RPK118 is a sphingosine kinase-1-binding protein that has been implicated in sphingosine 1 phosphate-mediated signaling. It contains a PX (phox homology) domain and two pseudo-kinase domains, and co-localizes with sphingosine kinase-1 on early endosomes. In this study we identified a novel RPK118-binding protein, PRDX3 (peroxiredoxin-3), by yeast two-hybrid screening. The interaction between these proteins was confirmed by pull-down assays and co-immunoprecipitation experi-ments. Deletion studies showed that RPK118 inter-acted with PRDX3 through its pseudokinase domains, and with early endosomes through its PX domain. Dou-ble immunofluorescence experiments demonstrated that PRDX3 co-localized with RPK118 on early endosomes in COS7 cells. PRDX3 is a member of the antioxidant family of proteins synthesized in the cytoplasm and functioning in mitochondria. Our findings indicate that RPK118 is a PRDX3-binding protein that may be involved in transporting PRDX3 from the cytoplasm to its mitochondrial site of function or to other mem-brane structures via endosome trafficking

Recently, poly-γ-glutamic acid synthetase A (pgsA) has been applied to display exogenous proteins on the surface of Lactobacillus casei or Lactococcus lactis, which results in a surfacedisplayed component of bacteria. However, the ability of carrying genes encoded by plasmids and the expression efficiency of recombinant bacteria can be somewhat affected by the longer gene length of pgsA (1,143 bp); therefore, a truncated gene, pgsA, was generated based on the characteristics of pgsA by computational analysis. Using murine IL-10 as an exogenous gene, recombinant Lactobacillus plantarum was constructed and the capacity of the surface-displayed protein and functional differences between exogenous proteins expressed by these strains were evaluated. Surface expression of IL-10 on both recombinant bacteria with anchorins and the higher expression levels in L. plantarum-pgsA’-IL-10 were confirmed by western blot assay. Most importantly, up-regulation of IL-1β, IL-6, TNF-α, IFN-γ, and the nuclear transcription factor NF-κB p65 in RAW264.7 cells after stimulation with Poly(I:C) or LPS was exacerbated after co-culture with L. plantarum-pgsA. By contrast, IL-10 expressed by these recombinant strains could reduce these factors, and the expression of these factors was associated with recombinant strains that expressed anchorin (especially in L. plantarum-pgsA’-IL-10) and was significantly lower compared with the anchorin-free strains. These findings indicated that exogenous proteins could be successfully displayed on the surface of L. plantarum by pgsA or pgsA’, and the expression of recombinant bacteria with pgsA’ was superior compared with bacteria with pgsA.

This paper studies the problem of H∞ model reference tracking control for continuous time networked control systems (NCSs) with communication constraints. By using the continuous Jensen inequality, linear matrix inequality (LMI)-based H∞ model reference tracking controller design for nominal NCSs with controller-to-actuator communication constraints is presented, and a new method is proposed to design H∞ model reference tracking controllers for NCSs with both controller-to-actuator and sensor-to- controller communication constraints. The results are also extended to NCSs with norm-bounded parameter uncertainties. The merits of the proposed methods lie in their less computational complexity and less conservatism, which are achieved by adopting continuous Jensen inequality and proposing new controller design methods, respectively. The simulation results illustrate the effectiveness of the proposed H∞ model reference tracking controller design for NCSs with communication constraints.
Genome-Wide Association Study in East Asians Identifies Novel Susceptibility Loci for Breast Cancer

<P>Genetic factors play an important role in the etiology of both sporadic and familial breast cancer. We aimed to discover novel genetic susceptibility loci for breast cancer. We conducted a four-stage genome-wide association study (GWAS) in 19,091 cases and 20,606 controls of East-Asian descent including Chinese, Korean, and Japanese women. After analyzing 690,947 SNPs in 2,918 cases and 2,324 controls, we evaluated 5,365 SNPs for replication in 3,972 cases and 3,852 controls. Ninety-four SNPs were further evaluated in 5,203 cases and 5,138 controls, and finally the top 22 SNPs were investigated in up to 17,423 additional subjects (7,489 cases and 9,934 controls). SNP rs9485372, near the TGF-β activated kinase (<I>TAB2</I>) gene in chromosome 6q25.1, showed a consistent association with breast cancer risk across all four stages, with a <I>P</I>-value of 3.8×10<SUP>−12</SUP> in the combined analysis of all samples. Adjusted odds ratios (95% confidence intervals) were 0.89 (0.85–0.94) and 0.80 (0.75–0.86) for the A/G and A/A genotypes, respectively, compared with the genotype G/G. SNP rs9383951 (<I>P</I> = 1.9×10<SUP>−6</SUP> from the combined analysis of all samples), located in intron 5 of the <I>ESR1</I> gene, and SNP rs7107217 (<I>P</I> = 4.6×10<SUP>−7</SUP>), located at 11q24.3, also showed a consistent association in each of the four stages. This study provides strong evidence for a novel breast cancer susceptibility locus represented by rs9485372, near the <I>TAB2</I> gene (6q25.1), and identifies two possible susceptibility loci located in the <I>ESR1</I> gene and 11q24.3, respectively.</P><P><B>Author Summary</B></P> <P>Breast cancer is one of the most common malignancies among women worldwide. Genetic factors play an important role in the etiology of breast cancer. The twin cam mixer (TCM), as a general-purpose mixer, shares many attributes in common with 3D industrial mixers, like the internal mixer. We investigated the mixing process in a 2D TCM with two identical isosceles triangular cams rotating at 0.5 rpm. A 2D numerical model coupled with the species transport model was employed to study the influence of cam height-base ratio and eccentricity qualitatively and quantitatively, and both were found to have a significant effect on the mixing behavior of the mixer. Furthermore, a dimensionless parameter, named the modified pressurization coefficient, is put forward to quantify the geometry of the mixer. The logarithmic relationship between the modified pressurization coefficient and the mixing quality was discovered and expected to provide new ideas for establishing the relationship between the geometric parameters of a mixer and its mixing performance.
Development of TREND dynamics code for molten salt reactors

The Molten Salt Reactor (MSR), one of the six advanced reactor types of the 4th generation nuclear energy systems, has many impressive features including economic advantages, inherent safety and nuclear non-proliferation. This paper introduces a system analysis code named TREND, which is developed and used for the steady and transient simulation of MSRs. The TREND code calculates the distributions of pressure, velocity and temperature of single-phase flows by solving the conservation equations of mass, momentum and energy, along with a fluid state equation. Heat structures coupled with the fluid dynamics model is sufficient to meet the demands of modeling MSR system-level thermal-hydraulics. The core power is based on the point reactor neutron kinetics model calculated by the typical Runge-Kutta method. An incremental PID controller is inserted to adjust the operation behaviors. The verification and validation of the TREND code have been carried out in two aspects: detailed code-to-code comparison with established thermal-hydraulic system codes such as RELAP5, and validation with the experimental data from MSRE and the CIET facility (the University of California, Berkeley's Compact Integral Effects Test facility).The results indicate that TREND can be used in analyzing the transient behaviors of MSRs and will be improved by validating with more experimental results with the support of SINAP.
Identification of novel rheumatoid arthritis-associated MiRNA-204-5p from plasma exosomes

Rheumatoid arthritis (RA) is an autoimmune disease characterized by infiltration of immune cells in the synovium. However, the crosstalk of immune cells and synovial fibroblasts is still largely unknown. Here, global miRNA screening in plasma exosomes was carried out with a custom microarray (RA patients vs. healthy controls = 9:9). A total of 14 exosomal miRNAs were abnormally expressed in the RA patients. Then, downregulated expression of exosomal miR-204-5p was confirmed in both the replication (RA patients vs. healthy controls = 30:30) and validation groups (RA patients vs. healthy controls = 56:60). Similar to the findings obtained in humans, a decreased abundance of exosomal miR-204-5p was observed in mice with collagen-induced arthritis (CIA). Furthermore, Spearman correlation analysis indicated that plasma exosomal miR-204-5p expression was inversely correlated with disease parameters of RA patients, such as rheumatoid factor, erythrocyte sedimentation rate, and C-reactive protein. In vitro, our data showed that human T lymphocytes released exosomes containing large amounts of miR-204-5p, which can be transferred into synovial fibroblasts, inhibiting cell proliferation. Overexpression of miR-204-5p in synovial fibroblasts suppressed synovial fibroblast activation by targeting genes related to cell proliferation and invasion. In vivo assays found that administration of lentiviruses expressing miR-204-5p markedly alleviated the disease progression of the mice with CIA. Collectively, this study identified a novel RA-associated plasma exosomal miRNA-204-5p that mediates the communication between immune cells and synovial fibroblasts and can be used as a potential biomarker for RA diagnosis and treatment.
