        Interactions between Indole-3-acetic Acid Producing Acinetobacter sp. SW5 and Growth of Tomato Plant

        Many rhizobacteria can promote plant growth through various direct or indirect mechanisms, and their production of phytohormones such as indole-3-acetic acid (IAA) may have pronounced effects on growth and development of plants. Rhizobacterial strain isolated from rhizosphere of foxtail (Setaria viridis), Acinetobacter sp. SW5 produced 118.1 mg/L of IAA and 4.5 mg/L of gibberellin ($GA_3$) in brain heart broth medium at 2 and 1 day of incubation, respectively. In a pot test the lengths of stem and root and fresh weight of the germinated tomato seedlings treated with Acinetobacter sp. SW5 significantly increased by 26.3, 33.3, and 105.3%, respectively compared to those of the uninoculated control in 12 weeks of cultivation. When the root exudate secreted from tomato seedlings was analyzed by HPLC, 3.75 ng mg tomato $root^{-1}$ of tryptophan which is an IAA precursor was detected. Acinetobacter sp. SW5 could produce $4.06{\mu}M$ of IAA from root exudate from 8 tomato seedlings. Together with the capability of growth of Acinetobacter sp. SW5 in the tomato root exudates, this IAA secreted by bacteria might contribute to enhance the growth of tomato plants. 많은 근권세균들이 다양한 직간접적인 방법을 통해 식물생장을 촉진할 수 있으며 indole acetic acid (IAA) 같은 식물호르몬의 생산과 분비는 식물의 생장과 발달에 큰 영향을 미친다. 강아지풀의 근권에서 분리된 Acinetobacter sp. SW5는 BHB 배지에서 각각 2일과 1일 배양 시 118.1 mg/L의 IAA와 4.5 mg/L의 지베렐린을 생성하였다. 소규모 재배실험에서 토마토 유묘에 이 균주를 2주 간격으로 처리하면서 12주 재배 시 토마토 식물의 shoot와 뿌리 길이 및 습윤중량이 비접종 대조군과 비교할 때 통계적으로 유의하게 각각 26.3, 33.3과 105.3% 증가하였다. 토마토 유묘의 뿌리로부터 분비되는 삼출물을 HPLC로 분석한 결과 IAA의 전구물질인 아미노산 트립토판이 3.75 ng/mg tomato root 검출되었으며 Acinetobacter sp. SW5는 8개의 토마토 유묘뿌리 삼출물로부터 $4.06{\mu}M$의 IAA를 생성하였다. 토마토 뿌리 삼출물에서 Acinetobacter sp. SW5의 생장능과 더불어 이 세균에 의해 생성된 IAA가 토마토 식물의 생장을 촉진시키는데 관여했을 것으로 추정된다.

        Environmental concern over conventional agricultural fertilization and disease control measures has led to increased interests in finding the environment friendly alternatives including the plant growth promoting bacteria. In this study some photosynthetic bacteria and heterotrophic bacteria were isolated from the aquatic environments. Phylogenetic analyses based on 16s rDNA gene sequences showed that the selected strains JFR, CR, JJ3 and DB are belonged to Rhodobacter blasticus, Rhodobacter johrii, Klebsiella veriicola and Proteus vulgaris, respectively, sharing 98.224, 98.672, 98.452 and 99.717% of sequence similarity with the closest neighbor of each type strain. They are able to exert the beneficial effects upon plant growth by production of phytohormones. Among the isolates, R. blasticus JFR showed the highest production rate of 1,388.102 ㎍ ㎎ protein-¹ of indole acetic acid, 1,251.375 ㎍ ㎎ protein-¹ of indole butyric acid in the modified Biebl and Pfenning medium. R. johrii JJ3 showed the soluble phosphate production of 536.3 ㎎ L-¹ in 5 days of cultivation in LB medium. The production rates of 5-aminolevulinic acid by R. blasticus JFR and R. johrii CR in LB medium were 2.54 and 0.96 ㎍ ㎎ protein-¹, respectively in 1 day. Dry weight of germinated tomato seedling inoculated with R. blasticus JFR was increased by 85.7% (p<0.005) than that of the control in 21 days. These photosynthetic bacteria and heterotrophic bacteria may be utilized as environment-friendly biofertilizer in the agriculture.

        냉동 건조 탈회 동종골 이식후 골막이 골 형성 과정에 미치는 영향에 관한 실험적 연구

        Periosteum in general is described as a specialized fibrous membrane of mesenchymal origin consisting of two basis layers : outer fibrous layer consists of irregularly arranged dense connective-tissue with fibroblasts, and inner osteogenic or cambial layer is composed of more loosely arranged fibers, greater vascularity and flatted spindle-shaped pre-osteoblasts. This periosteum may serve in controlling bone growth, especially mandibular growth has been emphasized. But, the periosteum enwrapping the facial skeleton have been studied for many years leaving a controversy in opinion regarding the function of these structures. We evaluated the bone formation activity of te periosteum in allogeneic bone grafts which bones are made of freeze-dried preparation preoperatively. We made the calvarial bone defects, 5×7mm sized, amd grafted with allogeneic bone in rats, which a half of specimens has dissected the overlying periosteum and a rest intacted. After bone grafting, we evaluated the capacity ofbone formation of periosteum, 1, 2, 4, 6, 8 weeks postoperatively. There are subtle differences of bone formation during early healing period after demineralized allogeneic bone grafting between control groups with periosteum and experimental groups without periosteum.
