Mancozeb와 Pendimethalin에 의한 CHL Cell의 소핵생성과 세포사멸 여부 규명

Mancozeb is a widely used broad-spectrum fungicide known to be genotoxic and induces tumors in rodents at various sites. Pendimethalin is a member of family dinitroanaline of herbicides. Pendimethalin are well known to be more or less toxic to all living organisms. In this study, we investigated whether mancozeb and pendimethalin induce micronucleus and apoptosis in CHL (Chinese hamster lung) cells through flow-cytometry analysis for high throughput. We identified mancozeb (20, 40, 80 ug/ml) and pendimethalin (5, 10, 20 ug/ml) mediated micronuclei formation and apoptosis in a dose dependent manner. We evaluated an increase of micronuclei formation and apoptosis by flow-cytometry analysis and microscopy analysis. We identified that MMC (mitomycin) induced micronuclei formation as positive control and negative control (DW) has no effect on CHL cells. We demonstrated that mancozeb and pendimethalin may induce micronuclei formation and apoptosis in CHL cells. We also suggest that flow-cytometry analysis could be efficient high-throughput method for quantifying the formation of micronuclei.

In this study, performance analysis of a Dual Fluidezed-Bed(DFB) biomass gasification system was conducted. The target system is composed of a bubbling fluidized be gasifier and a circulating fluidized bed combustor. Appropriate heat and mass balances of the two reactors are important for increasing cold gas efficiency and total efficiency of the system. The capacity of gasifier is 15 ton/day and the highest Cold Gas Efficiency(CGE) is 73.11% at ER=0.2

Economic feasibility of power generation system of biomass syngas engine and direct combustion of biomass with steam turbine was investigated. Due to the low energy density of biomass, there is a limit in the capacity of biomass power generation plant. In this study, small scale biomass power generation plants ranging from 0.5㎿e to 5㎿e was considered. As a result, syngas engine system using bio-syngas is more economical than steam turbine system at low capacity system. The steam turbine system becomes better than syngas engine if the capacity is over 5MWe.

순산소 연소 기술에서는 공기에서 질소가 분리된 순산소와 재순환된 배기가스를 산화제로 이용하여, 배기가스의 수분 및 불순물을 제거하면 고순도의 이산화탄소만 배출된다. 이를 이산화탄소 저장 및 이용 기술에 적용하여 궁극적으로 zero-emission을 실현할 수 있는 기술이다. 이를 구현하기 위해서는 배기가스의 먼지, NOx, SOx, 물 등을 제거하는 것이 중요하다. 본 연구에서는 연료다변화가 가능한 순환유동층 공정에 순산소 연소 기술을 적용한 0.1 MW 급 순산소 순환유동층 설비를 이용하여 다양한 연료들의 공기/순산소 연소 및 대기오염물질 배출 특성을 알아보았다. 기존 공기 연소와 비교하여 연소 효율은 2% 이상 상승하였고, 대기오염물질 배출량은 후처리 공정 없이 80% 이상 감소됨을 확인하였다.

Woodchip, a carbon neutral fuel, is one of the fastest growing renewable energy sources. Among many conversion methods to energy, gasification is the best way in terms of energy efficiency and various options for end use like heat, steam, electronics, gaseous and liquid fuel. In the gasification process of woodchip, tar treatment is very important to avoid any mechanical problems and to increase heating value of product gas. However, tar generation process is not fully understood yet. In this study, we investigated tar generation characteristics of woodchip in devolatization and char gasification process. We found that different kinds of tar was generated in the devolatization and gasification process. The effects of temperature and steam amount are also studied.

Biomass, a carbon neutral fuel, is one of the fastest growing renewable energy sources. In this study, economic feasibility of small scale power generation system using syngas from biomass has been investigated. Effects of important variables such as capital investment, cost of waste wood, renewable energy certificate(REC), system marginal price(SMP) on the benefit of business have been analyzed. Internal rate of return(IRR) was predicted as 16.90%, which implicates the business is promising based on the assumptions such as SMP of 137 Won/kWh, capital cost of 4.7 billion won, and waste wood price of 30 thousand won. Major factors affecting the benefit of business are as follows; system marginal price, operational rate, capital investment, expenditure of waste wood, certified emission reduction.
유세포분석기를 이용한 농약의 in vitro 소핵생성 평가

Pesticides have been recognized as one of major concern to human and environmental health. This study is aimed to evaluate whether flow cytometry analysis is a suitable method for micronuclei test, one of genotoxicity test of pesticides on human. We evaluated the number of micronuclei in the positive control (mitomycin C, colchicine) treated groups in Chinese hamster lung cells using flow cytometry. As a results, micronucleus rate in positive control were 2.4~4.2 much higher than in negative control group. These results indicate that flow cytometry method is a quicker and more efficient method for obtaining similar results to the conventional microscopic observation method. Also, pendimethalin (PM) and mancozeb (MZ), which are reported as pesticides causing genotoxicity, were treated with CHL cells, and the occurrence of micronuclei was confirmed by flow cytometry analysis. The occurrence of micronuclei in PM and MZ treated group was significantly increased with a dose-dependent manner, compared with negative control group. These results suggest that micronuclei assay using flow-cytometry analysis could be efficient high-throughput method for quantifying the formation of micronuclei.
