Cytoplasmic and Nuclear Localization of DcHSP17.7, a Small Heat Shock Protein

The cellular localization of DcHSP17.7, a small heat shock protein (smHSP) from carrot (Daucus carota L.), was studied using green fluorescent protein (GFP) as a marker. A fusion gene construct, the DcHSP17.7 gene fused with the GFP gene, driven by the cauliflower mosaic virus 35S promoter was introduced into potato (Solanum tuberosum L. cv. ‘Desiree’) via Agrobacterium-mediated transformation. Transgenic plants were successfully regenerated and the DcHSP17.7-GFP fusion protein was observed using confocal laser scanning microscopy. Our results showed that DcHSP17.7 is localized in the cytoplasm and nucleus of the cells in leaf and stem tissues. Its general cellular compartmentation did not alter after heat stress (40℃, 4 hours). However, DcHSP17.7 was present at a higher level in the perinuclear region of the cytoplasm than inside the nucleus in the heat-stressed cells of stem tissues. The possible roles of DcHSPI7.7 in enhancement of thermotolerance under heat stress conditions were discussed based on its cellular localization. The integration of the transgene into the genome of transgenic plants and the accumulation of the fusion protein were confirmed by using polymerase chain reaction and immunodetection analyses, respectively.

A simple and efficient protocol was developed for culturing Cu-tolerant and Cu-accumulating plants via pre-adaptation to Cu during plant tissue culture. We induced multiple shoots from begonia (Begonia evansiana Andr.) leaf explants on MS medium supplemented with naphtaieneacetic acid and benzyladenine. After 3 months, small plantlets were transferred to MS medium supplemented with $100{\mu}M\;CuCl_2$ for pre-adaptation to Cu and cultured for 5 months. Then, these plantlets were individually planted in pots containing artificial soil. An additional 500 mg of Cu dissolved in 1/4 strength MS solution was applied to each pot during irrigation over the course of 2 months. We planted pre-adapted and control begonias in soil from the II-Kwang Mine, an abandoned Cu mine in Pusan, Korea, to examine their ability to tolerate and accumulate Cu for phytore-mediation. Pre-adapted begonias accumulated $1,200{\mu}g$ Cu/g dry root tissue over the course of 45 days. On the other hand, non-Cu-adapted controls accumulated only $85{\mu}g$ Cu/g dry root tissue. To enhance Cu extraction, chelating agents, ethylenediamine tetraacetic acid (EDTA)-dipotassiun and pyridine-2,6-dicarboxylic acid (PDA), were applied. While the chelating agents did not enhance accumulation of Cu in the roots of control begonias, EDTA application increased the level of Cu in the roots of pre-adapted begonias twofold (to $2,500{\mu}g$ Cu/g dry root tissue). Because pre-adapted begonias accumulated a large amount of Cu, mainly in their roots, they could be used for phytostabilization of Cu-contaminated soils. In addition, as a flowering plant, begonias can be used to create aesthetically pleasing remediation sites.

DcHsp17.7 is a heat-inducible small heat shock protein (sHSP) in carrot (Daucus carota L.). Moreover, here we report that DcHsp17.7 is constitutively expressed in vegetative and reproductive tissues at defined developmental stages. Immunoblot analysis using a polyclonal antibody raised against DcHsp17.7 showed that the protein was present in non-stressed leaf tissue and gradually accumulated upon heat stress (40℃). In the heat-induced level of DcHsp17.7, there were transient, but reproducible declines, suggesting dynamic expression regulation. The protein was also present in dry and germinating seeds. It is worth noting that unlike most other seed sHSPs, which disappear upon completion of germination, DcHsp17.7 was continuously present in young seedling. In flower bud and open flower, the protein was constitutively expressed. Heat stress increased the amount of DcHsp17.7 in these floral tissues. Under the opposite temperature extreme, cold stress (2℃), DcHsp17.7 was also present in these tissues, but the levels were lower than those under heat stress. Constitutive expression of DcHsp17.7 suggests that it may play physiological role(s) in the absence of thermal stress. Alternatively, DcHsp17.7 in non-stressed tissues may confer advanced protection to sudden temperature changes by immediately enhancing thermotolerance of cells.

히라야마병은 감각 및 중추신경계의 증상 없이 상지의 근위축과 근력약화가 진행하는 경추척수증으로 매우 드문 질환이다. 이에 저자들은 일측 손목 하수와 일측 근력약화, 근위축을 보인 소아에서의 히라야마병 2예를 보고하는 바이다. 첫번째 환자는 16세 남아로 좌측 손목 하수와 함께 동측의 2-5번째 손가락의 떨림이 진행하는 것으로 내원하였고, 신체진찰상 중수수지, 지절간관절의 근력약화, 배측골간근, 소지외전근의 근위축이 확인되었다. 두번째 환자는 14세 남아로 우측 손의 근력약화와 글씨를 바르게 쓰기 어려운 것이 진행하여 내원하였고, 신체진찰상 천지굴근, 모지대립근, 충양근, 척측수근굴근, 심지굴근, 소지굴근, 무지내전근, 배측골간근, 수장측골간근의 근력약화와 무지구근, 중간구근의 근위축이 확인되었다. 두 환자는 특별한 과거력은 없었고, 전해질 및 혈당 검사는 정상이었다. 운동신경전도검사에서 척골신경과 요골신경의 운동신경활동전위진폭이 감소하였고, 척추자기공명영상검사에서 전자는 정상이었으나 후자는 C5-C7의 앞쪽 척수가 약간 비대칭적으로 얇아진 소견이 확인되었다. 두 환자 모두 경추굽힘을 피하도록 하면서 재활치료를 받으면서 증상의 추가적인 진행은 없었다. 히라야마병은 유병률이 높지 않아 쉽게 간과되거나 오진될 수 있으나 조기에 진단하여 반복적인 경추굽힘으로 인해 병이 진행하지 않도록 예방할 수 있는 병이다. 그러므로 소아에서 감각이상 없이 원위부 근력 약화나 근위축을 보일 때 히라야마병의 가능성을 고려하는 것이 중요하겠다. We report two pediatric cases with Hirayama disease-a 16-year-old boy with a left wrist drop and a 14-year-old-boy with weakness and muscle atrophy of right hand. Motor nerve conduction study revealed decreased motor nerve action potential amplitudes in the ulnar nerve and radial nerve of the affected hands. The former patient showed normal magnetic resonance imaging (MRI) of the cervical spine, but the latter showed mild, asymmetric thinning of the anterior spinal cord at levels C5 to C7. Following active rehabilitation and avoidance of neck flexion, no further progression of neurological findings was noticed. These clinical findings were typical of Hirayama disease. We show that timely and accurate diagnosis for Hirayama disease is possible with awareness of disease history, careful physical examination, and the use of neurophysiological studies and MRI studies.
Small Heat Shock Proteins Can Confer Tolerance to Nanomaterial-induced Toxicity

<P>The expression of a small heat shock protein (sHSP) in plants and its possible function in conditions related to nanomaterial exposure were examined. Multiwalled carbon nanotubes (MWCNTs) and silver nanoparticles (AgNPs) induced toxicity that was indicated by the bending and curling of carrot leaf tissues. Both nanomaterials induced the expression of a small heat shock protein in carrot, DcHsp17.7, but reduced the level of a constitutive heat shock cognate 70. To examine the possible function of DcHsp17.7, the coding gene was heterologously expressed in <I>Escherichia coli</I>. Both nanomaterials reduced the viability of <I>E. coli</I> cell lines. However, the transgenic cell line heterologously expressing DcHsp17.7 showed higher levels of cell viability, compared with vector controls, when exposed to MWCNTs and, more notably, to AgNPs. To the best of our knowledge, this is the first study reporting the influence of nanomaterials on the expression of a plant sHSP and its possible function in conferring tolerance to nanomaterial stress.</P>

Horse population has been gradually increasing with the growth of the racing industry in Republic of Korea (ROK). Approximately 22,941 horses including native ponies were raised on 1,142 farms in 2006. Great care has been taken to prevent viral, bacterial and parasitic infections in horses. However, there has been few reports on equine infectious anemia (EIA) in ROK until recently since 1987. Therefore, we conducted a serological survey of EIA in ROK from 2007 through 2009. Using the agar gel immunodiffusion test, a total of 2,601 horses were tested in Foreign Animal Disease Division of Animal, Plant, and Fisheries Quarantine and Inspection Agency. This survey found no serological evidence of EIA presence in ROK. Although our surveillance found no evidence of EIA in ROK during the surveillance period, it is possible that EIA could be introduced into ROK in the near future. Increased cooperation between the government and other agencies, such as horse-racing authorities, is important for both the early detection of the disease and the development of effective veterinary and public health strategies.
