        Interaction between the p75 neurotrophin receptor and a novel adaptor protein

        The neurotrophin plays an important role in the development, differentiation and survival of the nervous system in vertebrates. It exerts its cellular effects through two different receptors, the Trk receptor tyrosine kinase neurotrophin receptor and the p75 neurotrophin receptor, a member of the tumor necrosis factor receptor superfamily. Trk and p75 neurotrophin receptors utilize specific target proteins to transmit signals into the cell. An ankyrin-rich membrane spanning protein (ARMS) was identified as a new p75 interacting protein and serves as a novel downstream target of p75 neurotrophin receptor. We sought to delineate the interaction between p75 and ARMS by deletion constructs of p75 and green fluorescent protein (GFP)-tagged ARMS. We examined the interaction between these two proteins after overexpressing them in HEK-293 cells. Using both Western blot analysis and immunocytochemistry followed by confocal laser scanning microscopy, we found out that the intracellular domain of the p75 neurotrophin receptor was important for the interaction with ARMS. The results from this study suggest that ARMS may play an important role for mediating the signals from p75 neurotrophin receptor into the cell.

        Boronate-functionalized hydrogel as a novel biosensing interface for the glycated hemoglobin A1c (HbA<sub>1c</sub>) based on the competitive binding with signaling glycoprotein

        <P><B>Abstract</B></P> <P>According to recent increases in public healthcare costs associated with diabetes mellitus, the development of new glycemic monitoring techniques based on the biosensing of glycated hemoglobin A1c (HbA<SUB>1c</SUB>), a promising long-term glycemic biomarker, has become a major challenge. In the development of HbA<SUB>1c</SUB> biosensors for point-of-care applications, the selection of an effective biorecognition layer that provides a high reaction yield and specificity toward HbA<SUB>1c</SUB> is regarded as the most significant issue. To address this, we developed a novel HbA<SUB>1c</SUB> biosensing interfacial material by the integration of boronate hydrogel with glass fiber membrane. In the present study, a new boronate-functionalized hydrogel was designed and spatio-selectively photopolymerized on a hydrophilic glass fiber membrane by using N-hydroxyethyl acrylamide, 3-(acrylamido)phenylboronic acid, and bis(<I>N</I>,<I>N</I>′-methylene-bis-acrylamide). Using this approach, the boronic acid group, which specifically recognizes the cis-diol residue of glucose on the HbA<SUB>1c</SUB> molecule, can be three-dimensionally coated on the surface of the glass fiber network with a high density. Because this network structure of boronate hydrogel-grafted fibers enables capillary-driven fluid control, facile HbA<SUB>1c</SUB> biosensing in a lateral flow assay concept could be accomplished. On the proposed HbA<SUB>1c</SUB> biosensing interface, various concentrations of HbA<SUB>1c</SUB> (5–15%) in blood-originated samples were sensitively measured by a colorimetric assay using horseradish peroxidase, a glycoenzyme can generate chromogenic signal after the competitive binding against HbA<SUB>1c</SUB> to the boronic acid residues. Based on the demonstrated advantages of boronate hydrogel-modified membrane including high analytical performance, easy operation, and cost-effectiveness, we expect that the proposed biorecognition interfacial material can be applied not only to point-of-care HbA<SUB>1c</SUB> biosensors, but also to the quantitative analysis of other glycoprotein biomarkers.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Boronic acid (BA) hydrogel was synthesized for HbA<SUB>1c</SUB> biosensing. </LI> <LI> BA hydrogel was patterned on the glass fiber membrane via photo-crosslinking. </LI> <LI> HbA<SUB>1c</SUB> and peroxidase were competitively bound to BA hydrogel-modified membrane. </LI> <LI> HbA<SUB>1c</SUB> could be efficiently quantified by peroxidase-based chromogenic reaction. </LI> <LI> HbA<SUB>1c</SUB> in the whole blood sample was determined within a range of 4.6–15.2%. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Effect of Polysaccharides from Acanthopanax senticosus on Intestinal Mucosal Barrier of Escherichia coli Lipopolysaccharide Challenged Mice

        To investigate the role of polysaccharide from Acanthopanax senticosus (ASPS) in preventing lipopolysaccharide (LPS)-induced intestinal injury, 18 mice (at 5 wk of age) were assigned to three groups with 6 replicates of one mouse each. Mice were administrated by oral gavage with or without ASPS (300 mg/kg body weight) for 14 days and were injected with saline or LPS at 15 days. Intestinal samples were collected at 4 h post-challenge. The results showed that ASPS ameliorated LPS-induced deterioration of digestive ability of LPS-challenged mice, indicated by an increase in intestinal lactase activity (45%, p<0.05), and the intestinal morphology, as proved by improved villus height (20.84%, p<0.05) and villus height:crypt depth ratio (42%, p<0.05), and lower crypt depth in jejunum (15.55%, p<0.05), as well as enhanced intestinal tight junction proteins expression involving occludin-1 (71.43%, p<0.05). ASPS also prevented intestinal inflammation response, supported by decrease in intestinal inflammatory mediators including tumor necrosis factor ${\alpha}$ (22.28%, p<0.05) and heat shock protein (HSP70) (77.42%, p<0.05). In addition, intestinal mucus layers were also improved by ASPS, as indicated by the increase in number of goblet cells (24.89%, p<0.05) and intestinal trefoil peptide (17.75%, p<0.05). Finally, ASPS facilitated mRNA expression of epidermal growth factor (100%, p<0.05) and its receptor (200%, p<0.05) gene. These results indicate that ASPS can prevent intestinal mucosal barrier injury under inflammatory conditions, which may be associated with up-regulating gene mRNA expression of epidermal growth factor and its receptor.

        Poverty, Food Insecurity and Helplessness among the Poor in Bangladesh during the COVID-19 Pandemic - A Qualitative Study -

        본 연구는 코로나19 팬데믹으로 인해 빈곤층에서 증가한 경제적 위기와 영양적 위험, 그 대응전략을 탐색하기 위하여, 방글라데시 랑푸르도 카하롤군 5세 미만 자녀를 둔 여성 45명을 대상으로 심층면 접조사를 실시하였다. 대부분의 응답자는 코로나19 대유행 이후 실직 및 소득 감소를 경험하였고, 주식 및 영양가 있는 식품을 구매할 수 없었으며, 특히, 아동과 임산부는 영양적으로 부족하고 불균 형한 음식을 섭취하고 있었다. 이러한 위기에 대응하기 위해 각 가구들은 가축을 팔거나, 대출을 받고, 저축을 사용하는 등 다양한 임시방편적인 전략을 취했다. 특히, 대출을 받은 경우 대출금을 상환할 수 없어 다시금 심각한 경제적⋅정신적 스트레스의 악순환에 빠졌다. 응답자들은 이러한 위기를 미래에도 극복할 수 없을 것이라는 만연한 무력감과 좌절감을 표현했다. 조사 당시 지역정부 와 NGO의 지원이 부족했고, 지역사회 주민들 간 상호 부조 역시 경제적 어려움으로 인해 거의 발견 되지 않았다. 본 연구는 지속 가능한 대응전략으로서 자급자족을 촉진하는 소득증대 지원 등의 확대 와 지역사회 회복력 강화를 위한 후속 연구를 제안하였다. Globally, the COVID-19 pandemic heightened the risk of economic and nutritional instability, particularly among the poor population. We conducted in-depth interviews with 45 women with young children in Kaharole Upazila, Rangpur Division, in Bangladesh. Most respondents reported reduced income or job loss since the pandemic and could not afford to purchase staples and nutrient-rich foods. Young children and pregnant women were at risk of having inadequate diets. The households used stress coping mechanisms such as selling livestock, taking loans, and spending savings several times. The coping mechanisms again put the respondents into a vicious economic and severe mental stress cycle as they had no way to repay the loan. Respondents expressed helplessness and frustration as a result of the economic downturn. There was a lack of relief assistance from local governments or civil societies. Our findings showed poor Bangladesh women faced financial hardship, limited food choices, and provisional coping strategies. It is advised that disaster relief efforts be directed toward helping the affected population maintain their standard of living and building local community resilience.

        학생 능동수업의 교육적 효과에 대한 초ㆍ중ㆍ고등학교 학생들의 인식

        본 연구는 학생 능동 수업 유형과 학생 능동 수업의 교육적 효과에 대한 초ㆍ중ㆍ고등학생들의 인식을 살펴본 것이다. 본 연구는 2016년 7월부터 12월까지 수행되었다. 연구 참여자로는 학생 능동수업의 경험을 가진 초ㆍ중ㆍ고등학교 학생 460명을 대상으로 진행하였다. 연구 결과를 살펴보며, 본 연구를 위해 학생 능동수업 성과 지표를 개발하고, 학생 능동수업 유형별 교육적 효과 차이와 학생 능동수업의 교육적 효과에 대한 인식 차이를 분석하였다. 첫째, 언어적인 활동이 중심이 되는 수업보다 는 학생들은 조작적이며 수공적인 유형의 수업에서 학생 능동 수업의 효과가 높게 나타났다. 고등학생보다는 초ㆍ중학생이, 여학생보다는 남학생이, 학업 성적은 중, 하 수준의 학생보다 상 수준의 학생들이 능동 수업의 모든 유형에서 높은 효과가 나타났다. 6개월에서 1년 미만 능동 수업 경험을 가진 경우에 학생 능동 수업의 교육적 효과가 높게 나타났다. 둘째, 학생 능동 수업의 교육적 효과에서는 모든 영역에서 고르게 높게 나타났다. 그러나, ‘과학적 사고력’, ‘자기 주도 학습 능력’, ‘정보 매체 활용 능력’,‘발표 토론 참여 빈도 증가’ 부분에서는 효과가 나타나지 않는다는 인식이 많았다. 학교급별 효과 차이는 ‘심미적 감성 역량’, ‘자기 관리 역량’, ‘과학적 참여’, ‘자기 주도 학습 시간의 증가’ 부분에서 중학생들의 효과가 초등, 고등학생들에 비해 크게 나타났다. ‘과학적 사고력’, ‘자기관리’, ‘과학적 참여’ 부분에서는 남학생에게 효과가 있었고, 1년 이상 학생 능동 수업을 받은 학생들은 ‘자기 주도 학습 시간의 증가’ 면에서 효과가 크게 나타났다. 따라서, 학생들의 학업 성적과 능동 수업의 경험에 따른 학생들의 잠재적인 발달 수준의 간격을 줄일 수 있는 방안 제시와 학교급별, 성별에 따른 학생 능동수업의 교육적 효과에 대한 차이가 나타나는 원인을 양적 및 질적 분석을 통해 좀 더 심도 깊게 밝히고 구체적인 대안의 마련이 필요하다. This study examined the perceptions of elementary, middle and high school students on the educational effects of student active class types and student active classes. This study was conducted from July to December, 2016. Participants were 460 elementary, midddle, and high school students with active experience in student activities. As a result, first, students active classroom activities were more effective than active classroom activities. The results of this study were as follows. First, the students with high academic achievement were more effective in all types of active class than the students with middle and lower grades. In the case of students who have active class experience for 6 months to less than 1 year, the active effect of student active class is high. Second, the educational effect of student active class was e enly higher in all areas. Howe er, there were many perceptions that scientific thinking , self-directed learning ability , ability to use information media , and The effect of school level difference on the effect of middle school students on aesthetic emotional competence , self-management competency , scientific participation , and increase of self-directed learning time was higher than that of elementary and high school students. Scientific thinking , Self-management , and Scientific participation were effective for male students, and students who received active classes for more than one year were more effective in increasing self-directed learning time. Therefore, it is important to identify ways to reduce the gap between the student s potential developmental level and the educational effect of active classroom activities, depending on the school s grade and gender, It is necessary to clarify more deeply through the analysis and to prepare specific alternatives.
