        SUV39H1 is a New Client Protein of Hsp90 Degradated by Chaetocin as a Novel C-Terminal Inhibitor of Hsp90

        Hsp90 is often overexpressed with activated form in cancer cells, and many key cellular proteins are dependent upon the Hsp90 machinery (these proteins are called “client protein”). Nowadays, more client proteins and more inhibitors of Hsp90 are being discovered. Chaetocin has been identified as an inhibitor of histone methyl transferase SUV39H1. Herein, we find that Chaetocin is an inhibitor of Hsp90 which binds to the C-terminal of Hsp90α. Chaetocin inhibited a variety of Hsp90 client proteins including AMl1-ETO and BCL-ABL, the mutant fusion-protein in the K562 and HL-60 cells. SUV39H1 mediates epigenetic events in the pathophysiology of hematopoietic disorders. We found that inhibition of Hsp90 by Chaetocin and 17-AAG had ability to induce degradation of SUV39H1 through proteasome pathway. In addition, SUV39H1 interacted with Hsp90 through co-chaperone HOP. These results suggest that SUV39H1 belongs to a client protein of Hsp90. Moreover, Chaetocin was able to induce cell differentiation in the two cells in the concentration range of Hsp90 inhibition. Altogether, our results demonstrate that SUV39H1 is a new client protein of Hsp90 degradated by Chaetocin as a novel C-terminal inhibitor of Hsp90. The study establishes a new relationship of Chaetocin and SUV39H1, and paves an avenue for exploring a new strategy to target SUV39H1 by inhibition of Hsp90 in leukemia.

      • Efficient Zero-Knowledge Proofs of Knowledge of Double Discrete Logarithm

        Zero-knowledge proof protocol is a basic cryptographic technique. And zero-knowledge proof of double discrete logarithm has some particular properties, so it has been widely applied in many security systems. But the efficient problem of zero-knowledge proof of double discrete logarithm has not been solved to this day, since there are some special difficulties in computing this kind of knowledge proof. Hence, the time complexity and the space complexity of existing schemes are all O(k), where k is a security parameter. After redesigning the basic construction of knowledge proof, we provide a new zero-knowledge proof of double discrete logarithm, which is the first scheme with O(1) time complexity and O(1) space complexity. If introducing an off-line TTP (trusted third party), we can provide two additional zero-knowledge proof schemes of double discrete logarithm, one is even more efficient than the first one, the other one solves another open problem, which is how to efficiently prove the equality of double discrete logarithms in zero-knowledge way, and the existing techniques cannot solve this problem. We also provide the detailed security proofs of our designs and efficiency analysis, comparing with the existing schemes. The significant improvement in efficiency of this basic cryptographic technique is also helpful for many security systems.

        Compact E-Cash with Practical and Complete Tracing

        E-cash has its merits comparing with other payment modes. However, there are two problems, which are how to achieve practical/complete tracing and how to achieve it in compact E-cash. First, the bank and the TTP (i.e., trusted third party) have different duties and powers in the reality. Therefore, double-spending tracing is bank’s task, while unconditional tracing is TTP’s task. In addition, it is desirable to provide lost-coin tracing before they are spent by anyone else. Second, compact E-cash is an efficient scheme, but tracing the coins from double-spender without TTP results in poor efficiency. To solve the problems, we present a compact E-cash scheme. For this purpose, we design an embedded structure of knowledge proof based on a new pseudorandom function and improve the computation complexity from O(k) to O(1). Double-spending tracing needs leaking dishonest users’ secret knowledge, but preserving the anonymity of honest users needs zero-knowledge property, and our special knowledge proof achieves it with complete proofs. Moreover, the design is also useful for other applications, where both keeping zero-knowledge and leaking information are necessary.

        Low cycle fatigue properties of hydrogenated welding sheets of Zr–Sn–Nb alloy using funnel-shaped flat specimens

        Low cycle fatigue tests on the hydrogenated welding seam of ZreSneNb alloy at room temperature and 360 C had been carried out by using the funnel-shaped flat specimens. The relationships between nominal stress & strain directly measured across the funnel and local stress & strain at the root of the funnel are given by considering cyclic plasticity correction. The results show that the fatigue resistance of welding seam at room temperature is only slightly better than that at 360 C. Probabilistic fatigue life curves are obtained by using a two-parameter power function.

        Theoretical Investigation on Collinear Phase Matching Stimulated Polariton Scattering Generating THz Waves with a KTP Crystal

        We present a theoretical research concerning terahertz (THz) wave generation with KTiOPO 4 (KTP) by collinear phase matching (CPM) stimulated polariton scattering (SPS). Both CPM and corresponding nonzero nonlinear coefficients can be simultaneously realized with s→f+f in yz plane, s→f+s with θ < Ω in xz plane and s→f+f with θ > Ω in xz plane. The effective nonlinear coefficients including electronic nonlinearities and ionic nonlinearities are calculated. Based on the parameter values of refractive indices, absorption coefficients and effective nonlinear coefficients, we simulate THz wave intensities generated with CPM SPS by solving coupled wave equations and give the relationship among the maximum THz wave intensity, optimal crystal length and the angle θ. The calculation results demonstrate that CPM SPS with KTP can generate THz waves with high intensities and quantum conversion efficiencies.

        Thermal-structural coupling analysis of motor rotor under extreme variable working conditions

        Extreme variable working conditions were proposed, and the thermal-structural coupling analysis of the rotor was conducted with an electric drive system as the research object to make the structural strength of the rotor of a vehicle motor satisfy all operating conditions as much as possible. The overall temperature rise characteristics of the electric drive system were obtained through simulation, and the accuracy of simulation results was verified through testing. On the basis of material tests at different temperatures, the interference ft, and the rotating centrifugal force as loads, a comparative analysis of the strength of the rotor was conducted for variable working conditions and single working conditions. Results show that the stress value in the variable working condition is 74.9 MPa greater than that in the single working condition. This finding indicates that the analysis of rotor stresses under variable working conditions has more engineering importance.

        LINC00943 knockdown exerts neuroprotective effects in Parkinson’s disease through regulates CXCL12 expression by sponging miR-7-5p

        Background Parkinson’s disease (PD) is a common neurodegenerative movement disorder, but the pathogenesis is still unclear. Long non-coding RNAs (lncRNAs) have been reported to play a prominent role in PD. Objective This study is designed to explore the role and mechanism of long intergenic non-coding RNA 00943 (LINC00943) in the N-methyl-4-phenylpyridine (MPP+)-inducted PD model. Methods LINC00943, microRNA-7-5p (miR-7-5p), and the chemokine (C-X-C motif) ligand 12 (CXCL12, also referred to as SDF-1) level were examined by real-time quantitative polymerase chain reaction (RT-qPCR). Cell viability and apoptosis were analyzed by 3-(4, 5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetrazolium bromide (MTT), and fow cytometry assays, severally. Protein levels of interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and CXCL12 were assessed by western blot assay. The ROS generation and SOD activity were detected by the corresponding kits. The binding relationship between miR-7-5p and LINC00943 or CXCL12 was predicted by Starbase and then verifed by a dual-luciferase reporter and RNA Immunoprecipitation (RIP) assays. Results LINC00943 and CXCL12 were increased, and miR-7-5p was decreased in MPP+-inducted SK-N-SH cells. LINC00943 silencing promoted cell viability, and repressed apoptosis and the infammatory response in MPP+-treated SK-N-SH cells. The mechanical analysis discovered that LINC00943 acted as a sponge of miR-7-5p to regulate CXCL12 expression. Conclusions LINC00943 knockdown could attenuate MPP+-triggered neuron injury by regulating the miR-7-5p/CXCL12 axis, hinting at a promising therapeutic target for PD treatment.

        haracterization of a Recombinant Thermostable Xylanase from Hot Spring Thermophilic Geobacillus sp. TC-W7

        A xylanase-producing thermophilic strain, Geobacillus sp. TC-W7, was isolated from a hot spring in Yongtai (Fuzhou, China). Subsequently, the xylanase gene that encoded 407 amino acids was cloned and expressed. The recombinant xylanase was purified by GST affinity chromatography and exhibited maximum activity at 75℃ and a pH of 8.2. The enzyme was active up to 95℃ and showed activity over a wide pH range of 5.2 to 10.2. Additionally, the recombinant xylanase showed high thermostability and pH stability. More than 85% of the enzyme`s activity was retained after incubation at 70℃ for 90 min at a pH of 8.2. The activity of the recombinant xylanase was enhanced by treatment with 10 mM enzyme inhibitors (DDT, Tween-20, 2-Me, or TritonX-100) and was inhibited by EDTA or PMSF. Its functionality was stable in the presence of Li+, Na+, and K+, but inhibited by Hg2+, Ni2+, Co2+, Cu2+, Zn2+, Pb2+, Fe3+, and Al3+. The functionality of the crude xylanase had similar properties to the recombinant xylanase except for when it was treated with Al2+ or Fe2+. The enzyme might be a promising candidate for various industrial applications such as the biofuel, food, and paper and pulp industries.

        Chinese herbal injections for coronavirus disease 2019 (COVID-19): A narrative review

        Background The outbreak of Coronavirus disease 2019 (COVID-19) has caused more than 180 million infections and 3.9 million deaths. To date, emerging clinical evidence has shown the synergetic benefits of Chinese herbal injections in treating this contagious respiratory disease. This review aims to summarize and analyze the efficacy and safety of Chinese herbal injections in the therapy of COVID-19. Methods The literature from 3 electronic databases, PubMed, CNKI, and Web of Science, were searched using the search terms “COVID-19”, “SARS-CoV-2”, “traditional Chinese medicine”, “herb”, “herbal”, and “injection”. Then the identified articles were comprehensively evaluated. Results Limited data demonstrated that Chinese herbal injections could significantly improve the clinical outcomes of COVID-19 patients, especially in combination with conventional treatment strategies. The benefits of which were mainly associated with the relief of symptoms, prevention of secondary infection, regulation of inflammation and immune function. There was also evidence showing the inhibitory effects on SARS-CoV-2 replication in vitro. Nevertheless, available real-world data suggested the increased risk of adverse event. Furthermore, the defects of existing researches and the insights for discovering novel antiviral drugs were prospectively discussed. Conclusion Evidence-based advances revealed that Chinese herbal injections such as XueBiJing injection and ShenMai injection, exerted potent effects against COVID-19. Further laboratory researches and clinical evaluation are needed to gather scientific evidence on the efficacy and safety.
