        Prediction of itching diagnostic marker through RNA sequencing of contact hypersensitivity and skin scratching stimulation mice models

        Pruritus (itching) is classically defined as an unpleasant cutaneous sensation that leads to scratching behavior. Although the scientific criteria of classification for pruritic diseases are not clear, it can be divided as acute or chronic by duration of symptoms. In this study, we investigated whether skin injury caused by chemical (contact hypersensitivity, CHS) or physical (skin-scratching stimulation, SSS) stimuli causes initial pruritus and analyzed gene expression profiles systemically to determine how changes in skin gene expression in the affected area are related to itching. In both CHS and SSS, we ranked the Gene Ontology Biological Process terms that are generally associated with changes. The factors associated with upregulation were keratinization, inflammatory response and neutrophil chemotaxis. The Kyoto Encyclopedia of Genes and Genomes pathway shows the difference of immune system, cell growth and death, signaling molecules and interactions, and signal transduction pathways. Il1a, Il1b and Il22 were upregulated in the CHS, and Tnf, Tnfrsf1b, Il1b, Il1r1 and Il6 were upregulated in the SSS. Trpc1 channel genes were observed in representative itching-related candidate genes. By comparing and analyzing RNA-sequencing data obtained from the skin tissue of each animal model in these characteristic stages, it is possible to find useful diagnostic markers for the treatment of itching, to diagnose itching causes and to apply customized treatment.

        Fast and Efficient Isolation of Mouse Bone Marrow-Derived Mesenchymal Stem Cells by Using a Biocompatible Polymer

        Mesenchymal stem cells (MSCs) differentiate into bone, fat, cartilage, tendon, and other organ progenitor cells. The rarity of MSCs in bone marrow necessitates fast and efficient isolation and/or in vitro expansion prior to clinical and biomedical applications. Previously, we reported that UV-exposed diphenylamino-s-triazine bridged p-phenylene vinylene (DTOPV-UV) with a hydrophilic and negative surface-containing carboxyl group is highly biocompatible and provides a substrate for efficient human bone marrow-derived MSC attachment. In this study, we applied this polymeric film to early adhesion and enrichment of MSCs from mouse bone marrow. With its high protein- binding capacity, DTOPV-UV film was more efficient in early capture of adherent bone marrow cells than conventional tissue culture polystyrene (TCPS). Cell binding to DTOPV-UV reached full capacity within 1 hr, whereas cell attachment to TCPS gradually increased over time. The isolated and culture-expanded MSCs from mouse bone marrow displayed typical morphology, phenotype, and differentiation into osteoblasts, adipocytes, and chondrocytes. Here, we demonstrate a novel method for isolating MSCs from mouse bone marrow using a biocompatible polymer. This method will aid the development of rapid and efficient isolation and in vitro expansion protocols for rare adherent cells.

        거주자의 유형을 고려한 공동주택 저층부 특화디자인에 관한 연구

        (Background and Purpose) Lower part of an apartment possesses an unique characteristic that it is placed adjacent to both upper ground and underground. Such a nature allows us to design the floor in various ways benefited from its spatial potential. In this thesis, we first analyze determining factors of lower part of an apartment in an effort to offer an specialized design. Finally, we suggest a specialized design for lower part of an apartment which suits various types of residents. (Method)In this study, we come up with a new designing methodology for low part of an apartment by analyzing relevant past studies and applying derived key factors. We intend to design a residential space for each type of lower part of an apartment dwellers according to their life cycle. (Result)We provide specialized designs for dwellers in each of four types within the life cycle with various spatial compositions; single families in family independency period, newly married couples with young children in family formation period, families with grown-up children in family expansion and elderly households in family reduction period. We further discuss limitations to this study in order to provide our findings as a basis reference to future specialized designs for lower part of an apartment.
