      • Cell Response to Micro-patterned Polymeric Substrate with the Gradient of Rigidity

        Here we introduce a bio-interactive sensing platform that enables investigation of the migratory behavior of cells cultured on a flexible PDMS substrate. Unlike other approaches, a pure gradient of rigidity on the surface was achieved without the effect of surface morphology and surface chemistry maintained homogeneous. It is well known that the migration of cells is strongly affected by the rigidity of the surrounding environment in biological processes. To observe cell-substrate interaction thorough this biomechanical “sensing” in a more proper way, a PDMS layer was used to create a rigidity gradient between the stiff (2.5㎛) and soft (7.5㎛) regions of PDMS over a glass trench. The changes of surface rigidity were caused by the thickness variation of the PDMS layer on the glass structure. The stiffness change was 48.8% in normal direction (stiff: 2.9N/m, soft: 1.4N/m) and 25.5% in lateral direction (stiff 1N/m, soft: 0.8N/m), respectively. The result of migratory cell experiment clearly showed that the current cell preferred migrating toward the stiff region. These findings will provide important cues for cell differentiation and tissue engineering occurring through a bio-sensing mechanism.

        Recently, nanotechnology has been developed to a stage that makes it possible to produce, characterize, and specifically tailor the functional properties of the MNPs for biological application. This has led to various opportunities such as improving the quality of magnetic resonance imaging, hyperthermic treatment for malignant cells, drug delivery, immunoassay, and biomolecules separation. Specially, bacterial magnetic nanoparticle (BMP) has been played a role in a biomedical application. BMP has useful qualities of dispersibility and facilitative functionalization since they covered with a lipid membrane. In most studies, BMPs were generally used as a carrier which was immobilized with the specific targeting biomolecules. Thus, manipulation of biomolecules and organelles in a cell using BMP has been less developed. Here, we demonstrate the manipulation of chromosomes conjugated with BMP-antibodies complexes in a live mouse oocyte cell using the external magnetic field.

        장애와 빈곤의 이중위험을 경험하는 장애인당사자 및 가족을 위한 지원방안 고찰

        연구목적: 본 연구는 장애와 빈곤이라는 이중위험 상황에 놓인 장애인당사자와 그들 가족에 대한 국내외 선행연구의 동향을 분석함으로써 그들이 가진 실제적 어려움과 지원요구를 파악하고 구체적인 지원방안을 모색하고자 하였다. 연구방법: 2000년부터 2017년까지 발표된 국내외 학술문헌 중 장애와 빈곤의 이중위험 관련문헌을 68편 선정하여 연구주제, 발표연도, 연구방법, 연구대상자별 분석을 실시하고 장애인당사자와 가족이 가지는 지원요구와 구체적 지원사례를 분석하였다. 연구결과: 가장 많은 연구가 집중된 주제영역은 ‘장애와 빈곤간의 관련성 분석’ 및 ‘장애와 빈곤의 이중위험 당사자의 실제적 지원요구’로 국내외 간의 큰 차이는 나타나지 않았으나, 국외의 경우 해당 주제에 대한 연구가 본격화된 시기가 국내보다 빠르며 지원의 필요성 분석을 바탕으로 장애아동 및 청소년과 그 가족을 대상으로 한 실제 지원을 제공하고 효과성에 대한 분석까지 이루어진 연구가 국내보다 더 많은 것으로 나타났다. 결론: 연구결과를 토대로 장애와 빈곤의 이중위험을 경험하는 장애인당사자와 가족을 지원하기 위해 다음과 같은 제언이 이루어졌다: 1) 장애와 빈곤의 이중위험 당사자에 대한 지원의 폭과 지원내용 확대, 2) 연구기반의 지원정책 마련, 3) 각 개인과 가정이 가진 특수성을 고려한 개별사례접근 활성화. Purpose: The purpose of this study is to analyze the trends in research on people with disabilities and their families who experience a dual risk situation of disability and poverty, and to identify their difficulties and support needs and ultimately to find ways to support them. Method: A total of 68 articles published from 2000 to 2017 was selected, which were related to the dual risk of disability and poverty. Research trends were analyzed according to the subject, publication year, research method, research participants, and finally, specific support cases were analyzed. Results: The subject area on which most of the research had been focused was "the analysis of the relationship between disability and poverty" and "the demand for support from the dual risk parties of disability and poverty". In foreign countries, research on the subjects was started earlier and research that provided actual supports for children with disabilities and their families and an analysis of the effectiveness was published more frequently than that in Korea. Conclusion: Based on these findings, the following implications were provided to support people with disabilities and their families who experience dual risk of disability and poverty: (a) to expand the range of support for persons who experience dual risk of disability and poverty, (b) to establish the policy based on research, and (c) to promote the individual case approach based on individualized support needs.

      • 불연속적 강성구배에 의한 생쥐 섬유아세포 움직임에 대한 연구

        A directional cell migration is demonstrated on a discrete rigidity gradient enabled by an array of doublelayered micropillars. This micropillar array has varying diameters (3 ㎛ to 7 ㎛) to accomplish desired rigidity gradient. Fibronectin used as an extra cellular matrix (ECM) is immobilized on pillar “tops”, providing an environment for focal adhesion of cells. In order to avoid the size effect of the ECM on cell reactions, the top diameter of every pillar is maintained identical by fabricating the pillars with double-layered SU-8 mold. NIH3T3 mouse fibroblasts are cultured on the double-layered micro pillar array device, and the motion of the cells is observed by a live imaging instrument. It is found that the rigidity gradient (Δk) needs to be larger than a certain value to initiate a directional and consistent migration. Our device proves that the cells migrate toward stiffer pillars and stay within the stiff region once the directional migration starts.

        It is getting attractive to study the function of microenvironmental stimulus towards cell behavior and cancer cell fate these days. In order to investigate the functions of microenvironmental cues that affect cell or cancer cell behavior, it requires an ability to design these systems in vitro and assay responses of cells to these multiple signals. Recognizing how cell activities are controlled by the microenvironmental cues is, however, much more complex. To control and investigate cell behavior related in migration, we present micro pillar array device that can offer the rigidity gradient. Our experimental platform will be used for studying the mechanism of the mechanically driven cell migration to understand cancer metastasis. Also, findings from this research on such environmental-driven cell movement are providing new insights into many biomedical applications including implantable medical devices, biomaterials and tissue engineering.

        한국판 SWPBIS Tiered Fidelity Inventory Tier 1 (TFI T1)의 내용 타당도 연구

        연구목적: 본 연구의 목적은 학교 차원의 긍정적 행동지원의 실행충실도 평가도구인 SWPBIS Tiered Fidelity Inventory Tier 1 (TFI T1)을 번역하고 원문의 의도를 벗어나지 않으면서 국내 환경에 맞게 수정하는 것이다. 연구방법: TFI T1을 역번역 절차에 따라 번역하고, 전문가(5명)의 내용 일치도 및 내용 타당도 평가와 보편적 지원 관련 업무를 담당하거나 관련 경험이 있는 특수교사(18명)의 내용 이해도 검토를 통해 초기 번역을 수정하고 보완하였다. 연구결과: 총 2회에 걸쳐 실시된 전문가의 내용 일치도(1차 = .96, 2차 = .89) 및 내용 타당도(1차 = .96, 2차 = 1.00) 평가 결과는 대체로 양호한 수준이었고, 100% 동의를 얻지 못한 문항을 중심으로 수정과 보완이 이루어졌다. 또한, 내용 이해도 설문 결과 96% 이상의 교사가 모든 문항을 이해할 수 있다고 응답하였다. 교사 의견을 참고하여 최종 수정과 보완이 이루어졌다. 결론: 본 연구에서는 한국판 TFI T1의 개발과 관련하여 학교 환경에 적합하고 교사들이 이해할 수 있는 용어를 선택하여 제시하였다. 이 연구는 한국판 TFI T1의 내용 타당도에 관한 초기 연구로써 향후 실행충실도 평가도구의 학교 활용에 기여할 수 있을 것이다. Purpose: This study aims to translate into Korean language and adapt the SWPBIS Tiered Fidelity Inventory Tier 1 (TFI T1) fitting to the Korean school settings but adhering to the original intentions. Method: The TFI T1 was translated systematically, including the backward translation. Then, the expert panel (n = 5) reviewed and rated the translated TFI T1 in terms of its content validity and content equivalence in two sessions. Also, the special education teachers (n = 18), who were involved in coordinating or implementing universal support, assessed the updated TFI T1 in terms of their content comprehension. All these inputs allowed the initial translation to be adapted and supplemented. Results: The result of the expert panel’s ratings indicated that the content equivalence (1st = 0.96, 2nd = 0.89) and the content validity (1st = 0.96, 2nd = 1.00) appeared appropriate. In contrast, the items with less than 100% agreement among the experts were additionally reviewed and revised. Also, 96% and more special educators reported in their ratings of the content comprehension that they were able to understand all items of the TFI T1. Final revisions were made based on these teachers’ feedback. Conclusion: This initial translation study gave more weight to selecting the terminology teachers can understand and use in their environment, which may contribute to the wider school use of the fidelity measures to guide effective implementation of universal support.
