Salinomycin 생산균주인 Streptomyces albus로 부터 Polyketide Synthase유전자 분리

Streptomyces albus wild type ATCC 21838 produced salinomycin, polyether antibiotic. To clone genes related salinomycin production, chromosomal DNA of the strain ATCC 21838 was used for the construction of genomic library using pKC 505 cosmid vector. The genomic library was screened using act I and act Ⅲ as DNA hybridization probes. pWHM210 was isolated, which contained an approximately 24 kb of insert DNA. Strepto-myces lividans transformed with the pWHM210 produced a very small amount of an ionophoric substance with the same chromatographic and mass spectromectric properties as salinomycin. As inducing production of a presumptive salinomycin by pWHM210 in salinomycin nonproducing organism, the 24 kb of DNA is very likely to comprize most of the gences for salinomycin biosynthesis. The 4.4 kb region of pWHM210 was hybridized with act I and act Ⅲ. This 4.4 kb DNA fragment in pIJ 702 complemented a mutant of Streptomyces glaucescens which was blocked in β - keto acyl synthase mediated step in tetracenomycin C biosynthesis. Se-quence analysis of 4.4 kb region revealed five open reading frames found in the same direction and they were homologous, in the predicted amino acid sequences, to tcm I,tcmK. tcmL, tcmM and act Ⅲ, respectively(tcm I, K, L are genes for tetracenomycin biosynthesis in Streptomyces glaucescens). tcm I was proposed to be a gene coding for a cyclase and tcmK and tcmL are genes for heterodimeric β -keto acylsynthase.
