Cytokinins overproduction에 따른 담배형질전환체의 변화

식물의 주요 phytohormone의 하나인 cytokinin은 식물체의 줄기와 뿌리성장 그 외에도 영양의 전달이나 노화방지, 열매숙성 등 식물의 성장과 발달에 미치는 영향은 크고 다양하다. Cytokinin 생합성에 관여하는 효소를 생산하는 것으로 알려져 있으며 토양박테리아 Agrobacterium tumefaciens에 존재하는 유전자인 isopentenyl transferase (jpt)를 이용한 많은 분자생물학적 연구가 진행되어 왔는데 그 중 하나로 이 연구에서 jpt 유전자에 의한 cytokinin의 overproduction이 식물체에 성장과 발달에 어떠한 영향을 주는지 관찰하고 그 결과가 제시할 수 있는 작물의 유전 공학적 이용가능성에 대하여 알아본다. Cytokinin is one of major growth regulators in plants. In this study, the gene isopentenyl transferase (jpt) which encodes a key enzyme involved in the biosynthesis of the growth regulator cytokinin isolated from Agrobacterium tumefaciens was introduced ito tobacco plant via Agrobacterium-mediated transformation. The jpt gene was modulated using the proteinase inhibitor II (PI-IIK) promotor. In general, this promoterlipt gene fusion resulted in overproduction of cytokinins throughout the transgenic plants. The overproduction of cytokinin caused dramatic changes in morphology of the plant, including stem thickness and reduced root development. The studies reported in this paper were initiated to examine the consequences of overproduction of cytokinin in plant.

In a SI engine, three- way catalyst converter has the best efficiency when A/F ratio is near the stoichiometry. The feedback control using oxygen sensors in the commercial engine has limits caused by the system delays. So it is necessary to control fuel Quantity in accordance with intake air amount in order to reduce exhaust emission and improve the specific fuel consumption. Precise A/F ratio control requires measurement of air amount with respect to the cylinder and injection fuel accounting to the air amount In this paper, we applied nonlinear fuel injection model, and developed the algorithm of A/F ratio control. This algorithm includes the methods of measurement of transient air mass flowing into each cylinder, of calculation of injection pulse width for measured air mass, and the method of feedback and estimation control by using lambda sensor. Also we developed control program for IBM-PC by using C++ Builder, and tested it in the commercial engine. The results we have got from the work better than those of the commercial ECU.

$CO_2$ 처리에 의한 배추 화주 유전자의 특이적 발현 연구

Chinese cabbage (Brassica campestris), one of the major vegetable crops in Korea, undergoes self-incompatible pathway for reproduction. To maintain inbred lines of chinese cabbage, a method in that $CO_2$ gas is treated to the pistils to break the self-incompatibility and thereby self-pollens can successfully make germination and fertilization has been selectively used in speed company. In this study, the pistil genes induced by the $CO_2$ treatment was investigated by mRNA differential display (DD-PCR) method. The result shows PCR products amplified in a differential pattern from both $CO_2$ gas treated- and untreated-pistil mRNAs, suggesting that the pistil genes are probably regulated positively and also negatively by the $CO_2$ gas. $CO_2$ gas 처리가 배추 자가불화합성 타파에 미치는 영향을 조사하고자 배추 inbred line의 암술에서 mRNA를 분리하여 DD-PCR을 수행하였다. $CO_2$ gas를 처리한 암술에서 분리한 mRNA와 처리하지 않은 꽃에서 분리한 암술 mRNA 발현과의 특이적인 차이를 DD-PCR로 조사한 결과 세 가지 각기 다른 anchor primer에 대해 모두 18개의 특이적 증폭 DNA fragment를 얻을 수 있었다. 이 결과로 미루어 보아 $CO_2$ gas 처리에 의한 암술 mRNA발현의 변화가 배추 자가불화합성 타파에 영향을 미치는 것으로 생각된다.
