We established an efficient transformation method for thermophile Geobacillus kaustophilus HTA426 using conjugative transfer from Escherichia coli of host-mimicking plasmids that imitate DNA methylation of strain HTA426 to circumvent its DNA restriction barriers. Two conjugative plasmids, pSTE33T and pUCG18T, capable of shuttling between E. coli and Geobacillus spp., were constructed. The plasmids were first introduced into E. coli BR408, which expressed one inherent DNA methylase gene (dam) and two heterologous methylase genes from strain HTA426 (GK1380-GK1381 and GK0343-GK0344). The plasmids were then directly transferred from E. coli cells to strain HTA426 by conjugative transfer using pUB307 or pRK2013 as a helper plasmid, pUCG18T was introduced very efficiently (transfer efficiency, 10-5-10-3 recipient-1), pSTE33T showed lower efficiency (10-7-10-6 recipient-1) but had a high copy number and high segregational stability. Methylase genes in the donor substantially affected the transfer efficiency, demonstrating that the host-mimicking strategy contributes to efficient transformation. The transformation method, along with the two distinguishing plasmids, increases the potential of G. kaustophilus HTA426 as a thermophilic host to be used in various applications and as a model for biological studies of this genus. Our results also demonstrate that conjugative transfer is a promising approach for introducing exogenous DNA into thermophiles.

DNA methylation in Streptomyces griseus IFO 13350 was analyzed by high-performance liquid chromatographic analysis and bisulfite-based analysis to reveal two methylation sites, 5`-GC5mCGGC-3` and 5`-GAG5mCTC-3`. The methylation was reconstituted in Escherichia coli by simultaneous expression of S. griseus SGR4675 and S. achromogenes M.SacI. The E. coli cells produced plasmids that mimicked the methylation profile of S. griseus DNA, which was readily introduced into S. griseus. The results of this study raise the possibility of a promising approach to establish efficient transformation in several streptomycetes.
Analysis of the Nodal Structure for a Nonintegrable Two-Dimensional Quartic Oscillator

We extend studies of the nodal structure in quantum-mechanical wave functions to a twodimensional quartic oscillator and investigate the transition from integrable to chaotic dynamics. We find that the number of nodal domains is drastically reduced as the dynamics of the system changes from integrable to nonintegrable and then gradually increases as the system becomes chaotic. We also calculate the area distribution of nodal domains and study the emergence of a power-law behavior with a Fisher exponent in the chaotic limit.
Readiness Estimation for a Take-Over Request in Automated Driving on an Expressway

Automated driving is attracting attention as a solution to road traffic problems. At Level 3, a take-over request (TOR) is issued to transfer driving operations from the system to a driver when it is unable to continue. In such cases, the driver must be monitored to ensure a proper takeover of the driving operations. This study aims to measure drivers’ brain activity before and after the TOR by analyzing time-series signals of brain activity with machine learning algorithms. We developed driving scenarios with a TOR trigger on a rainy expressway at night. We used a portable functional near-infrared spectroscopy (fNIRS) device to measure cerebral blood oxygenation changes (ΔHbO) at the frontal pole. We used a long short-term memory (LSTM) network on this data for time-series learning and prediction after multivariate and multilayering modifications to improve accuracy. We conducted driving questionnaires beforehand and used two classification methods to categorize subjects into several groups with similar driving characteristics. Experimental results of a ΔHbO drop revealed that brain activity tended to decrease during automated driving. Moreover, success in obstacle avoidance and mean squared error (MSE) for each driver group demonstrated that the behavior toward an obstacle after the TOR trigger influenced changes in brain activity.

When distributed generators such as photovoltaic (PV) systems spread widely in distribution networks, it is important to detect islanding caused by power system fault for ensuring electrical safety. We developed the nonlinear magnetizing characteristics model of a pole transformer for analyzing islanding-prevention of PV system. Basic experiments of the islanding-detection were conducted by using the RLC resonant circuit for verification of the developed model. Analytical results by the proposed model agreed well with experimental results.

In this paper, we propose an extraction method of ride comfort according to the difference of the tire by electroencephalogram (EEG). Recently, the sensory evaluation is used for development of the product. However, the sensory evaluation by a general user who doesn’t have a clear standard is unstable. Moreover, ride comfort depends on the sensibility and the preference of the individual. It is considered that these psychological changes influence the brain that rules center of sensation. Therefore, we pay attention to the EEG. We use the EEG that it is possible to measure it simply in the brain function measurement technique as an objective evaluation. In this study, the feature of the EEG during the driving is extracted by the factor analysis (FA). Moreover, we investigate the correlation of the subjective evaluation and the EEG features. From the result, the EEG features and subjective evaluation features has correlation. Thus, the effectiveness of the proposed method as an objective evaluation method was shown.

When large penetration of the distributed generators (DGs) such as photovoltaic (PV) systems is growing up in grid system, it is important to quickly prevent islanding caused by power system fault to ensure electrical safety. We propose a novel active method for islanding prevention by harmonic injection synchronized with the exciting current harmonics of the pole transformer to avoid mutual interference between active signals. We confirm the validity of the proposed method by performing the basic tests of islanding by using a current source superimposed the harmonic active signal. Further, we carry out the simulation using PSCAD/EMTDC, and verify the fast islanding detection.
