      • Viscum Album Var Hot Water Extract Mediates Anti-cancer Effects through G1 Phase Cell Cycle Arrest in SK-Hep1 Human Hepatocarcinoma cells

        Viscum album var (VAV) also known as mistletoe, has long been categorized as a traditional herbal medicine in Asia. In addition to its immunomodulating activities, mistletoe has also been used in the treatment of chronic hepatic disorders in China and Korea. There are numerous reports showing that VAV possesses anti-cancer effects, however influence on human hepatocarcinoma has never been elucidated. In the present study, hot water extracts of VAV was evaluated for its potential anti-cancer effect in vitro. SK-Hep1 cells were treated with VAV (50-400ug/ml) for both 24 and 48 hours then cell viability was measured by cell counting kit-8 (CCK-8). Flow cytometry analysis was used to measure the proportion of SK-Hep1 in the different stages of cell cycle. RT-PCR and Western blot analysis were conducted to measure expression of cell cycle arrest related genes and proteins respectively. VAV dose dependently inhibited the proliferation of SK-Hep1 cells without any cytotoxicity with normal Chang liver cell (CCL-13). Flow cytometry analysis showed that VAV extract inhibited the cell cycle of SK-Hep1 cells via G1 phase arrest. RT-PCR and Western blot analysis both revealed that cyclin dependent kinase 2 (Cdk2) and cyclin D1 gene expression were significantly down regulated while p21 was upregulated dose dependently by VAV treatment. Combined down regulation of Cdk2, Cyclin D1 and up regulation of p21 can result in cell death. These results indicate that VAV showed evidence of anti-cancer activity through G1 phase cell cycle arrest in SK-Hep1 cells.

        This study focused on the significant differences in choosing a destination brought by the socio-demographics of DLSU-D employees using a theoretical framework formulated in relation to Push-Pull theory. Unlike other studies, this considered all the demographics in examining its differences. Through Analysis of Variance and t-test, income and religion were known to show a significant difference in destination selection. Moreover, the results reveal that international destinations appeal to the respondents the most. The demographics that brought a significant difference in choosing a destination were religion and trip expenditure.

        <P><B>Abstract</B></P> <P>In this study, novel carbon dots were synthesized from polyurethane foam via a one-step pyrolysis method. Polyurethane foam is an interesting precursor for carbon dot synthesis because: (1) polyurethane foam waste generation is alarmingly high and (2) polyurethane is rich in nitrogen content which is important for producing highly photoluminescent carbon dots. The carbon dot synthesis parameters namely acid concentration, reaction time, and reaction temperature were modeled and optimized using Box-Behnken design (BBD) of response surface methodology (RSM). Temperature was found to be the most important parameter affecting the photoluminescence quantum efficiency of the carbon dots. The as-prepared carbon dots in aqueous solution showed a blue-green emission under UV light with a relatively high quantum yield of 33%, agreeing well with the value predicted using the RSM. The carbon dots exhibited distinct selectivity toward silver ion (Ag<SUP>+</SUP>) with a limit of detection of 2.8 μM. The Ag<SUP>+</SUP> contents in real water samples were accurately determined with acceptable repeatability. Based on these results, the RSM is a useful tool for investigating the effects of reaction parameters on the quantum efficiency of carbon dots. Furthermore, pyrolysis was proved to be an effective means for producing label-free, highly photoluminescent carbon dots from polyurethane, enabling new, effective recycling strategy and various potential sensing applications.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Ethanol Extract of Ulmus pumila Ameliorates Heat Stress through the Induction of Heat Shock Proteins Expression in RAW264.7 Macrophage Cells

        Heat stress is a significant burden to animal production in most areas of the world. Improving our knowledge of physiological and metabolic mechanisms of acclimation may contribute to the development of procedures that may help to maintain health and production efficiency under hot temperature. The effect of Ulmus pumila (UP) extract in inducing Heat Shock Proteins (HSPs) expression in heat-stressed RAW264.7 macrophage cells was investigated. Cell viability assay showed a dose dependent increase in cells after treatment with UP for 24 hours. RT-PCR and western blot analysis showed that increasing concentrations of UP induce the expression of Heat Shock Factor 1 (HSF1) and dose dependently upregulated the expression of Heat shock protein 70 (Hsp70) and Hsp90. LPS-induced nitric oxide was dose-dependently reduced while phagocytic activity greatly recovered with UP treatment. These data demonstrated that UP can be a potential candidate in the development of cytoprotective agent against heat stress.

      • Philippine COPD Pathway Project

        Chronic Obstructive Pulmonary Disease (COPD), as a chronic airway disease, is associated with a significant burden and has been a major public health problem in the Philippines. It remains to be one of the top ten causes of mortality and morbidity. The Philippine College of Chest Physicians (PCCP) Council on COPD and Pulmonary Rehabilitation last published the guidelines for COPD management in the Philippines last 2009 which was revised in 2014. With the passage of Universal Health Care (UHC) Act in the Philippines last February 2019, consensus statements on the diagnosis and management of COPD were drafted which aims to provide a simple guide to all health care workers (in the different levels of care- primary, secondary and tertiary) who manage possible COPD patients within the universal health care framework of the Philippines and the presence of COVID-19 infection. This guideline was drafted by the members of Council of COPD and Pulmonary Rehabilitation of PCCP. The Philippine Academy of Family Physicians (PAFP) also participated as active members in the working committees in the process. The process of the update formulation included working committees on diagnosis, pharmacologic and nonpharmacologic treatment reviewing international guidelines and relevant literature on the sections and formulating the algorithms. The Global Initiative on Obstructive Lung Diseases (GOLD) served as the major reference of the pathway. This updated guideline/ consensus statements covered a wide range of sections including the diagnosis of COPD, management of stable COPD, management of COPD exacerbations and monitoring and non-pharmacologic management of COPD in the different levels of care.

        Case Presentation: Carbimazole is an antithyroid drug with a rare potentially fatal complication of agranulocytosis. Presented is a case of a 42-year-old male with Graves` disease and was admitted for fever. He initially presented with loose bowel movement, tremors, palpitation, and tachycardia and unexplained weight loss. He was maintained on Methimazole, but was later shifted to Carbimazole, when he developed rashes while taking the former drug. However, within a week of Carbimazole treatment, he developed fever, sore throat and agranulocytosis. Working diagnosis at that time was: Febrile neutropenia with acute tonsillopharyngitis, in Sepsis: Agranulocytosis secondary to Carbimazole: Graves` disease, in Thyroid Storm. Discussion: Carbimazole - induced agranulocytosis usually spontaneously resolves within one to two weeks of stopping the drug. The use of haemopoietic growth factors to stimulate the proliferation and differentiation of progenitor cells accelerates neutrophil recovery, as shown in this case. Conclusion: Agranulocytosis from carbimazole is an uncommon, life-threatening complication. With the potential to reduce morbidity and mortality, the use of haemopoietic growth factors certainly seems a justifiable option, instead of awaiting spontaneous recovery. Diagnosis and management of this case conveyed a dilemma that was resolved afterwards. Keywords: carbimazole, agranulocytosis, thyroid storm, haemopoietic growth factor, G-CSF

        Low lysine stimulates adipogenesis through ZFP423 upregulation in bovine stromal vascular cells

        Adipogenesis is a complex process comprising commitment and a differentiation stages. Through research, many different transcriptional factors were found to mediate preadipocyte commitment and differentiation. Lysine has a potential of regulating the commitment and differentiation of preadipocytes. In the present study, intramuscular stromal vascular cells (SVC) isolated from Hanwoo beef cattle were used to elucidate the effects of low lysine level on adipogenesis. SVC were isolated and incubated with various concentrations of lysine (0, 37.5, 75, 150 and 300 μg/mL). No significant difference were observed in the proliferation of SVC after 24 and 48 h of incubation with different concentration of lysine. On preadipocyte determination, reducing the level of lysine significantly increased the expression of preadipocyte commitment gene Zinc finger protein 423 and Preadipocyte factor-1. Upon differentiation, Oil Red O staining revealed that lipid accumulation and triglyceride content significantly increased with the decreasing lysine levels in the media. Expression levels of peroxisome proliferator-activated receptor-γ, CCAAT enhancer binding protein-α, sterol regulatory element binding protein-1c, Fatty Acid Binding Protein 4 and stearoyl CoA desaturase were upregulated by the decreased level of lysine. These data suggest the potential mechanism of action for the improved preadipocyte commitment and adipocyte differentiation in bovine intramuscular SVC upon treatment with low levels of lysine. These findings may be valuable in developing feed rations that promote deposition of intramuscular fat in beef cattle through lysine level modification.
