        Isoelectronic Pt(ii)- and Au(iii)-N-heterocyclic carbene complexes: a structural and biological comparison

        <P>To elucidate the intrinsic biological activities displayed by platinum <I>versus</I> gold-based organometallic reagents, the isoelectronic and isostructural square planar complexes [Pt(C∩N)(C)Cl][PF6] (2) and [Au(C∩N)(C)Cl][PF6]2 (3), which are supported by a common N-heterocyclic carbene, 1-methyl-2-pyridin-2-yl-2<I>H</I>-imidazo[1,5-<I>a</I>]pyridin-4-ylidene (1), were synthesized. The complexes were characterized using a range of spectroscopic techniques as well as single crystal X-ray diffraction and a series of computational studies were also performed to gain additional insight into their fundamental electronic bonding structures. The cytotoxicities of the complexes were tested <I>in vitro</I> against non-small lung carcinoma (A549), colorectal carcinoma (HCT-116) and human hepatocellular carcinoma (HepG2) cell lines. The measured IC50 values showed that the Au(iii)-NHC complex is more potent than its Pt analogue.</P>

      • Association of Two Polymorphisms of DNA Polymerase Beta in Exon-9 and Exon-11 with Ovarian Carcinoma in India

        Background: DNA polymerase beta ($pol{\beta}$) is a key enzyme in the base excision repair pathway. It is 39kDa protein, with two subunits, one large subunit of 31 kDa having catalytic activity between exon V to exon XIV, and an 8 kDa smaller subunit having single strand DNA binding activity. Exons V to VII have double strand DNA binding activity, whereas exons VIII to XI account for the nucleotidyl transferase activity and exons XII to XIV the dNTP selection activity. Aim: To examine the association between $pol{\beta}$ polymorphisms and the risk of ovarian cancer, the present case control study was performed using 152 cancer samples and non-metastatic normal samples from the same patients. In this study, mutational analysis of $pol{\beta}$ genomic DNA was undertaken using primers from exons IX to XIV - the portion having catalytic activity. Results: We detected alteration in DNA polymerase beta by SSCP. Two specific heterozygous point mutations of $pol{\beta}$ were identified in Exon 9:486, A->C (polymorphism 1; 11.18%) and in Exon 11:676, A->C (polymorphism 2; 9.86%). The correlation study involving polymorphism 1 and 4 types of tissue showed a significant correlation between mucinous type with a Pearson correlation value of 4.03 (p=0.04). The association among polymorphism 2 with serous type and stage IV together have shown Pearson ${\chi}^2$ value of 3.28 with likelihood ratio of 4.4 (p=0.07) with OR =2.08 (0.3-14.55). This indicates that there is a tendency of correlation among polymorphism 2, serous type and stage IV, indicating a risk factor for ovarian cancer. Conclusion: Hence, the results indicate that there is a tendency for $pol{\beta}$ polymorphisms being a risk factor for ovarian carcinogenesis in India.

        CFD simulations of the flow field of a laboratory-simulated tornado for parameter sensitivity studies and comparison with field measurements

        A better understanding of tornado-induced wind loads is needed to improve the design of typical structures to resist these winds. An accurate understanding of the loads requires knowledge of near-ground tornado winds, but observations in this region are lacking. The first goal of this study was to verify how well a CFD model, when driven by far field radar observations and laboratory measurements, could capture the flow characteristics of both full scale and laboratory-simulated tornadoes. A second goal was to use the model to examine the sensitivity of the simulations to various parameters that might affect the laboratory simulator tornado. An understanding of near-ground winds in tornadoes will require coordinated efforts in both computational and physical simulation. The sensitivity of computational simulations of a tornado to geometric parameters and surface roughness within a domain based on the Iowa State University laboratory tornado simulator was investigated. In this study, CFD simulations of the flow field in a model domain that represents a laboratory tornado simulator were conducted using Doppler radar and laboratory velocity measurements as boundary conditions. The tornado was found to be sensitive to a variety of geometric parameters used in the numerical model. Increased surface roughness was found to reduce the tangential speed in the vortex near the ground and enlarge the core radius of the vortex. The core radius was a function of the swirl ratio while the peak tangential flow was a function of the magnitude of the total inflow velocity. The CFD simulations showed that it is possible to numerically simulate the surface winds of a tornado and control certain parameters of the laboratory simulator to influence the tornado characteristics of interest to engineers and match those of the field.

        Study on the Quality of Market Dahi Compared to Laboratory Made Dahi

        Dahi is yoghurt like fermented dairy products available in Bangladesh made by traditional ways from raw milk. The aim of the present study was to check and compare the present quality status of the market dahi in reference to laboratory made dahi. For this, dahi was prepared in the laboratory (A) and collected from Kishoregonj district (B), Gazipur district (C), Bogra district (D) and Dhaka district (E and F). Consumer acceptance quality was judged on 100 point by a judge panel and chemical and microbiological quality test were done by established methods. ‘A’ type dahi was superior followed by E, D, F, B and C considering the consumer acceptance quality parameter. From chemical test, it appears that, B possess the lowest pH (3.75 ± 0.05) and highest was in C (4.46 ± 0.15). Total solids content of F type dahi (318.40 ± 4.44 g/kg) was highest and fat (52.00 ± 2.00 g/kg), protein (44.33 ± 2.00 g/kg) and ash (10.76 ± 0.31 g/kg) content were also higher in F type dahi. E and A type dahi were also with considerable figure in chemical quality parameter. Dahi B was inferior in respect of microbiological quality- total viable count (TVC) (× 105 cfu/ml) content was 94.00 ± 4.58, yeast (cfu/ml) content was 183.33 ± 15.28 and mold (cfu/ml) content was 53.33 ± 15.28. The TVC (× 105 cfu/ml), yeast (cfu/ml) and mold (cfu/ml) content of A type dahi were 39.67 ± 4.51, 50.00 ± 10.00 and 20.00 ± 10.00, respectively. Considering all the three major quality aspects dahi A was superior than others followed by F, D, E, C and B.         다히(dahi)는 원유를 이용하여 방글라데시 전통 방법으로 발효시킨 요구르트와 비슷한 발효 유제품으로 본 연구는 실험실에서 만든 다히(dahi)와 시장에서 판매되고 있는 다히(dahi)의 품질을 비교하기 위하여 수행되었다. 실험실에서 준비한 다히(dahi)는 (A), Kishoregonj 지역에서 수집한 다히(dahi)는 (B), Gazipur 지역의 다히(dahi)는 (C), Bogra 지역의 다히(dahi)는 (D), Dhaka 지역의 다히(dahi)는 (E, F)로 각각 구분하였다. 소비자 만족 품질은 심사위원들에 의해 100점 만점으로 판단되었으며 화학적 미생물학적 품질 시험은 확립된 방법에 의해 수행되었다. 'A' 타입의 다히(dahi)가 소비자 만족 품질에 있어서 가장 우수한 것으로 나타났으며 그 다음으로 E, D, F, B, C 순으로 나타났다. 화학적 시험 결과, 'B'타입의 다히(dahi)가 가장 낮은 pH ($3.75\;{\pm}\;0.05$)를 나타냈으며 'C'타입의 다히(dahi)가 가장 높은 pH ($4.46\;{\pm}\;0.15$)를 나타내었다. 'F' 타입의 다히(dahi)의 총 고형물, 지방, 단백질, 회분이 각 각 $318.40\;{\pm}\;4.44\;g/kg$, $52.00\;{\pm}\;2.00\;g/kg$, $44.33\;{\pm}\;2.00\;g/kg$, $10.76\;{\pm}\;0.31\;g/kg$으로 가장 높게 나타났다. 'B'타입의 다히(dahi)에서 total viable count (TVC)는 $94.00\;{\pm}\;4.58\;cfu/ml$, 효모가 $183.33\;{\pm}\;15.28\;cfu/ml$, 곰팡이가 $53.33\;{\pm}\;15.28\;cfu/ml$로 미생물학적 품질 측면에서 가장 좋지 않은 것으로 나타났다. 'A' 타입의 다히(dahi)는 TVC, 효모와 곰팡이가 각 각 $39.67\;{\pm}\;4.51\;cfu/ml$, $50.00\;{\pm}\;10.00\;cfu/ml$, $20.00\;{\pm}\;10.00\;cfu/ml$로 나타났다. 3가지 주요 품질을 고려하였을 때 'A'타입의 다히(dahi)가 가장 우수하였으며 그 다음으로 F, D, E, C, B 순으로 나타났다.         Relationship Between Heat Unit Requirement and Growth and Yield of Mulberry, Morns indica L.

        Growth and development of a plant over a growing season is closely related to the daily accumulation of heat. Heat unit measured by accumulated growing degree days (GDD) is becoming increasingly popular to estimate the growth of a plant or even in insect. GDD or heat accumulation per day is measured by calculating average daily temperature and then subtracting the base temperature below which growth does not occur. Heat accumulation per day is added for the desired period and accumulated GDD is determined. The present study was conducted in five seasons in an established garden with K-2, S-36 and V-1 mulberry varieties belonging to Morus indica L. grown under completely irrigated condition at the farm of CSRTI, Mysore during 2001 - 2002. Plants were pruned in each season and the growth of the plant measured by total shoot length and fresh leaf yield was recorded at an interval of 5 days starting from 30 days of pruning (DAP) to 70 days when all the plants were pruned. The accumulated GDD for the corresponding days were recorded and used for analysis. Accumulated growing degree days (GDD) have been found to be perfectly correlated with both growth and yield in all the seasons in all the varieties studied. The high $R^2$ values indicated a strong relationship between the accumulated GDD and, growth and yield of mulberry.

        In situ occurrence of a gall midge (Insecta, Diptera, Cecidomyiidae) on fossilized angiosperm leaf cuticle fragments from the Pliocene sediments of eastern India

        In situ preservation of fossil insect damage in plant fossils is an excellent tool to study the coevolution of flora and fauna through geological time, but finding both damage and the insect causing that damage in the same specimen is a very rare phenomenon. Galling is a common form of angiosperm leaf damage, which can be regarded as a kind of extended phenotype of the causal insects, essentially the gall midges, but galls usually lack remains of the insects themselves. Here we report the in situ occurrence of a gall midge (Insecta, Diptera, Cecidomyiidae) as well as its pupal exuviae on the abaxial cuticular surface of fossilized leaf cuticle fragments of Fabaceae leaves (cf. Albizia) that also bear galls, recovered from the latest Neogene (Rajdanda Formation, Pliocene) sediments of the Chotonagpur Plateau, Jharkhand, northeastern India. This Pliocene gall midge features well-preserved legs, segmented antenna with distinct and enlarged scape, elongate curved setae, and longer than broad terminal plate of the ovipositor lamellae. The in situ presence of a gall midge on a host fabaceous leaf cuticle indicates the existence of a host-ectoparasite relationship in the ancient warm and humid tropical monsoon-influenced forests of eastern India during the Pliocene. This is the first authentic fossil record of an in situ phytophagous insect of Cecidomyiidae from India, as well as southeast Asia. Although the identification of the recovered phytophagous insect associated with the fossil leaf cuticle is only possible to family level, this find reveals that such plant-insect relationships existed in the Pliocene of eastern India.

      • Exon 8-9 Mutations of DNA Polymerase β in Ovarian Carcinoma Patients from Haldia, India

        Background: Ovarian cancer is the number one killer among all the gynecological cancers. We undertook association study to identify potential alterations in the genomic DNA of a DNA repair gene, DNA polymerase beta ($pol{\beta}$), involved in base excision repair (BER), in ovarian carcinomas of patients from Haldia, India. Mutations, splice variants have been reported earlier in different tumors other than ovarian tumors. Aim: In this study we explored the possibility of association of any mutation of $pol{\beta}$ (Exon 8) with prognosis in 152 ovarian cancer samples. Results: Alteration in the exon 8 region (Exon 8:468, $A{\rightarrow}C$; 15.1%) was noted among fifty seven polymorphism positive samples. Alteration in the intervening sequence 8 (IVS8, -25, $A{\rightarrow}C$; 3.9%) was also noted. All alterations are heterozygous in nature. Conclusions: We found no significant association among the samples from serous type, stage IV, and the $pol{\beta}$ mutations ($P{\leq}0.01$). Only a slight tendency of association was evident between IVS8, -25, A to C; and stage III. Further analysis with a larger number of samples is needed.

        Variability in Specific Leaf Weight in Mulberry Germplasm and Its Inheritance Pattern

        Specific leaf weight (SLW), defined as the mass of tissue per unit leaf area has been found to be an important physiological parameter as it indicates the relative thickness of leaves. Greater SLW provides more photosynthetic potential per unit area of leaf and hence it is frequently been considered as correlated with photosynthesis in several plant species. Collections of 165 mulberry (Morus sp.) germplasm accessions, both Indian and exotic in origin were evaluated for their variability with respect to SLW. The mean specific leaf weight ranged from 35.3 to $72.3 g/m^{-2}$. The distribution of SLW was found to be normal. High heritability (97.08%) and a small difference between genotypic and phenotypic variance demonstrates the genetic control over SLW. Significant heterotic effect with respect to SLW was observed in crosses when parents with high and low SLW were chosen.
