        Changing Representations of Youth: “Youth Films” in the People`s Republic of China

        Youth culture is an indicator of social transformations. In a context where youth cultures have been shaped by globalization, contemporary world cinema increasingly becomes youth-oriented in terms of style, subject, and reception. The present paper aims to trace the development of youth films in the People`s Republic of China since the end of the 1940s within this global context. An examination of how youth films talk to and about young people-and how young people are seen in these films-is a particularly revealing way to explore the profound social and cultural changes that have been taking place in that society. It can also highlight the global-local interactions that have reshaped Chinese life and the ways in which certain developments and influences in youth culture have acquired a distinctive Chinese inflection.

        On the Rooftop: A Study of Marginalized Youth Films in Hong Kong Cinema

        Researchers of contemporary Hong Kong cinema have tended to concentrate on the monumental, metropolitan and/or historical works of such esteemed directors as Wong Kar-Wai, John Woo and Tsui Hark. This paper focuses instead on a number of low-budget films that circulated below the radar of Chinese as well as Western film scholars but were important to local young viewers, i.e. a cluster of films that feature deviant and marginalized youth as protagonists. They are very interesting as evidence of perceived social problems in contemporary Hong Kong. The paper aims to outline some main features of these marginalized youth films produced since the mid-1990s.

        Screening Education: Schools on Film in the People`s Republic of China

        Like many other national and/or semi-national cinemas today, contemporary Chinese film tends to be youth or teen-driven. Recent years have seen a plethora of youth films produced by Chinese filmmakers working in and outside the official studio system. Nevertheless, school film, i.e. those set on school campuses and focusing on education activities within the campus context, has not received sufficient attention despite its being a most definable subgenre of youth films. This paper aims to trace the development of school films in the People`s Republic of China since the 1950s. It also attempts to discuss some reasons why there has been a shortage of school films being made and why the filmic representation of teacher- and/or student-heroes is often stereotyped in the limited number of school films. The paper concludes with an in-depth study of a more recent school film Thirteen Princess Trees (Lu Yue, 2006), which displays some interesting departures from the existing tradition.

        Increased retinoic acid signaling decreases lung metastasis in salivary adenoid cystic carcinoma by inhibiting the noncanonical Notch1 pathway

        MYB-NFIB fusion and NOTCH1 mutation are common hallmark genetic events in salivary gland adenoid cystic carcinoma (SACC). However, abnormal expression of MYB and NOTCH1 is also observed in patients without MYB-NFIB fusion and NOTCH1 mutation. Here, we explore in-depth the molecular mechanisms of lung metastasis through single-cell RNA sequencing (scRNA-seq) and exome target capture sequencing in two SACC patients without MYB-NFIB fusion and NOTCH1 mutation. Twenty-five types of cells in primary and metastatic tissues were identified via Seurat clustering and categorized into four main stages ranging from near-normal to cancer-based on the abundance of each cell cluster in normal tissue. In this context, we identified the Notch signaling pathway enrichment in almost all cancer cells; RNA velocity, trajectory, and sub-clustering analyses were performed to deeply investigate cancer progenitor-like cell clusters in primary tumor-associated lung metastases, and signature genes of progenitor-like cells were enriched in the “MYC_TARGETS_V2” gene set. In vitro, we detected the NICD1-MYB-MYC complex by co-immunoprecipitation (Co-IP) and incidentally identified retinoic acid (RA) as an endogenous antagonist of genes in the “MYC_TARGETS_V2” gene set. Following this, we confirmed that all-trans retinoic acid (ATRA) suppresses the lung metastasis of SACC by correcting erroneous cell differentiation mainly caused by aberrant NOTCH1 or MYB expression. Bioinformatic, RNA-seq, and immunohistochemical (IHC) analyses of primary tissues and metastatic lung tissues from patients with SACC suggested that RA system insufficiency partially promotes lung metastasis. These findings imply the value of the RA system in diagnosis and treatment.

        Tube Voltage, DNA Double-Strand Breaks, and Image Quality in Coronary CT Angiography

        Objective: To evaluate the effects of tube voltage on image quality in coronary CT angiography (CCTA), the estimated radiation dose, and DNA double-strand breaks (DSBs) in peripheral blood lymphocytes to optimize the use of CCTA in the era of low radiation doses. Materials and Methods: This study included 240 patients who were divided into 2 groups according to the DNA DSB analysis methods, i.e., immunofluorescence microscopy and flow cytometry. Each group was subdivided into 4 subgroups: those receiving CCTA only with different tube voltages of 120, 100, 80, or 70 kVp. Objective and subjective image quality was evaluated by analysis of variance. Radiation dosages were also recorded and compared. Results: There was no significant difference in demographic characteristics between the 2 groups and 4 subgroups in each group (all p > 0.05). As tube voltage decreased, both image quality and radiation dose decreased gradually and significantly. After CCTA, γ-H2AX foci and mean fluorescence intensity in the 120-, 100-, 80-, and 70-kVp groups increased by 0.14, 0.09, 0.07, and 0.06 foci per cell and 21.26, 9.13, 8.10, and 7.13 (all p < 0.05), respectively. The increase in the DNA DSB level in the 120-kVp group was higher than those in the other 3 groups (all p < 0.05), while there was no significant difference in the DSBs levels among these latter groups (all p > 0.05). Conclusion: The 100-kVp tube voltage may be optimal for CCTA when weighing DNA DSBs against the estimated radiation dose and image quality, with further reductions in tube voltage being unnecessary for CCTA.

        Circulating MiRNA-21-enriched extracellular vesicles promote bone remodeling in traumatic brain injury patients

        Fracture combined with traumatic brain injury (TBI) is one of the most common and serious types of compound trauma in the clinic and is characterized by dysfunction of cellular communication in injured organs. Our prior studies found that TBI was capable of enhancing fracture healing in a paracrine manner. Exosomes (Exos), as small extracellular vesicles, are important paracrine vehicles for noncell therapy. However, whether circulating Exos derived from TBI patients (TBI-Exos) regulate the prohealing effects of fractures remains unclear. Thus, the present study aimed to explore the biological effects of TBI-Exos on fracture healing and reveal the potential molecular mechanism. TBI-Exos were isolated by ultracentrifugation, and the enriched miR-21-5 p was identified by qRT‒PCR analysis. The beneficial effects of TBI-Exos on osteoblastic differentiation and bone remodeling were determined by a series of in vitro assays. Bioinformatics analyses were conducted to identify the potential downstream mechanisms of the regulatory effect of TBI-Exos on osteoblasts. Furthermore, the role of the potential signaling pathway of TBI-Exos in mediating the osteoblastic activity of osteoblasts was assessed. Subsequently, a murine fracture model was established, and the effect of TBI-Exos on bone modeling was demonstrated in vivo. TBI-Exos can be internalized by osteoblasts, and in vitro, suppression of SMAD7 promoted osteogenic differentiation, whereas knockdown of miR-21-5 p in TBI-Exos strongly inhibited this bone-beneficial effect. Similarly, our results confirmed that preinjection of TBI-Exos led to enhanced bone formation, whereas knockdown of exosomal miR-21-5 p substantially impaired this bone-beneficial effect in vivo.

        Loss Analysis and Air-Cooled Design for a Cascaded Electrical Source Transmitter

        Air-cooling method is adopted on the basis of the requirements for the thermal stability and convenient field use of an electrical source transmitter. The power losses of the transmitter are determined after calculating the losses of the alternating current (AC)-direct current (DC) power supply, the constant-current circuit, and the output circuit. According to the analysis of the characteristics of a heat sink with striped fins and a fan, the engineering calculation expression of the Nusselt number and the design process for air-cooled dissipation are proposed. Experimental results verify that the error between calculated and measured values of the transmitter losses is 12.2%, which meets the error design requirements of less than 25%. Steady-state average temperature rise of the heat sink of the AC-DC power supply is $22^{\circ}C$, which meets the design requirements of a temperature rise between $20^{\circ}C$ and $40^{\circ}C$. The transmitter has favorable thermal stability with 40 kW output power.

      • Expression and Function of GSTA1 in Lung Cancer Cells

        Glutathione S-transferase A1 (GSTA1) appears to be primarily involved in detoxification processes, but possible roles in lung cancer remain unclear. The objective of this study was to investigate the expression and function of GSTA1 in lung cancer cells. Real-time PCR and Western blotting were performed to assess expression in cancer cell lines and the normal lung cells, then verify the A549 cells line with stable overexpression. Localization of GSTA1 proteins was assessed by cytoimmunofluorescence. Three double-strand DNA oligoRNAs (SiRNAs) were synthesized prior to being transfected into A549 cells with Lipofectamine 2000, and then the most efficient SiRNA was selected. Expression of the GSTA1 gene in the transfected cells was determined by real-time PCR and Western blotting. The viability of the transfected cells were assessed by MTT. Results showed that the mRNA and protein expression of A549 cancer cells was higher than in MRC-5 normal cells. Cytoimmunofluorescence demonstrated GSTA1 localization in the cell cytoplasm and/or membranes. Transfection into A549 cells demonstrated that down-regulated expression could inhibit cell viability. Our data indicated that GSTA1 expression may be a target molecule in early diagnosis and treatment of lung cancer.

        Alum sludge conditioning with ferrous iron/peroxymonosulfate oxidation: Characterization and mechanism

        Alum sludge produced by drinking water plants needs to be conditioned and dewatered before final disposal. In this study, a novel ferrous iron/peroxymonosulfate (PMS) oxidation process was employed to enhance alum sludge dewaterability. The effect of oxidative sulfate radicals generated by Fe2+ activated HSO5  on alum sludge was studied. The results showed that the optimal conditioning conditions for addition of Fe2+ and PMS were 0.5 g/g and 0.1 g/g TSS, respectively. Meanwhile, the capillary suction time (CST) and specific resistance to filtration (SRF) of alum sludge was reduced by 66% and 88%. Also found was that the absolute value of the zeta potential increased and the particle size decreased in alum sludge after Fe2+-PMS conditioning, which indicated that oxidative sulfate radicals destroyed the floc structure of alum sludge and smaller particles were generated. At the same time, the water contained in sludge flocs was released and enhanced sludge dewaterability, while leaching of aluminum ions also characterized decomposition of alum sludge.
