Pinellia ternata Breitenbach, the natural medicinal plant of the Araceae family, is a perennial plant originated from the East Asia, but also widely distributed in Europe and North America. Its tuber is used as traditional medicine for treatment of various diseases such as vomiting, inflammation, and traumatic injury. Pharmacological studies revealed that P. ternata possesses anticonvulsant, anti-tumor, insecticidal, and cytotoxic activities. Despite being well-known as the useful medicinal plant, there is no reliable, standardized method for origin discrimination. Ultra performance liquid chromatography-photodiode array detector and quadrupole time of flight-mass spectrometry based metaboliteprofiling was applied to explore significant metabolite for origin discrimination between Korean and Chinese P. ternata. One compound was isolated from Korean P. ternata using repeated ODS column chromatography by bioactivity guided fractionation, and determined as [6]-gingerol according to the results of spectroscopic data including nuclear magnetic resonance and MS. This compound was selected as cosmeceutical biomarker by fingerprints, and it was associated to melanin inhibitory effect determining its origin authenticity. Furthermore, the calibration curve of biomarker was prepared using validated method for the comparison of content between Korean and Chinese P. ternata. This is the report to address the selection and successful validation of the discriminant metabolite for confirmation of Korean P. ternata.
넓패 추출물이 HeLa 자궁암세포의 세포사멸에 미치는 영향

본 연구에서는 넓패 메탄올 추출물의 농도별 처리가 인체자궁암 세포 HeLa의 세포사멸에 미치는 영향을 확인하기 위하여 세포독성 측정, Hoechst 33258 staining, flow cytometry 분석을 통하여 세포사멸을 확인하였다. 넓패 메탄올 추출물 처리 시 HeLa 세포에서 농도 의존적으로 세포의 증식을 억제하였으며, 또한 넓패 메탄올 추출물은 농도 의존적으로 핵을 응축하고 apoptotic bodies을 생성하였다. 유세포 분석을 통하여 apoptosis를 측정한 결과, 넓패 메탄올 추출물의 농도가 증가함에 따라 유의적으로 apoptotic 세포가 증가하였다. Western blot을 통해 PARP 단백질의 절단 현상을 분석한 결과, 넓패 메탄올 추출물의 처리 농도와 시간에 따라 PARP 단백질의 절단 현상이 증가하였다. 또한 넓패 메탄올 추출물은 caspase-8, caspase-9 및 caspase-3 활성을 농도와 시간에 따라 증가시켰으며, caspase 저해제인 z-VAD-fmk로 처리 시 넓패 메탄올 추출물에 의한 세포사멸이 유의적으로 감소되어 넓패 메탄올 추출물에 의한 HeLa 세포의 apoptosis 유도에 caspase가 중요한 역할을 하고 있음을 확인하였다. 따라서 넓패 메탄올 추출물은 HeLa 자궁암 세포의 apoptosis를 유도하는 것으로 나타나 넓패의 항암효과 가능성을 제시하였다. The purpose of this study was to elucidate the anti-proliferative effect and the mechanisms underlying apoptosis induced by a methanol extracts from Ishige sinicola(ISE) in HeLa cells. ISE treatment for 24 hr significantly inhibited cell viability in a dose-dependent manner. Apoptosis was detected by Hoechst 33258 staining and an annexin V/PI assay after 24 hr treatment. Moreover, ISE treatment triggered the cleavage of caspase-8, -9, -3, and poly(ADP-ribose) polymerase (PARP) in dose-dependent and time-dependent manners. In addition, z-VAD-fmk, a general caspase inhibitor, blocked ISE-induced cell death. Taken together, these results suggest that ISE-induced apoptosis is mediated by the activation of a caspase cascade in HeLa cells.

The roots of Achyranthes japonica Nakai were extracted with 100% aqueous and concentrated subfraction was separated with ultra-performance liquid chromatography-based activity profiling. Three compounds were isolated from the subfraction 5 through the repeated prep- high performance liquid chromatography column chromatography. According to the results of physico-chemical and spectroscopic data including NMR and MS, the chemical structures of the compounds were determined as ecdysterone (1), 25S-inokosterone (2), and 25R-inokosterone (3). Three phytoecdysones were showed weak inhibitory activity for thymus and activation-regulated chemokine expression levels in tumor necrosis factor (TNF)-α plus IFN-γ induced HaCaT cells, respectively. However, those compounds 1-3 were exhibited the most potent inhibition (80-95% at 200 μg/mL) against TNF-α expression levels in A23187 plus phorbol-myrisrate acetate-induced RBL-2H3 cells. As result, 100% aqueous extract of A. japonica has an excellent anti-atopy activity. It could be used to a large range of functional anti-atopy cosmetics.

The rhizomes of Anemarrhena asphodeloide was extracted with 100% ethanol and concentrated subfractions were separated with medium pressure liquid chromatography-based activity profiling. One compound was isolated from the subfraction 10 through the repeated preparative high performance liquid chromatography (prep-HPLC). According to physico-chemical and spectroscopic data including NMR and MS, the chemical structures of the compound was determined as nyasol (1). Nyasol was exhibited potent inhibitory activity for NO (IC50: 12.5 μg/mL), tyrosinase (IC50: 12.5 μg/mL), melanin contents (IC50: 12.5 μg/mL), thymus and activation-regulated chemokine (TARC) production (IC50: 6.25 μg/mL). As a result, nyasol has an excellent inflammation-dependent anti-whitening and TARC production activity. It could be used to a large range of functional cosmetics.
