        Methods for Determination of Amino Acids Bioavailability in Pigs - Review -

        Methods developed for measuring digestibility and availability of amino acids in feedstuffs used in pig nutrition are reviewed. Digestibility is a proportion of an amino acid in a feed that is absorbed from the digestive tract and should be determined from the difference between the amount of amino acid consumed and passing the distal ileum. Techniques for ileal digesta sampling including various types of cannulas: a re-entrant, T-piece, IPV, IPVC and ileaorectal anastomosis are described and comparisons amongst these methods are presented. Other methodologies like mobile bag technique, in vitro assays and mathematical prediction method are also described. Significance and methodologies for measurement of endogenous nitrogen and amino acids losses at the distal ileum and their effect on the apparent and true nitrogen and amino acid digestibilities in feeds are discussed. Factors influencing the apparent and true amino acid digestibilities such as dry matter intake, protein, fibre and antinutritive compounds content in the diet are discussed. Amino acid bioavailability -the proportion of the total amino acid digested and absorbed in a form utilized in metabolism - measured by the growth assay may differ from its ileal digestibility. Chemical methods for determination of available lysine content in heat treated feeds are evaluated.

        Investigation of -308G>A and -1031T>C Polymorphisms in the TNFA Promoter Region in Polish Peptic Ulcer Patients

        Background/Aims: Tumor necrosis factor α (TNF-α) encoded by TNFA is a key mediator in inflammation, a precursor condition for peptic ulceration. Promoter polymorphisms of TNFA that influence its transcriptional activity and TNF-α production are known. TNFA-308G>A (rs1800629) and TNFA- 1031T>C (rs1799964), which are responsible for increased TNFA transcription, could influence the risk of peptic ulceration. This study aimed to investigate these polymorphisms and to evaluate their association with peptic ulcer disease and Helicobacter pylori infection in the Polish population. Methods: Gastric mucosa specimens obtained from 177 Polish peptic ulcer patients were used to conduct rapid urease tests and to assess the investigated polymorphisms by polymerase chain reaction-restriction fragment length polymorphism. Genotyping data were compared with the results obtained from healthy individuals of Polish origin. Results: There were no significant differences in genotype and allele frequency of the investigated polymorphisms between peptic ulcer patients and healthy individuals. No associations between the frequencies of particular genotypes and alleles for both single-nucleotide polymorphisms (SNPs) and the presence of H. pylori infection in peptic ulcer patients and in subgroups of men and women with peptic ulcer disease were found. Conclusions: The investigated SNPs are not risk factors for either peptic ulcer or H. pylori infection development in the Polish population. The results require verification in a larger cohort.         Bioavailability of Phosphorus in Feeds of Plant Origin for Pigs - Review -

        Phosphorus has been known as an essential component of animal body. However, the requirement has not been determined precisely because of the variable bioavailabilities of feedstuffs from plant origin. The bioavailability of P in various feedstuffs of plant origin varies from 10 to 60%. Digestibility and availability of the P differed considerably depending on the feed. The lowest values were found for maize (under 20%), the highest for wheat and triticale (over 50%). This is due to the proportion of phytate and the presence of intrinsic phytase. And the digestive tract of monogastric animals does not contain sufficient amounts of phytase, an enzyme that hydrolyses the unavailable phytate complexes to available, inorganic orthophosphates. Microbial phytase supplementation improves the P availability, and both intrinsic plant and microbial phytase improves the availability of P in feedstuffs of plant origin. In a mixture of feeds with low and high activity of intrinsic phytase and/or supplemented by commercial phytase, the P availability is additive. However, in the light of current results it seems that exceeding the P availability equal to 60-70% is unrealizable even at large microbial phytase doses.

        Apparent Digestibility of Phosphorus in Experimental Feeds and the Effect of Commercial Phytase

        The digestibility (apparent) of P and other nutrients from the RSM, SBM and 2 batches of maize, wheat and barley was investigated in two experiments with 24 castrated male growing pigs. The effect of supplemental microbial phytase (1,000 U/kg) was also evaluated. The diets contained 25% RSM (Exp. 1) or 40% SBM (Exp. 2) and had no inorganic P. In each period of digestive trial, after 9 days of adaptation, faeces were collected for 5 days. The digestibility of P contained in the RSM and SBM was calculated by difference method. The P digestibility in maize, wheat, barley was on average 20, 34 and 36%, respectively. The digestibility of P in the RSM and SBM estimated from maize-based diets were 19 and 24 %, respectively. Kind of cereal grain had significant (p < 0.05) influence on the digestibility of P which was lower in the diets based on maize than wheat or barley. The digestibility of P significantly increased with the supplemental microbial phytase (on an average of 17%). Moreover, inclusion of enzyme into the diets positively affected digestibility of other nutrients, namely the protein and organic matter.

        Growth Performance, Body Composition and Protein and Energy Utilization of Pigs Fed Ad Libitum Diets Formulated According to Digestible Amino Acid Content

        Four groups of six growing gilts each were fed ad libitum diets composed of cereals and soyabean (SBM) or rapeseed (RSM) meal and containing two levels of crude protein: high - 18.0% (RSM-H) and 16.9% (SBM-H) or low - 15.6% (RSM-L) and 15.1% (SBM-L). The diets were balanced by supplementation with crystalline amino acids and contained apparent ileal digestible lysine, methionine, threonine and tryptophan in proportions (1.00:0.32:0.57:0.18) according to CVB (1995). Voluntary feed intake, weight gain and slaughter and chemical body composition of animals were assessed. Protein and energy balances from 25 to 70 kg body weight were calculated by the comparative slaughter method. Protein source had a significant effect on voluntary feed intake; it was 0.12 kg/d lower in pigs fed the SBM than RSM-diets. Pigs fed on the SBM-L diet consumed the least amount of feed (2.17 kg). Daily gain (average, $900{\pm}12.59g$) and feed conversion ratio ($2.54{\pm}0.04kg/kg$) were not statistically affected by source (SMB and RSM) and protein level (high and low). In empty body similar amounts (g/kg) of protein (163 g), water (635 g) and ash (28 g) were found. However, pigs fed the RSM-L diet were fatter than those fed the SBM-L diet (188 vs. 161 g/kg). No statistical differences were observed in daily protein deposition, which on average amounted to $142{\pm}11g$, or carcass characteristics. An improvement of crude protein utilization by 6.3 percentage units was found by decreasing the protein concentration in the diets. Heat production in the body was not significantly affected by the treatments.
