Subpopulations of extracellular vesicles and their therapeutic potential

<P><B>Abstract</B></P> <P>Extracellular vesicles (EVs), such as exosomes and microvesicles, have over the last 10–15 years been recognized to convey key messages in the molecular communication between cells. Indeed, EVs have the capacity to shuttle proteins, lipids, and nucleotides such as RNA between cells, leading to an array of functional changes in the recipient cells. Importantly, the EV secretome changes significantly in diseased cells and under conditions of cellular stress. More recently, it has become evident that the EV secretome is exceptionally diverse, with many different types of EVs being released by a single cell type, and these EVs can be described in terms of differences in density, molecular cargos, and morphology. This review will discuss the diversity of EVs, will introduce some suggestions for how to categorize them, and will propose how EVs and their subpopulations might be used for very different therapeutic purposes.</P>

<P><B>Zwei Zinkphthalocyanin‐Photosensibilisatoren</B> mit verschiedenen Phosphinsäure‐Ankergruppen (siehe Schema) wurden synthetisiert. Solarzellen mit diesen Verbindungen verfügen über eine Photostromdichte von (7.6±0.2) mA cm<SUP>−2</SUP> bei geschlossenem Stromkreis, eine Spannung von (559±30) mV bei offenem Stromkreis und einen Füllfaktor von 0.76±0.03; dies entspricht einem Gesamtwirkungsgrad von 3.24 % unter 1 sun.</P>

<P><B>ABSTRACT</B></P><P>Cells secrete extracellular RNA (exRNA) to their surrounding environment and exRNA has been found in many body fluids such as blood, breast milk and cerebrospinal fluid. However, there are conflicting results regarding the nature of exRNA. Here, we have separated 2 distinct exRNA profiles released by mast cells, here termed high-density (HD) and low-density (LD) exRNA. The exRNA in both fractions was characterized by microarray and next-generation sequencing. Both exRNA fractions contained mRNA and miRNA, and the mRNAs in the LD exRNA correlated closely with the cellular mRNA, whereas the HD mRNA did not. Furthermore, the HD exRNA was enriched in lincRNA, antisense RNA, vault RNA, snoRNA, and snRNA with little or no evidence of full-length 18S and 28S rRNA. The LD exRNA was enriched in mitochondrial rRNA, mitochondrial tRNA, tRNA, piRNA, Y RNA, and full-length 18S and 28S rRNA. The proteomes of the HD and LD exRNA-containing fractions were determined with LC-MS/MS and analyzed with Gene Ontology term finder, which showed that both proteomes were associated with the term extracellular vesicles and electron microscopy suggests that at least a part of the exRNA is associated with exosome-like extracellular vesicles. Additionally, the proteins in the HD fractions tended to be associated with the nucleus and ribosomes, whereas the LD fraction proteome tended to be associated with the mitochondrion.</P><P>We show that the 2 exRNA signatures released by a single cell type can be separated by floatation on a density gradient. These results show that cells can release multiple types of exRNA with substantial differences in RNA species content. This is important for any future studies determining the nature and function of exRNA released from different cells under different conditions.</P>

MEASUREMENTS OF THE CORONAL ACCELERATION REGION OF A SOLAR FLARE

<P>The Reuven Ramaty High Energy Solar Spectroscopic Imager (RHESSI) and the Nobeyama Radioheliograph (NoRH) are used to investigate coronal hard X-ray and microwave emissions in the partially disk-occulted solar flare of 2007 December 31. The STEREO mission provides EUV images of the flare site at different viewing angles, establishing a two-ribbon flare geometry and occultation heights of the RHESSI and NoRH observations of similar to 16 Mm and similar to 25 Mm, respectively. Despite the occultation, intense hard X-ray emission up to similar to 80 keV occurs during the impulsive phase from a coronal source that is also seen in microwaves. The hard X-ray and microwave source during the impulsive phase is located similar to 6 Mm above thermal flare loops seen later at the soft X-ray peak time, similar in location to the above-the-loop-top source in the Masuda flare. A single non-thermal electron population with a power-law distribution (with spectral index of similar to 3.7 from similar to 16 keV up to the MeV range) radiating in both bremsstrahlung and gyrosynchrotron emission can explain the observed hard X-ray and microwave spectrum, respectively. This clearly establishes the non-thermal nature of the above-the-loop-top source. The large hard X-ray intensity requires a very large number (>5 x 10(35) above 16 keV for the derived upper limit of the ambient density of similar to 8 x 10(9) cm(-3)) of suprathermal electrons to be present in this above-the-loop-top source. This is of the same order of magnitude as the number of ambient thermal electrons. We show that collisional losses of these accelerated electrons would heat all ambient electrons to superhot temperatures (tens of keV) within seconds. Hence, the standard scenario, with hard X-rays produced by a beam comprising the tail of a dominant thermal core plasma, does not work. Instead, all electrons in the above-the-loop-top source seem to be accelerated, suggesting that the above-the-loop-top source is itself the electron acceleration region.</P>

<P>In this study, we examine the cross‐cultural differences in human resource (HR) managers’ beliefs in effective HR practices by surveying HR practitioners in Finland (N = 86), South Korea (N = 147), and Spain (N = 196). Similar to previous studies from the United States, the Netherlands, and Australia, there are large discrepancies between HR practitioner beliefs and research findings, particularly in the area of staffing. In addition, we find that interpersonal‐oriented aspects of HR practices tend to be more culturally bound than technical‐oriented aspects of HR practices. We interpret the differences using Hofstede's cultural dimensions (Power Distance, Individualism versus Collectivism, Masculinity versus Femininity, Long‐Term Orientation versus Short‐Term Orientation, and Uncertainty Avoidance). We discuss the overall nature of the science‐practice gap in HR management, and the implications for evidence‐based management. © 2014 Wiley Periodicals, Inc.</P>
MEGAMASER DISKS REVEAL A BROAD DISTRIBUTION OF BLACK HOLE MASS IN SPIRAL GALAXIES

<P>We use new precision measurements of black hole (BH) masses from water megamaser disks to investigate scaling relations between macroscopic galaxy properties and supermassive BH mass. The megamaser-derived BH masses span 10(6)-10(8) M-circle dot, while all the galaxy properties that we examine (including total stellar mass, central mass density, and central velocity dispersion) lie within a narrower range. Thus, no galaxy property correlates tightly with M-BH in similar to L* spiral galaxies as traced by megamaser disks. Of them all, stellar velocity dispersion provides the tightest relation, but at fixed sigma* the mean megamaser M-BH are offset by -0.6 +/- 0.1 dex relative to early-type galaxies. Spiral galaxies with non-maser dynamical BH masses do not appear to show this offset. At low mass, we do not yet know the full distribution of BH mass at fixed galaxy property; the non-maser dynamical measurements may miss the low-mass end of the BH distribution due to an inability to resolve their spheres of influence and/or megamasers may preferentially occur in lower-mass BHs.</P>

<P>A π-extended tetrathiafulvalene-boradiazaindacene chimera, ex-TTF-BODIPY, has been prepared. The resulting system undergoes sequential one-electron oxidations, allowing access to both the mono-oxidized radical cationic and dicationic states. Revealing community structures by ensemble clustering using group diffusion

<P><B>Abstract</B></P> <P>We propose an ensemble clustering approach using group diffusion to reveal community structures in data. We represent data points as a directed graph and assume each data point belong to single cluster membership instead of multiple memberships. The method is based on the concept of ensemble group diffusion with a parameter to represent diffusion depth in clustering. The ability to modulate the diffusion-depth parameter by varying it within a certain interval allows for more accurate construction of clusters. Depending on the value of the diffusion-depth parameter, the presented approach can determine very well both local clusters and global structure of data. At the same time, the ability to combine single outcomes of the method results in better cluster segmentation. Due to this property, the proposed method performs well on data sets where other conventional clustering methods fail. We test the method with both simulated and real-world data sets.
