by a simple design scheme. For different centerlines of the pole shoe and damper bar, the optimisation effects on the no-load voltage waveform are investigated in two tubular hydro-generators with different weighted powers (34 MW and 18 MW). The results are compared with those of the traditional stator-slots skewed design. The quality of the no-load voltage waveform was related to the shifting degree, and the different optimisation effects between the integer slot generator (q = 2) and the fractional slot generator (q = 11/2) were analysed. This research can improve the quality of the power output and no-load voltage waveform, and provide an effective reference for improving the industrial design and manufacture level of tubular hydro-generators.

      • IL-35 Over-expression is Associated with Genesis of Gastric Cancer

        Overexpression of interleukin (IL)-35 has been found in a variety of malignancies, but the expression status in gastric cancer has yet to be elucidated clearly. In the present study, positive expression of EBI3 and p35 was 63.3% and 70.0% of cases, respectively. EBI3 expression was strongly related with larger tumor size and invasion depth (P<0.05). Similarly, expression of p35 was also correlated with larger tumor size (P<0.05). These results indicate that IL-35 might be involved in growth of gastric cancer. Interestingly, EBI3 and p35 expressions were positive correlated with Ki-67 expression. Moreover, EBI3 immunoreactivity was associated with Bcl-2 staining. Our data suggest IL-35 is correlated with genesis of gastric cancer by regulating growth and apoptosis.

        Adsorption Behavior of Environmental Hormone Bisphenol A onto Mesoporous Silicon Dioxide

        Mesoporous silicon dioxide (meso-$SiO_2$) was prepared using cetyltrimethylammonium bromide as the structure-directing reagent and tetraethyl orthosicate as the silicon source. The influence of pH value on the adsorption behavior of bisphenol A (BPA) was investigated. The adsorption capacity of BPA onto meso-$SiO_2$ increases slightly with pH value from 2 to 6, and then gradually decreases as further improving pH value. The effect of temperature was also studied, and the adsorption capacity of BPA gradually declines with increasing temperature. The adsorption kinetics and thermodynamics of BPA were examined. It is found that the adsorption of BPA onto meso-$SiO_2$ is in good agreement with Langmuir adsorption model. The rate constant of adsorption is $5.17{\times}10^{-3}g\;mg^{-1}\;min^{-1}$, and the maximum adsorption capacity is as high as 353.4 $mg\;g^{-1}$ at 20 $^{\circ}C$.

        Assessment of Cortical Visual Impairment in Infants with Periventricular Leukomalacia: a Pilot Event-Related fMRI Study

        Objective: We wanted to investigate the usefulness of event-related (ER) functional MRI (fMRI) for the assessment of cortical visual impairment in infants with periventricular leukomalacia (PVL). Materials and Methods: FMRI data were collected from 24 infants who suffered from PVL and from 12 age-matched normal controls. Slow ER fMRI was performed using a 3.0T MR scanner while visual stimuli were being presented. Data analysis was performed using Statistical Parametric Mapping software (SPM2), the SPM toolbox MarsBar was used to analyze the region of interest data, and the time to peak (TTP) of hemodynamic response functions (HRFs) was estimated for the surviving voxels. The number of activated voxels and the TTP values of HRFs were compared. Pearson correlation analysis was performed to compare visual impairment evaluated by using Teller Acuity Cards (TAC) with the number of activated voxels in the occipital lobes in all patients. Results: In all 12 control infants, the blood oxygenation level-dependent (BOLD) signal was negative and the maximum response was located in the anterior and superior part of the calcarine fissure, and this might correspond to the anterior region of the primary visual cortex (PVC). In contrast, for the 24 cases of PVL, there were no activated pixels in the PVC in four subjects, small and weak activations in six subjects, deviated activations in seven subjects and both small and deviated activations in three subjects. The number of active voxels in the occipital lobe was significantly correlated with the TAC-evaluated visual impairment (p < 0.001). The mean TTP of the HRFs was significantly delayed in the cases of PVL as compared with that of the normal controls. Conclusion: Determining the characteristics of both the BOLD response and the ER fMRI activation may play an important role in the cortical visual assessment of infants with PVL.
