인삼의 에탄올 추출물(WGpet-alc)은 사람 혈청 속에 있는 글루탐산-옥살초산 트란스아마나아제(GOT) 및 글루탐산-피루브산 트란스아미나아제(GPT)의 활성을 증가시키는 효과를 가졌으며, 이러한 효과는 WGpet-alc의 농도가 0.50% 일때 가장 현저하였고, 농도가 이보다 클 때는 도리어 감소되었다. 이와는 반대로, GOT 및 GPT의 활성은 인삼의 석유에테르 추출물 (WGpet)에 의해 크게 감소된다는 것이 확인되었다. Ethanol extract of Ginseng(WGpet-alc) was observed to increase the activities of human serum GOT and GPT in vitro and it was found that both enzymes showed the maximum activity when the concentration of WGpet-alc was 0.5% and the enzyme activity was decreased as the concentration of WGpet-alc rised over 0.5%. It was also observed that petroleum ether extract of Ginseng (WGpet) showed even at very low concentration an inhibitory effect on both enzymes.

On the basis of our previous report on the biochemical studies of Ginseng substances, it was attempted to observe the effect of ethanol extract (WGpet-alc) and petroleum ether extract (WGpet) of White Ginseng on the activity of human serum GIDH and pure GlDH in vitro. The activity of human serum GlDH and pure GlDH was significantly increased in the presence of WGpet-alc. On the other hand, an inhibitory effect of WGpet on GlDH was. observed even at low concentration. It was also confirmed that these activating and inhibitory effects have the same pattern as the effects on albino rat hepatic GlDH.

It has been confirmed that petroleum ether extract from White Ginseng has a remarkable inhibitory effect on human erythrocyte carbonic anhydrase. This effect is very similar to the inhibitory effect of the carbonic anhydrase by acetazolamide, and therefore it may be assumed that the diuretic action of Ginseng results from its hydrophobic inhibiting substances.

인삼에 포함되어 있는 carbonic anhydrase 활성화 또는 안정화성분의 열, 무기산, 알칼리, 몇 가지 산화제 및 환원제에 대한 안정성을 검토하였다. 이 성분은 열이나 묽은 무기산, $Na_2S_2O_4$와 같은 환원제에 대해 비교적 안정하나, 알칼리와 산화제($H_2O_$, NaClO, $K_3Fe(CN)_6$)에 의해 그 작용을 쉽게 잃게 된다는 것을 확인하였다. Effects of heat, mineral acid, alkalis, and oxidizing and reducing agents on the stability of the carbonic anhydrase activating or stabilizing principle in Panax Ginseng are investigated. It is found that the substance is relatively stable to heat, dil. mineral acid (hydrochloric acid), and reducing agent such as Na-hydrosulfite. However, the action of alkalis and oxidizing agents ($H_2O_$, NaClO, $K_3Fe(CN)_6$) resulted in loss of its function.

백삼의 석유에테르추출물(WGpet)은 빵효모의 RNA생합성을 극히 미약하게 촉진시킬 뿐이지만, 백삼의 에탄올추출물(WGpet-alc) 및 홍삼정(RGE)의 성분은 다같이 RNA 생합성을 크게 촉진시킬뿐 아니라, 그 모노누클레오티드의 조성에까지 큰 변화를 일으키는 효과가 있음이 확인되었다. 즉 RGE에 의해 AMP와 CMP는 물론, 특히 UMP가 현저하게 감소된 반면, GMP는 크게 증가되었으며, 그 몰비는 총 모노누클레오티드의 78.5%에 달하였다. 이러한 결과는 인삼성분의 단백질 생합성 촉진효과와도 관련이 있는 퍽 흥미로운 점이라고 하겠다. It was found that petroleum ether extract of White Ginseng had a very slightly stimulating effect on the RNA biosynthesis of Baker's yeast, while both ethanol extract of White Ginseng(WGpet-alc) and Red Ginseng Extract(RGE) showed a significant stimulating effect on the RNA biosynthesis and its activity to change mononucleotide composition of the yeast RNA was noticeable; RGE induced a remarkable decrease of AMP, CMP and especially of UMP, however a marked increase of GMP, and it was observed that the molar ratio of GMP was 78.5 % of all mononucleotides of the yeast RNA. It is of interest that these results are closely related with the stimulating effect of protein biosynthesis of Baker's yeast cells.

사람 혈청 속에 있는 L-글루탐산-데히드로게나아제 (GlDH) 및 순수한 GlDH (Sigma사 제품)의 활성은 인삼의 에탄올 추출물(WGpet-alc)에 의해 다같이 증가되었지만, 인삼의 석유에테르 추출물(WGpet)에 의해서는 작은 농도에서도 감소되었다. 이러한 효소활성 증가효과 또는 감소효과는 흰쥐의 간 GlDH에 대한 실험결과와도 여러 점에서 공동된 양상을 나타낸다는 것이 확인되었다. The activity of Albino rat hepatic L-glutamate dehydrogenase (GIDH) was greatly increased in the presence of the ginseng extract (WGpet-alc) in vitro, and it was found that at the concentration of 0.1% and 1.5% WGpet-alc the specific activity of the enzyme was about two and four times that of control respectively. The G1DH`s inhibitory effect of GTP was found gradually decreased as the concentration of WGpet-alc rised. Petroleum ether extract of ginseng (WGpet) was observed to inhibit the G1DH`s activity remarkably even at very low concentration. In relation to the physiological activities of ginseng substances, it may be noted that ginseng contains at least two different active principles) which showed the activating and inhibiting effect on the activity of an enzyme system.

In the aerobic metabolism of bakers` yeast using glucose as substrate, it was found that the CO₂ output was increased remarkably in the presence of 1.0-1.5% of the ginseng extracts (WGpet-alc and WGpet) : the CO₂ output increasing effect of WGpet-alc was greater than that of WGpet. Furthermore both ginseng extracts were observed to stimulate O₂ uptake of bakers` yeast and the maximum effect was shown when the concentration of WGpet-alc and WGpet were 1.0% and 1.5-3.0% respectively. In the presence of succinate as substrate, WGpet-alc showed the pronounced lowering effect on O₂ uptake and the enhancing activity of Pi-incorporation into bakers` yeast cells while WGpet had little stimulating effect on O₂ uptake and a significant inhibitory effect on the Pi-incorporation. These results suggest that WGpet-alc stimulates the oxidative phosphorylation in bakers` yeast cells and WGpet may act as an uncoupler.

인삼의 석유-에테르 추출물은 사람의 적혈구로부터 얻은 carbonic anhydrase의 활성을 크게 억제하는 작용을 가졌음이 확인되었다. 이 억제효과는 acetazolamide에 의한 carbonic anhydrase 억제효과와 매우 유사하며, 따라서 인삼의 이른바 이뇨작용은 이 소수성 억제 물질에 기인하는 것으로 추측된다. It has been confirmed that petroleum ether extract from White Ginseng has a remarkable inhibitory effect on human erythrocyte carbonic anhydrase. This effect is very similar to the inhibitory effect of the carbonic anhydrase by acetazolamide, and therefore it may be assumed that the diuretic action of Ginseng results from its hydrophobic inhibiting substances.
