The p53 protein is a well-known tumor suppressor that plays an important role in preventing cancer. The mutations of p53 such as functional damage of its DNA binding domain lead to tumors. Specially, the mutants of p53 at hot spot have unique changes of local electron density that affect specificity of DNA binding compared with the wild types. This can be detected by Rayleigh light scattering according to the changes of localized surface plasmon resonance (LSPR). The LSPR Δλmax shift of mutant p53 proteins is sifnificantly lower than that of wild types. These differences propose that hot-spot mutants can be easily detected by local electron density alterations that disturbs the specificity of DNA binding domain of p53 on the nanoplasmonic sensor. By this strategy, the nanoplasmonic sensor can detect mutant p53 proteins from samples of breast cancer patients and thus suggesting itself as a promising tool for using in cancer diagnosis and predict the effect from adjuvant treatment.

Signal transducer and activator of transcription 3 (STAT3) protein signaling is crucial for the survival and growth of human cancer cells; thus, STAT3 protein is an ideal target for a new drug screening system. Herein, we developed a label-free sensor for anticancer drug-discovery based on the LSPR hift response by tracking of STAT3 signaling including phosphorylation and dimerization. In screening of anticancer candidates, the system worked well in the presence of STA-21 which inhibits STAT3 dimerization. The LSPR λmax shift in the inhibition condition is three times lower than that in the absence of an inhibitor. Interestingly, the system reveals high specificity, reproducibility and compatibility with real samples (MCF-7 cell line). Therefore, these results demonstrated that this system has strong potential to be an accurate and effective sensor for tracking of signaling pathways and drug screening of anticancer candidates for anticancer therapy.
p53 hot-spot mutant based cancer detection using localized surface plasmon resonance

The Hair designers' self esteem and duty-satisfaction, and to investigate the influence on duty-satisfaction of self esteem. this thesis investigated hair designers working in Seoul-si Iksan-si, Jeonju-si, and Namwon-si. The first objects were 464 Hair designers, 16 to June 22, 2009, for three months. This thesis performed SPSS 14.0 Program, The result is as followings. The analyzed differences in self esteem, classified with general traits, The longer the carrer and the higher the average income, the higher was self esteem. As for analyzed difference in duty-satisfaction, classified with general traits, the higher the age, the higher was duty-satisfaction. As for position, The higher the position, the higher was duty-satisfaction, and for hair designers with more than 5 years' career. The hair designers with more than average 2 million's income had the highest duty satisfaction. As shown above, the higher the position, the more the wages, the more duty-satisfaction with self esteem hair designers felt. With the duty-satisfaction. hair disigners will apply themselves to qualitative enhancement.
