        Esophageal Nocturnal Baseline Impedance and Post-reflux Swallow-induced Peristaltic Wave Index in Identifying Proton Pump Inhibitor-refractory Non-erosive Reflux Disease

        Background/Aims Esophageal mean nocturnal baseline impedance (MNBI) levels and post-reflux swallow-induced peristaltic wave (PSPW) index could increase the diagnostic value of 24-hour multichannel intraluminal impedance and pH monitoring in patients with gastroesophageal reflux disease. This study aims to compare the MNBI and PSPW index in patients with no evidence of erosive reflux disease. Methods Impedance-pH monitoring tracings from 70 patients, 50 with non-erosive reflux disease (NERD) and 20 with functional heartburn (FH), were reviewed. According to proton pump inhibitors (PPI) treatment response, NERD patients were divided into NERD/PPI responders and NERD/PPI nonresponders. MNBI, PSPW index, and intercellular spaces were measured and compared among each group. Results MNBI values and PSPW index were lower in NERD patients than in FH (P < 0.01 and P < 0.05, respectively). MNBI positively correlated with PSPW index (r = 0.525, P < 0.001). NERD/PPI responders had lower MNBI values and PSPW index compared to NERD/PPI nonresponders (both P < 0.01). MNBI and PSPW index distinguished NERD from FH patients with an area under the curve of 0.914 and 0.677, respectively. Wider intercellular space could be identified in patients with NERD (P < 0.01). Conclusion MNBI and PSPW index may differentiate NERD from FH patients and relate to PPI treatment efficacy in patients with NERD. (J Neurogastroenterol Motil 2021;27:525-532)

        Gamma linolenic acid suppresses hypoxia-induced proliferation and invasion of non-small cell lung cancer cells by inhibition of HIF1α

        Background The main therapies for cancer often results in many side effects and drug resistance. Gamma linolenic acid (GLA) is a kind of natural reagent with negligible cytotoxicity. Objective This work aims at detecting whether GLA possesses anti-cancer activity in NSCLC cells and elucidating the potential molecular mechanism. Methods Cytotoxicity of GLA was evaluated by MTT assay and soft agar colony formation method. Immunoblotting analysis examined the effect of GLA on protein expressions of cell proliferation markers (e.g., PCNA, Ki-67 and MCM2), pro-survival protein bcl-2, apoptosis-associated proteins (e.g., bax and cleaved caspase 3), HIF1α and VEGF. Wound healing assay and transwell invasion assay were performed to test the effect of GLA on hypoxia-induced cell migration and invasion. Cell transfection was used to overexpress HIF1α followed by the treatment of GLA to test the effect of HIF1α overexpression on the tumoricidal activity of GLA in NSCLC cell lines. Results MTT and soft agar colony formation tests showed that GLA dose-dependently suppressed cell proliferation in both Calu-1 and SK-MES-1 cell lines. Immunoblotting analysis demonstrated that GLA suppressed protein expressions of PCNA, Ki-67, MCM2 and bcl-2, while GLA induced bax and cleaved caspase 3 expressions. Wound healing assay and transwell invasion assay revealed that GLA was very effective on the inhibition of NSCLC cell migration and invasion. Immunoblotting analysis and cell transfection method indicated that GLA inhibited hypoxia-induced cell proliferation and invasion by suppressing HIF1α-VEGF pathway. Conclusion GLA suppresses hypoxia-induced proliferation and invasion of NSCLC cells by inhibition of HIF1α pathway in vitro.

        Digital image tamper detection by analyzing the statistical properties of the image content of the authenticity of the digital image, the scene authenticity and integrity of authentication methods, namely determine whether the image is the original image, whether the image is real and whether it contains other secret information image, these techniques is the digital image forensics. Digital image forensics technology is dependent on the basis of the original carrier may be divided into active and passive two technologies forensics evidence. This paper generated image detection research for digital image forensics scene authenticity certification in natural images and computer. Combining the results of recent research, based on systematic analysis of the differences between two types of image generation, high-end features for wavelet conducted a detailed study of the effectiveness, from the validity of the characteristic data, proposed several general-purpose computer generating an image detection algorithm. Compared with the previous detection algorithm, strong stability of the proposed algorithm, computational complexity is low, and has easy implementation.

        Overexpression of a maize MYB48 gene confers drought tolerance in transgenic arabidopsis plants

        MYBs are a vital family of transcription factorsthat play critical roles in plant development and stressresponse. However, knowledge concerning the functions ofMYBs in the non-model plants is still limited. In this study,we isolated a R1-type MYB gene from maize (Zea mays L.),designated as ZmMYB48. Quantitative RT-PCR analysisdemonstrated that ZmMYB48 expression was induced bydrought and ABA treatments. Subcellular localization analysisrevealed that ZmMYB48 protein was targeted to the nucleusin tobacco leaf epidermal cells. Transactivation assay in yeastdemonstrated that ZmMYB48 had transcriptional activationability. Heterologous overexpression of ZmMYB48 inArabidopsis remarkably improved plant tolerance to droughtstress, as determined through physiological analyses ofsurvival rate, relative water content, malonaldehyde content,relative electrolyte leakage and proline content. Moreover,overexpression of ZmMYB48 enhanced the expression ofstress/ABA-responsive genes such as P5CS1, RD22, RD29Band ABI1. In addition, ZmMYB48-overexpressing plantsaccumulated higher content of ABA than WT plants underdrought stress. These results demonstrate that ZmMYB48might act as a positive regulator that participates in thedrought stress response through ABA signalling.

        Machining characteristics of USV-MF complex assisted WEDM-LS based on multi-physical coupling

        Ultrasonic vibration (USV) and magnetic field (MF) complex assisted techniques are combined with low speed wire electrical discharge machining (USV-MF complex assisted WEDM-LS) to improve the machining performance of conventional WEDM-LS. Firstly, the simulation results of multi-physical coupling simulation model show that under the influence of ultrasonic vibration, the working fluid fluidity is greatly enhanced and its temperature is significantly decreased. Then, a three-phase flow model of bubble, working fluid and debris in the machining area is established to investigate the effects of ultrasonic vibration on bubble and debris, and the process of bubble rupture is simulated. The simulation results show that with the increase of ultrasonic frequency, the movement of bubble and debris becomes fiercer and fiercer, when ultrasonic frequency is 30 kHz and ultrasonic amplitude is 10 μm, the bubble is collapsed. Finally, comparison of the experimental results of conventional WEDM-LS, USV assisted WEDM-LS, MF assisted WEDM-LS and USV-MF complex assisted WEDM-LS in machining TiNi-01 shape memory alloy reveals that the ratio of normal discharge and the material removal rate are both increased, the workpiece surface roughness value is decreased and the workpiece surface quality is improved in the process of USV-MF complex assisted WEDM-LS.

        IL-11 promotes the treatment efficacy of hematopoietic stem cell transplant therapy in aplastic anemia model mice through a NF-κB/microRNA-204/thrombopoietin regulatory axis

        Hematopoietic stem cell (HSC) transplantation could be of therapeutic value for aplastic anemia (AA) patients, and immunosuppressants may facilitate the efficiency of the procedure. As anti-inflammatory cytokine interleukin-11 (IL-11) has a thrombopoietic effect, its use in cases of chronic bone marrow failure, such as AA, has been proposed to induce HSC function. However, the putative mechanisms that may support this process remain poorly defined. We found that decreased miR-204-5p levels were coincident with increased proliferation in mouse HSCs following exposure to IL-11 in vitro. Through inhibiting NF-кB activity, miR-204-5p repression was demonstrated to be a downstream effect of IL-11 signaling. miR-204-5p was shown to directly target thrombopoietin (TPO) via sequence-dependent 3′-UTR repression, indicating that this microRNA-dependent pathway could serve an essential role in supporting IL-11 functions in HSCs. Increased TPO expression in HSCs following IL-11 exposure could be mimicked or blocked by inhibiting or overexpressing miR-204-5p, respectively. Consistent with these in vitro findings, IL-11 promoted HSC engraftment in a mouse model of AA, an effect that was attenuated in cells overexpressing miR-204-5p. The reduction in miR-204-5p levels is an integral component of IL-11 signaling that may play an essential role in treating AA.

        Original Article : The levels of liver enzymes and atypical lymphocytes are higher in youth patients with infectious mononucleosisthan in preschool children

        Background/Aims: Infectious mononucleosis (IM) is the clinical presentation of primary infection with Epstein-Barr virus. Although the literature contains a massive amount of information on IM, most of this is related specifically to only children or adults separately. In order to distinguish any differences between preschool children and youth patients, we retrospectively analyzed their demographic and clinical features. Methods: Records of patients hospitalized from December 2001 to September 2011 with a diagnosis of IM were retrieved from Peking University First Hospital, which is a tertiary teaching hospital in Beijing. The demographic data and clinical characteristics were collected. Results: IM was diagnosed in 287 patients during this 10-year period, with incidence peaks among preschool children (≤7 years old, 130/287, 45.3%) and youth patients (>15 and <24 years old, 101/287, 35.2%). Although the complaints at admission did not differ between these two patient groups, the incidence of clinical signs (tonsillopharyngitis, lymphadenopathy, hepatomegaly, and edema of the eyelids) was much higher in preschool children. The incidence of liver lesion and percentage of atypical lymphocytes were significantly higher in the youth group (P<0.001), and the average hospital stay was longer in this group. Pneumonia was the most common complication, and there was no case of mortality. Conclusions: The incidence of IM peaks among preschool children and youth patients in Beijing, China. The levels of liver enzymes and atypical lymphocytes increase with age.

        An Improved Level Set Method to Image Segmentation Based on Saliency

        In order to improve the edge segmentation effect of the level set image segmentation and avoid the influence ofthe initial contour on the level set method, a saliency level set image segmentation model based on local Renyientropy is proposed. Firstly, the saliency map of the original image is extracted by using saliency detectionalgorithm. And the outline of the saliency map can be used to initialize the level set. Secondly, the local energyand edge energy of the image are obtained by using local Renyi entropy and Canny operator respectively. Atthe same time, new adaptive weight coefficient and boundary indication function are constructed. Finally, thelocal binary fitting energy model (LBF) as an external energy term is introduced. In this paper, the contrastexperiments are implemented in different image database. The robustness of the proposed model forsegmentation of images with intensity inhomogeneity and complicated edges is verified.

        Volunteering at the Olympic and Youth Olympic Games: More Than a Distant Memory?

        In recent years, cities have shown an interest in using the hosting of sport mega-events as a leverageable resource in realizing long-term impacts for individual volunteers. However, previous research has provided limited evidence of the realization of antici- pated impacts in the post-event period. This study focuses on the long-term impacts of Olympic and Youth Olympic volunteering in terms of memory, attitudes, and behaviors. Data were collected from a sample of 353 volunteers who have been involved with the 2008 Beijing Olympic and Paralympic Games, the 2010 Singapore Youth Olympic Games, and the 2014 Nanjing Youth Olympic Games. Results show that volunteers can easily access their Olympic and Youth Olympic memory, with vivid, emotional, and visual details, while the overall tone of their volunteer mem- ory is positive over a long period. Volunteering at the Olympic and Youth Olympic Games can be understood as a positive life- changing event and participants have reported high scores on the attitudes in terms of the interest in the Olympic Movement. Findings reveal that the sharing of the memory by volunteers and the time perspective have been found to contribute significantly to broader impacts in terms of attitudes about the Olympic Movement and subsequent volunteer behaviors. Implications for sport mega-event volunteer management are discussed.
