The root of Polygala tenuifolia has been utilized as an expectorant in chinese medicine. Chemical researches on polygala root have been reported by a number of investigators. But since the pharmacological basis of polygala root was not known, we have attempted to study the influence of saponin fraction extracted from the root of Polygala tenuifolia on cardiovascular system in rabbits. Results obtained from the study are as follows: 1) The saponin fraction of Polygala tenuifolia administered intravenously (4mg/kg) to anesthetized rabbits produced characteristically a transitory hypotension followed by a considerably longer hypertensive response. 2) The intravenous administration of atropine 2mg/kg or propranolol 1mg/kg ,did not inhibit the hypotensive response to intravenous polygala saponin 4mg/kg. 3) The hypotensive component of polygala saponin response was prolonged by pretreament with phentolamine lmg/kg and the hypertensive component of the polygala saponin response was slightly reduced by phentolamine 1mg/kg. 4) In case of isolated left atrium of rabbit, a negative isotropic effect was exhibited and the threshold stimulus to induce contraction was increased. 5) The number of perfusion drops in ear of rabbits treated with polygala saponin were reduced significantly in comparison with those of control group. 6) The tonus of aorta spiral segment was increased by polygala saponin 80,ug/ mI. It is, therefore, suggested that the transitory hypotensive response to polygala saponin is dependent on the possibility of cardiac participation and a considerably longer hypertensive component is vascular origin.
