옥수수 뿌리로부터 분리한 Membrane - bound ATPase 의 특성에 관한 연구

The membrane-bound ATPases were separated on sucrose gradient from corn roots and characterized by pH optima, sensitivity to monovalent salt, Km and Vmax. The pH optima for the activity of all the ATPases associated with 13,000g pellet and 13,000∼80,000g pellet were 5 and 9, respectively. The ATPases in Fractions B and C of the 13,000g pellet were more active at pH 4 than pH 9. While, in the case of Fractions D, E and F, they were reverse. The activities of the ATPase in Fractions A and C of the 13,000∼80,000g pellet were greater at pH 5 than pH 9. On the other hand, the ATPases in Fractions B, D, E, and F were more active at pH 9 than pH 5. The optimum concentration of ATP for the assay was about 3 to 5mM. The Km`s for the membrane-bound ATPases in 13,000g pellet and in 13,000∼80,pppg pellet were 0.25mM. While Vmax values for 13,000g pellet were from 8.0 to 12.5 μM Pi/㎎ protein/hr, according to pH values, those for 13,000∼80,000g pellet were from 35.7 to 55.6 μM Pi/㎎ protein/hr. Activities of the membrane-bound ATPases in both 13,000g pellet and 13,000∼80,000g pellet were stimulated with increasing the concentration of K^+.

단백질의 생합성에 필요한 여러가지 components의 활성에 미치는 cAMP 의 효과를 조사하였다. cAMP로 처리된 ribosome의 활성은 대조구에 비하여 27% 증가하였으며 EF-1 과 EF-2의 활성은 cAMP에 의하며 각각 17%와 20% 증가 하였다. As a part of the studies on the regulatory mechanism of gene expression by gibberellic acid, the effects fo GA_3 on the protein biosynthesis and phosphorylation in maize seedlings were investigated in the presence of actinomycin D. The activites of protein biosynthesis and phosphorylation in germination seeds treated with GA_3 were greater than these of the control at the 3-day point after germination. It is assumed that the enhancement of protein biosynthesis by GA_3 in the presence of actinomycin D is due to the effects of GA_3 on the translational processes in which protein is produced from the mRNA synthesized previously.
Agrobacterium tumefaciens KU12로부터 Octopine형 Ti 및 잠재 플라스미드의 제거에 의한 숙주 개발

Agrobacterium tumefaciens KU12내에 존재하는 240kb 크기으 octopine형 Ti 플라스미드인 pTiKU12와 45kb 크기의 잠재 플라스미드인 pTi12를 제거하여 무독성의 A. tumefaciens 균주를 제조하였다. Octopine형 Ti 플라스미드인 pTiKU12는 고온(37${\circ}C$)에서의 배양과 ethidium bromide가 첨가되어 있는 배지에서의 배양을 각각 실시하여 제거하였으며, 잠재 플라스미드인 pTi12는 pTi12의 복제기원이 클로닝되어 잇는 재조합 플라스믿인 pYWXP와의 비화합성을 이용하여 제거하였다. pYWXP는 ethidium bromide가 첨가되어 있는 배지에서 고온(37${\circ}C$)으로 배양하여 제거하였다. Agrobacterium tumefaciens KU12 contains pTiKU12 (240kb) of the octopine type Ti plamsid and pTi12 (45 kb) of the cryptic plasmid. To make the avirulent A. tumefaciens, the octopine type Ti plasmid, pTiKU12, was cured with elevated temperature (37${\circ}C$) and ethidium bromide (EtBr), respectively. Also the cryptic plasmid, pTi12, was cured by the introduction of recombinant plasmid, pYWXP, made by pTi12 replication origin and pUC19. pYWXP was cured by elevated temperature (37${\circ}C$) and EtBr simultaneously.

cAMP가 $GA_3$의 작용기작에 관여하는 지를 밝히기 위해 단백질의 생합성과 인산화반응에 미치는 cAMP의 효과를 구명하였던바, 정상상태에서의 단백질의 생합성 및 인산화 반응은 cAMP에 의해 모두 촉진되었다. Transcription이 억제된 상태에서는 단백질의 생합성은 전 발아기간 중 대조구에 비해 촉진되었으나 인산화 반응은 발아 3일 후 cAMP에 의해 증가되었다. 또한 cAMP는 발아 초기에는 단백질의 인산화 반응 보다는 생합성을 촉진시키고, 발아가 진행됨에 따라 단백질의 생합성 보다는 인산화 반응을 촉진시킨다. 이러한 결과는 단백질의 생합성 및 인산화반응에 미치는 $GA_3$의 촉진효과와 동일한 점으로 보아 cAMP는 $GA_3$의 작용기작과 밀접한 관계가 있는 것으로 추정된다. In order to investigate cAMP's involvement in the action mechanisms of gibberellic acid, the effects of cAMP on the biosynthesis and phosphorylation of proteins in the germinating maize seeds were studied. cAMP promoted the biosynthesis and phosphorylation of proteins. In the presence of actinomycin D, the stimulation of protein biosynthesis by cAMP was continuous during the germination period, while the protein phosphorylation was increased at the 3-day point after germination by cAMP. The stimulatory effect of cAMP on the protein biosynthesis was greater than that on the prtein phosphorylation during the early stages of germination. On the other hand, this pattern is reversed during the latter period. The above mentioned results are consistent with the facilitation effects of $GA_3$ on the protein biosynthesis and phosphorylation. Therefore, it seems to be that cAMP is related to the action mechanisms of $GA_3$.

In order to investigate cAMP`s involvement in the action mechanisms of gibberellic acid, the effects of cAMP on the biosynthesis and phosphorylation of proteins in the germinating maize seeds were studied. cAMP promoted the biosynthesis and phosphorylation of proteins. In the presence of actinomycin D, the stimulation of protein biosynthesis by cAMP was continuous during the germination period, while the protein phosphorylation was increased at the 3-day point after germination by CAMP. The stimulatory effect of cAMP on the protein biosynthesis was greater than that on the prntein phosphorylation during the early stages of germination. On the other hand, this pattern is reversed during the latter period. The above mentioned results are consistent with the facilitation effects of GA₃ on the protein biosynthesis and phosphorylation.
