This experiment was carried out to establish the basic information of mass production of gladiolus (cv. Firebrand and Topaz) through callus culture. The callus formation from the cormel explant was depending on the levels and kinds of auxin to the MS solid medium; callus formation was best at 10ppm 2,4-D, and callus was initiated in about 40days cultures. Morphological characteristics of gladiolus callus on agar medium with 0.5ppm 2,4-D was nonchlorophyllous, light yellow, soft, friable and consisted of small clusters of spherical parenchyma cells. When callus was further subcultured on the medium with 0.5ppm 2,4-D, regenaration ability was reduced after 18 months of subculture. For regenaration of plantlets from the callus, low levels of cytokinin (BA 0.1ppm and kinetin 0.1-1.0ppm) was effective but higher concentration rather inhibited organ formation. The MS solid medium with 1.0ppm kinetin (containing 2-3% sucrose, 0.5-1.1% agar, and pH 5.7) gave the best regenaration of plantlets from the fresh callus under 16 hrs light. Formation of adventitious roots and shoots from the fresh callus was initiated 20 and 30days after subculture, respectively. However, cytokinin free medium was more effective than the medium with 1.0ppm kinetin for organ formation in call older than 6 months.
