        System identification of the suspension tower of Runyang Bridge based on ambient vibration tests

        A series of field vibration tests are conducted on the Runyang Suspension Bridge during both the construction and operational stages. The purpose of this study is devoted to the analysis of the dynamic characteristics of the suspension tower. After the tower was erected, an array of accelerometers was deployed to study the evolution of its modal parameters during the construction process. Dynamic tests were first performed under the freestanding tower condition and then under the tower-cable condition after the superstructure was installed. Based on the identified modal parameters, the effect of the pile-soil-structure interaction on dynamic characteristics of the suspension tower is investigated. Moreover, the stiffness of the pile foundation is successfully identified using a probabilistic finite model updating method. Furthermore, challenges of identifying the dynamic properties of the tower from the coupled responses of the tower-cable system are discussed in detail. It\'s found that compared with the identified results from the freestanding tower, the longitudinal and torsional natural frequencies of the tower in the tower-cable system have changed significantly, while the lateral mode frequencies change slightly. The identified modal results from measurements by the structural health monitoring system further confirmed that the vibrations of the bridge subsystems (i.e., the tower, the suspended deck and the main cable) are strongly coupled with one another.

        MicroRNA-217 Functions as a Tumour Suppressor Gene and Correlates with Cell Resistance to Cisplatin in Lung Cancer

        MiR-217 can function as an oncogene or a tumour suppressor gene depending on cell type. However, the function of miR-217 in lung cancer remains unclear to date. This study aims to evaluate the function of miR-217 in lung cancer and investigate its effect on the sensitivity of lung cancer cells to cisplatin. The expression of miR-217 was detected in 100 patients by real-time PCR. The effects of miR-217 overexpression on the proliferation, apoptosis, migration and invasion of SPC-A-1 and A549 cells were investigated. The target gene of miR-217 was predicted by Targetscan online software, screened by dual luciferase reporter gene assay and demonstrated by Western blot. Finally, the effects of miR-217 up-regulation on the sensitivity of A549 cells to cisplatin were determined. The expression of miR-217 was significantly lower in lung cancer tissues than in noncancerous tissues (p < 0.001). The overexpression of miR-217 significantly inhibited the proliferation, migration and invasion as well as promoted the apoptosis of lung cancer cells by targeting KRAS. The up-regulation of miR-217 enhanced the sensitivity of SPC-A-1 and A549 cells to cisplatin. In conclusion, miR-217 suppresses tumour development in lung cancer by targeting KRAS and enhances cell sensitivity to cisplatin. Our results encourage researchers to use cisplatin in combination with miR-217 to treat lung cancer. This regime might lead to low-dose cisplatin application and cisplatin side-effect reduction.

        According to the retweeting mechanism based on user relations, we introduce the contacted state of an agent, and build a model of information propagation process in social networks. In the model, agents have two absorbing states, that is, the infected and refractory state. Experimental prove that small spreading rete can make information diffuse widely, and the degree based density of infected agents increases with the degree monotonously. The influence between information occurs only when different information originates in the same local neighborhood.

        IDENTIFICATION OF THERMODYNAMIC PARAMETERS OF ARCTIC SEA ICE AND NUMERICAL SIMULATION

        This paper studies the multi-domain coupled system of one dimensional Arctic temperature field and establishes identification model about the thermodynamic parameters of sea ice (heat storage capacity, density and conductivity) by the so-called output least-square estimate according to the temperature data acquired by a monitor buoy installed in the Arctic ocean. By the optimal control theory, the existence and dependability of weak solution and the identifiability of identification model have been given. Moreover, necessary optimality condition is proposed. Furthermore, the optimal algorithm for the identification model is constructed. By using the optimal thermodynamic parameters of Arctic sea ice, the numerical simulation is implemented, and the numerical results of temperature distribution of Arctic sea ice are demonstrated.

        Interface contact pressure-based virtual gradient material model for the dynamic analysis of the bolted joint in machine tools

        Bolted joints have significant influence on the dynamic characteristic of the machine tool assembly. This study proposes a new modeling method using virtual gradient material. On the basis of Hertz contact theory and fractal geometry theory, the relationship between contact stiffness and normal contact load is deduced. The contact pressure distribution at the interface is obtained through finite element contact analysis. An orthogonal material model is introduced to describe the properties of virtual material. The virtual material is divided into several layers, and the elastic modulus and shear modulus in each sub-layer are derived based on Hooke’s Law. Both experimental and theoretical results for a bolted lap structure are obtained. The experimental results are compared with theoretical ones in terms of qualitative comparison of similar mode shapes and quantitative comparison of corresponding natural frequencies. The comparison shows that the theoretical first three-order vibration mode shapes are in good line with the experimental ones. The relative errors of corresponding natural frequencies between the virtual gradient material model and the experiment are less than 9%, which are smaller than those between the traditional virtual uniform material model and the experiment. The proposed virtual gradient material method would be useful to accurately model the bolted joint in CNC machine tools.

        Performances of Prognostic Models in Stratifying Patients with Advanced Gastric Cancer Receiving First-line Chemotherapy: a Validation Study in a Chinese Cohort

        Purpose: While several prognostic models for the stratification of death risk have been developed for patients with advanced gastric cancer receiving first-line chemotherapy, they have seldom been tested in the Chinese population. This study investigated the performance of these models and identified the optimal tools for Chinese patients. Materials and Methods: Patients diagnosed with metastatic or recurrent gastric adenocarcinoma who received first-line chemotherapy were eligible for inclusion in the validation cohort. Their clinical data and survival outcomes were retrieved and documented. Time-dependent receiver operating characteristic (ROC) and calibration curves were used to evaluate the predictive ability of the models. Kaplan-Meier curves were plotted for patients in different risk groups divided by 7 published stratification tools. Log-rank tests with pairwise comparisons were used to compare survival differences. Results: The analysis included a total of 346 patients with metastatic or recurrent disease. The median overall survival time was 11.9 months. The patients were different into different risk groups according to the prognostic stratification models, which showed variability in distinguishing mortality risk in these patients. The model proposed by Kim et al. showed relative higher predicting abilities compared to the other models, with the highest χ<sup>2</sup> (25.8) value in log-rank tests across subgroups, and areas under the curve values at 6, 12, and 24 months of 0.65 (95% confidence interval [CI]: 0.59-0.72), 0.60 (0.54-0.65), and 0.63 (0.56-0.69), respectively. Conclusions: Among existing prognostic tools, the models constructed by Kim et al., which incorporated performance status score, neutrophil-to-lymphocyte ratio, alkaline phosphatase, albumin, and tumor differentiation, were more effective in stratifying Chinese patients with gastric cancer receiving first-line chemotherapy.

        Determination of Survival of Gastric Cancer Patients With Distant Lymph Node Metastasis Using Prealbumin Level and Prothrombin Time: Contour Plots Based on Random Survival Forest Algorithm on High-Dimensionality Clinical and Laboratory Datasets

        Purpose: This study aimed to identify prognostic factors for patients with distant lymph node-involved gastric cancer (GC) using a machine learning algorithm, a method that offers considerable advantages and new prospects for high-dimensional biomedical data exploration. Materials and Methods: This study employed 79 features of clinical pathology, laboratory tests, and therapeutic details from 289 GC patients whose distant lymphadenopathy was presented as the first episode of recurrence or metastasis. Outcomes were measured as any-cause death events and survival months after distant lymph node metastasis. A prediction model was built based on possible outcome predictors using a random survival forest algorithm and confirmed by 5×5 nested cross-validation. The effects of single variables were interpreted using partial dependence plots. A contour plot was used to visually represent survival prediction based on 2 predictive features. Results: The median survival time of patients with GC with distant nodal metastasis was 9.2 months. The optimal model incorporated the prealbumin level and the prothrombin time (PT), and yielded a prediction error of 0.353. The inclusion of other variables resulted in poorer model performance. Patients with higher serum prealbumin levels or shorter PTs had a significantly better prognosis. The predicted one-year survival rate was stratified and illustrated as a contour plot based on the combined effect the prealbumin level and the PT. Conclusions: Machine learning is useful for identifying the important determinants of cancer survival using high-dimensional datasets. The prealbumin level and the PT on distant lymph node metastasis are the 2 most crucial factors in predicting the subsequent survival time of advanced GC.

        Therapeutic Benefits of Mesenchymal Stromal Cells in a Rat Model of Hemoglobin-Induced Hypertensive Intracerebral Hemorrhage

        Previous studies have shown that bone marrow mesenchymal stromal cell (MSC) transplantation significantly improves the recovery of neurological function in a rat model of intracere-bral hemorrhage. Potential repair mechanisms involve anti-inflammation, anti-apoptosis and angiogenesis. However, few studies have focused on the effects of MSCs on inducible nitric oxide synthase (iNOS) expression and subsequent per-oxynitrite formation after hypertensive intracerebral hemorrhage (HICH). In this study, MSCs were transplanted intracerebrally into rats 6 hours after HICH. The modified neurological severity score and the modified limb placing test were used to measure behavioral outcomes. Blood–brain barrier disruption and neuronal loss were measured by zonula occludens-1 (ZO-1) and neuronal nucleus (NeuN) expression, respectively. Concomitant edema formation was evaluated by H&E staining and brain water content. The effect of MSCs treatment on neuroinflammation was analyzed by immunohistochemical analysis or polymerase chain reaction of CD68, Iba1, iNOS expression and subsequent peroxynitrite formation, and by an enzyme-linked immunosorbent assay of pro-inflammatory factors (IL-1 and TNF-). The MSCs-treated HICH group showed better performance on behavioral scores and lower brain water content compared to controls. Moreover, the MSC injection increased NeuN and ZO-1 expression measured by immunochemistry/immunofluorescence. Furthermore, MSCs reduced not only levels of CD68, Iba1 and pro-inflammatory factors, but it also inhibited iNOS expression and peroxynitrite formation in perihematomal regions. The results suggest that intracerebral administration of MSCs accelerates neurological function recovery in HICH rats. This may result from the ability of MSCs to suppress inflammation, at least in part, by inhibiting iNOS expression and subsequent peroxynitrite formation.
