RH-531을 施用한 湖南野山開發地區産大豆中의 Globulin 및 Glutelin의 加水分解에 依해서 生成되는 아미노酸의 組成
The crude globulin and crude glutelin were fractionated from the RH-531 treated soybeans and after the hydrolysis of these proteins by 6N-HCl solution their amino acids were separated and identified by the thin layer chromatography. The results are summarized as follows: 1. The hydrolysis of globulin and glutelin prepared from six kinds of RH-531 treated soybeans afforded the mixtures of 15 kinds of amino acids, which were identified by thin layer chromatography, respectively, as aspartic acid, lysine, glutamic acid, serine, glycine, arginine, threonine, alanine, histidine, tyrosine, valine, methionine, leucine, proline and phenylalanine. 2. The yield of crude globulin was highest in sample 3 and except the control the yield of crude glutelin was also highest in sample 3. And the total yield of NaCl-soluble fraction and NaOH-soluble fraction was also highest in sample 3. 3. In the one-dimensional chromatogram, depending on the increase of RH-531 amount, the contents of the globulin-compssing amino acids increased, while the contents of glutelin-composing amino acids decreased. 4. In the two-dimensional chrematogram the change of the composition of the amio acids of globulin and glutelin was hardly observed but, sometimes, methionine and histidine were not detected. On the other hand, the chang in the contents of amino acids of these two proteins was observable and was similar to the results reported by another authors.
Interferon 및 gamma-globulin이 實驗的 Pneumocystis carinii 肺炎의 治療에 미치는 影響
This study was performed to obtain the additional informations on the therapeutic effects of interferon and gamma-globulin in experimental Pneumocyatis carinii pneumonia. Total 135 female BALB/c mice were used. The mice were administered an immunosuppressive regimen of 2.5 mg of cortisone acetate injected subcutaneously twice weekly, a low(8w) protein diet, and tetracycline(1㎎/㎖) for 11 weeks. At 9 weeks, the mice were randomly divided into control and five different experimental groups(15 mice per group). Control group received no anti-P. carinii therapy, but experimental groups administered trimethoprim-sulfamethoxazole(TMP-SMZ; 10-50 mg/mouse/day), recombinant mouse interferon-gamma(rIFN-r: 5×10⁴units/mouse/day) and mouse gamma-globulin(r-globulin: 20 ㎎/mouse/day) for 3 weeks. The 5 mice were sacrified at weekly intervals to study anti-P. carinii effects in control and experimental group. Evaluations of drug therapy were based on body weights of mice, histopathologic and electron micrascopic findings of lungs, and number of P. carinii cysts by Gomori's methenamine silver stain. Body weights of mice were significantly increased in combination therapy of TWP-SMX with rIFN-r or r-globulin(p<0.05), and treatment with TMP-SMZ alone(p<0.05). But it was little effect in the treatment with r-globulin alone. Histopathologic findings of P. carinii pneumonia were best improved in combination therapy of TMP-SMZ with rINF-r. Treatment with rINF-r alone was similar histopathologic findings as compared with treatment with TMP-SMZ. In electron microscopic findings of P. carinii pneumonia, the number of trophozoites and cysts were reduced in treatment with either TMP-SMZ or rIFN-r as compared with control group. And most of cysts were empty-cyst form or containing one or two intracystic bodies. Treatment with either TMP-SWZ or rIFN-r significantly reduced the number of cysts in the P. carinii pneumonia(p<0. 05) But r-globulin alone was ineffective in eradicating P. carinii cysts . From this study, treatment with IFN-r caused a significant reduction in the number of cysts in P. carinii pneumonia in the lung, and combination therapy of TMP-SWZ with IFN-r were synergistic effects in the treatment of P. carinii pneumonia in mice. But further study will be needed to understand the action mechanism of IFN-y in the treatment of P. carinii pneumonia.
