Differential expression of heat shock protein 90, 70, 60 in chicken muscles postmortem and its relationship with meat quality

        Zhang, Muhan,Wang, Daoying,Geng, Zhiming,Sun, Chong,Bian, Huan,Xu, Weimin,Zhu, Yongzhi,Li, Pengpeng Asian Australasian Association of Animal Productio 2017 Animal Bioscience Vol.30 No.1

Objective: The aim of this study was to investigate the expression of heat shock protein (HSP) 90, 70, and 60 in chicken muscles and their possible relationship with quality traits of meat. Methods: The breast muscles from one hundred broiler chickens were analyzed for drip loss and other quality parameters, and the levels of heat shock protein (HSP) 90, 70, and 60 were determined by immunoblots. Results: Based on the data, chicken breast muscles were segregated into low (drip loss${\leq}5%$), intermediate (5%<drip loss<9.5%) and high (drip loss${\geq}9.5$) drip loss groups. The expression of HSP90 and HSP60 were significantly lower in the high drip loss group compared to that in the low and intermediate drip loss group (p<0.05), while HSP70 was equivalent in abundance in all groups (p>0.05). Conclusion: Results of this study suggests that higher levels of HSP90 and HSP60 may be advantageous for maintenance of cell function and reduction of water loss, and they could act as potential indicator for better water holding capacity of meat.

Evaluation of antioxidant property of heat shock protein 90 from duck muscle

        Zhang Muhan,Wang Daoying,Xu Xinglian,Xu Weimin 아세아·태평양축산학회 2021 Animal Bioscience Vol.34 No.4

Objective: The objectives of this study were to investigate the direct antioxidative effect of 90 Kda heat shock protein (Hsp90) obtained from duck muscle. Methods: The interaction of Hsp90 with phospholipids and oxidized phospholipids was studied with surface plasmon resonance (SPR), and their further oxidation in the presence of Hsp90 was evaluated with thiobarbituric acid reactive substances (TBARS) assay. The scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azinobis (3-ethylbenzthiazoline-6-sulfonic acid (ABTS) was measured, and the electron paramagnetic resonance (EPR) spectroscopy in combination with 5-tert-Butoxycarbonyl-5-methyl-1- pyrroline-N-oxide and 2-phenyl-4,4,5,5,-tetramethylimidazoline-1-oxyl-3-oxide (PTIO) was utilized to determine the abilities of Hsp90 in scavenging hydroxyl and PTIO radicals. Results: SPR showed Hsp90 could bind with both phospholipids and oxidized phospholipids, and prevent their further oxidation by the TBARS assay. The DPPH and ABTS scavenging activity increased with Hsp90 concentration, and could reach 27% and 20% respectively at the protein concentration of 50 μM. The EPR spectra demonstrated Hsp90 could directly scavenge ·OH and PTIO· radicals. Conclusion: This suggests that Hsp90, a natural antioxidant in meat, may play an important role in cellular defense against oxidative stress, and may have potential use in meat products. Effect of myoglobin, hemin, and ferric iron on quality of chicken breast meat

Zhang, Muhan,Yan, Weili,Wang, Daoying,Xu, Weimin Asian Australasian Association of Animal Productio 2021 Animal Bioscience Vol.34 No.8

Objective: The objective was to evaluate the impact of different forms of iron including myoglobin, hemin, and ferric chloride on the quality of chicken breast meat. Methods: Chicken breast muscles were subjected to 1, 2, 3 mg/mL of FeCl3, myoglobin and hemin treatment respectively, and the production of reactive oxygen species (ROS) and malondialdehyde, meat color, tenderness, water holding capacity and morphology of meat was evaluated. Results: Hemin was found to produce more ROS and induce greater extent of lipid oxidation than myoglobin and ferric chloride. However, it showed that hemin could significantly increase the redness and decrease the lightness of the muscle. Hemin was also shown to be prominent in improving water holding capacity of meat, maintaining a relatively higher level of the immobilized water from low-field nuclear magnetic resonance measurements. Morphology observation by hematoxylin-eosin staining further confirmed the results that hemin preserved the integrity of the muscle. Conclusion: The results indicated that hemin may have economic benefit for the industry based on its advantage in improving water holding capacity and quality of meat.

Out-of-Band Measurement of LED-based Solar Blind UV Filters

        Muhan Cui,Xue Chen,Feng Yan,Mingchao Zhang,Huaijiang Yang,Yue Zhou 한국광학회 2014 Current Optics and Photonics Vol.18 No.3

Due to the difficulty in measuring very low out-of-band cutoff depths of solar blind UV filters, we propose a cutoff depth adjustable measurement system (CDAM) to test deep cutoff filters with a large dynamic range. The CDAM utilizing the substitution method is elaborately composed of several parts, including narrow-band LED light sources, standard reflective neutral attenuators with known attenuation coefficients, and a photomultiplier (PMT). This paper also presents an attenuator combination method ensuring that the PMT works within its linear response range. In addition, numerical simulation testifies to the method, and experiment shows that the CDAM system can achieve an extension of dynamic range from 0—6 OD to 0—10 OD, which is sufficient for the measurement of out-of-band cutoff depths of solar blind UV filters. Above all, the CDAM system, being easily implemented, of wide dynamic range, and highly precise, could be widely used in the measurement of filter cutoff depth.

The influence of ultrasound and adenosine 5’-monophosphate marination on tenderness and structure of myofibrillar proteins of beef

        Ye Zou,Heng Yang,Muhan Zhang,Xinxiao Zhang,Weimin Xu,Daoying Wang 아세아·태평양축산학회 2019 Animal Bioscience Vol.32 No.10

Objective: The aim was to investigate the influence of ultrasound and adenosine 5’-monophosphate (AMP) marination (UAMP) on tenderness and structure of myofibrillar proteins of beef. Methods: Five groups, the untreated meat (Control), deionized water marination (DW), ultrasound followed by DW (UDW), AMP marination (AMP), and ultrasound followed by AMP (UAMP) were studied. Myofibrillar fragmentation, cooking loss, shear force, thermograms, histological observation of meats and myofibrillar proteins properties were investigated in these different treatments. Results: The results showed that UAMP significantly increased myofibrillar fragmentation index from 152 (Control), 231 (AMP), and 307 (UDW) to 355 (p<0.05), respectively. The lowest cooking loss, shear force and peak denaturation temperature were observed in UAMP. In histological observation, UDW and UAMP had more fragmented muscular bundles than the others. Furthermore, a drastic increase in α-helix and decrease in β-sheet of myofibrillar proteins was observed in UAMP, implying the disaggregation of protein samples. The synchronous fluorescence spectra of myofibrillar proteins in UAMP suggested the combination of ultrasound and AMP could accelerate the unfolding molecular structure and destroying hydrophobic interactions. The results of circular dichroism and synchronous fluorescence spectra for myofibrillar proteins coincided with the microstructures of beef. Conclusion: The results indicate that ultrasound combined with AMP improved meat tenderness not only by disruption in muscle integrity, increasing water retention, but also altering their spatial structure of myofibrillar proteins.

      • Switching terahertz waves with gate-controlled active graphene metamaterials

        Lee, Seung Hoon,Choi, Muhan,Kim, Teun-Teun,Lee, Seungwoo,Liu, Ming,Yin, Xiaobo,Choi, Hong Kyw,Lee, Seung S.,Choi, Choon-Gi,Choi, Sung-Yool,Zhang, Xiang,Min, Bumki Nature Publishing Group 2012 Nature materials Vol.11 No.11

The extraordinary electronic properties of graphene provided the main thrusts for the rapid advance of graphene electronics. In photonics, the gate-controllable electronic properties of graphene provide a route to efficiently manipulate the interaction of photons with graphene, which has recently sparked keen interest in graphene plasmonics. However, the electro-optic tuning capability of unpatterned graphene alone is still not strong enough for practical optoelectronic applications owing to its non-resonant Drude-like behaviour. Here, we demonstrate that substantial gate-induced persistent switching and linear modulation of terahertz waves can be achieved in a two-dimensional metamaterial, into which an atomically thin, gated two-dimensional graphene layer is integrated. The gate-controllable light??matter interaction in the graphene layer can be greatly enhanced by the strong resonances of the metamaterial. Although the thickness of the embedded single-layer graphene is more than six orders of magnitude smaller than the wavelength (<貫/1,000,000), the one-atom-thick layer, in conjunction with the metamaterial, can modulate both the amplitude of the transmitted wave by up to 47% and its phase by 32.2째 at room temperature. More interestingly, the gate-controlled active graphene metamaterials show hysteretic behaviour in the transmission of terahertz waves, which is indicative of persistent photonic memory effects.

An Automatic Corona-discharge Detection System for Railways Based on Solar-blind Ultraviolet Detection

        Jiaqi Li,Yue Zhou,Xiangyu Yi,Mingchao Zhang,Xue Chen,Muhan Cui,Feng Yan 한국광학회 2017 Current Optics and Photonics Vol.1 No.3

Corona discharge is always a sign of failure processes of high-voltage electrical apparatus, includingthose utilized in electric railway systems. Solar-blind ultraviolet (UV) cameras are effective tools for coronainspection. In this work, we present an automatic railway corona-discharge detection system based on solarblindultraviolet detection. The UV camera, mounted on top of a train, inspects the electrical apparatus, including transmission lines and insulators, along the railway during fast cruising of the train. An algorithmbased on the Hough transform is proposed for distinguishing the emitting objects (corona discharge) fromthe noise. The detection system can report the suspected corona discharge in real time during fast cruises. An experiment was carried out during a routine inspection of railway apparatus in Xinjiang Province, China. Several corona-discharge points were found along the railway. The false-alarm rate was controlled to lessthan one time per hour during this inspection.
