        Kinesin Family Member 11 Enhances the Self-Renewal Ability of Breast Cancer Cells by Participating in the Wnt/β-Catenin Pathway

        Purpose: Our previous studies have shown that kinesin family member 11 (KIF11) is markedly overexpressed in human breast cancer cells or tissues and positively correlated with distant metastasis and prognosis in patients with breast cancer, suggesting an important role in the regulation of cancer stem cells. Herein, we examined the role of KIF11 in breast cancer stem cells. Methods: In the current study, we validated our previous findings through analysis of data collected in The Cancer Genome Atlas. Endogenous KIF11 was stably silenced in MCF-7 and SKBR-3 cells. Flow cytometry was used to measure the proportion of side-population (SP) cells. Mammosphere culture and tumor implantation experiments in immunodeficient mice were used to assess the self-renewal ability of breast cancer cells. Real-time polymerase chain reaction, western blot, immunofluorescence staining, luciferase reporter assays and Wnt agonist treatment were conducted to investigate the signaling pathways regulated by KIF11. Results: We found that the expression level of KIF11 was positively correlated with stem cell-enrichment genes. The proportion of SP cells was significantly reduced in KIF11-silenced cells. Silencing endogenous KIF11 not only reduced the size and number of mammospheres in vitro, but also reduced the ability of breast cancer cells to form tumors in mice. Simultaneously, we found that KIF11 was involved in regulating the activation of the Wnt/β-catenin signaling pathway. Conclusion: Endogenous KIF11 enhances the self-renewal of breast cancer cells by activating the Wnt/β-catenin signaling pathway, thereby enhancing the characteristics of breast cancer stem cells.

        454 Pyrosequencing Analysis of Bacterial Diversity Revealed by a Comparative Study of Soils from Mining Subsidence and Reclamation Areas

        Significant alteration in the microbial community can occur across reclamation areas suffering subsidence from mining. A reclamation site undergoing fertilization practices and an adjacent coal-excavated subsidence site (sites A and B, respectively) were examined to characterize the bacterial diversity using 454 high-throughput 16S rDNA sequencing. The dominant taxonomic groups in both the sites were Proteobacteria, Acidobacteria, Bacteroidetes, Betaproteobacteria, Actinobacteria, Gammaproteobacteria, Alphaproteobacteria, Deltaproteobacteria, Chloroflexi, and Firmicutes. However, the bacterial communities` abundance, diversity, and composition differed significantly between the sites. Site A presented higher bacterial diversity and more complex community structures than site B. The majority of sequences related to Proteobacteria, Gemmatimonadetes, Chloroflexi, Nitrospirae, Firmicutes, Betaproteobacteria, Deltaproteobacteria, and Anaerolineae were from site A; whereas those related to Actinobacteria, Planctomycetes, Bacteroidetes, Verrucomicrobia, Gammaproteobacteria, Nitriliruptoria, Alphaproteobacteria, and Phycisphaerae originated from site B. The distribution of some bacterial groups and subgroups in the two sites correlated with soil properties and vegetation due to reclamation practice. Site A exhibited enriched bacterial community, soil organic matter (SOM), and total nitrogen (TN), suggesting the presence of relatively diverse microorganisms. SOM and TN were important factors shaping the underlying microbial communities. Furthermore, the specific plant functional group (legumes) was also an important factor influencing soil microbial community composition. Thus, the effectiveness of 454 pyrosequencing in analyzing soil bacterial diversity was validated and an association between land ecological system restoration, mostly mediated by microbial communities, and an improvement in soil properties in coalmining reclamation areas was suggested.

        Crack Resistance and Strength Properties of Red Clay Modified with Lignocellulose and Guar gum

        Desiccation cracking is a common problem of red clay under wet and dry conditions. The presence of cracks could reduce soil strength properties, which is the major reason for shallow failure of red clay slopes. This study aims to propose some additives to improve the crack resistance as well as the strength of red clay. To this end, three natural fibers and three natural gels were primarily selected to modify red clay. Desiccation crack tests were conducted to identify the best fiber and gel in terms of crack resistance. Then, direct shear tests and uniaxial tensile tests were performed on the soil specimens modified by the best fiber and gel to determine the optimal dosages regarding strength improvements. The results demonstrated that natural gels reduced the crack resistance of red clay, while natural fibers could enhance the crack resistance. Lignocellulose was the best additive in improving the crack resistance of red clay, and the modified effect was positively correlated with the fiber dosage. Adding 1.5% lignocellulose into red clay could reduce the crack rate from 2.19% to 0.30%, with a reduction of 0.86 times. The tensile strength, shear strength and its parameters of lignocellulose-modified red clay showed a trend of increasing first and then decreasing with increasing lignocellulose dosage. At the optimal lignocellulose dosage of 0.75%, the shear strength and tensile strength were increased by 44.63% and 18.21%, respectively. The strength of red clay modified by both lignocellulose and guar gum was positively correlated with the dosage of guar gum, and the desirable dosage of guar gum was 1.0%. In engineering practice, the wet red clay mixed with 0.75% lignocellulose and grass seeds can be employed as planting soil and is sprayed uniformly onto the slope surface after excavation using a spraying planting machine. To further improve the strength of the planting soil, 1.0% guar gum solution can be sprayed on the soil surface. The findings could provide useful guidance for the protection of red clay slopes.

        Numerical Investigation of the Turbulent Cavitating Flow over Submerged Bodies

        A numerical method for the calculation of turbulent cavitating flow over submerged objects is proposed in present work. Cavitation is modeled via a single-fluid cavitation model which is derived based on a truncated form of the Rayleigh-Plesset equation and the mixture multiphase theory. The approach has been implemented by user-define function which is widely used in ANSYS FLUENT. Detailed results are presented for sheet cavitation over a submerged hemispherical object in a wide range of cavitation numbers and the cloud cavitation around a Clark-Y hydrofoil. In particular, for the hemispherical body, we compared the surface pressure distribution with experimental data which was available in literature. Later the cloud cavitation structure and its effect on the forces of the hydrofoil were studied. The comparisons between the simulating and experimental results show that present numerical approach has good capability to predict the surface pressure coefficient and the pulsation frequency at cavitation number σ=0.4, 0.55 and 0.65 of the hemispherical body under cavitation conditions. Meanwhile, for the hydrofoil, the proposed approach is sufficiently robust to predict the characteristics of the time-averaged lift and drag coefficients and the evolution of the cloud cavity with time.

        Study on the Crystalline Structure and the Thermal Stability of Silver-oxide Films Deposited by Using Direct-current Reactive Magnetron Sputtering Methods

        Silver-oxide (AgxO) films were deposited on glass substrates by direct-current reactive magnetron sputtering at different oxygen flux ratios (OFR = [O2]/[Ar]) and substrate temperatures (Ts). An X-ray diffraction analysis indicates that the AgxO films are biphased (Ag + Ag2O) when deposited at low OFR values and that Ag2O-dominated AgxO film can only be synthesized at higher OFR values, as con¯rmed by X-ray photoelectron spectroscopy. This result may be due to the Ag2O phase being preferably produced at high OFR value. The AgO phase is thermodynamically unsta-ble compared with the Ag2O phase. In order to further offer deep insight into the ¯lm's thermal stability, Ag2O-dominated AgxO ¯lms were thermally treated by using a rapid thermal processing technique at different annealing temperatures for different annealing times. The Ag2O phase is thermodynamically stable at temperatures below the threshold of the thermal decomposition tem-perature which approaches 175 ±C. The domination of the Ag2O phase in the AgxO film may be attributed to the chain reaction AgO → Ag2O ↔ Ag + O, AgO + Ag → Ag2O.

        The use of fluorescent techniques in combination with flow cytometry for fast counting of Bifidobacterium longum ATCC BAA-2753 in BIFICO capsule

        BIFICO is a main microecological drug that the main ingredient is Bifidobacterium longum ATCC BAA- 2753. It’s necessary to detect the number of Bifidobacterium longum ATCC BAA-2753 for quality control during the production of BIFICO. Plate count assay (PCA) is the most commonly used method for counting microbial cells. However, not all microorganisms can be counted by PCA. (e.g. uncultured microorganisms under specific growth conditions). In this study, a method of fluorescent techniques in combination with flow cytometry (FCM) was established for the counting of Bifidobacterium longum BAA-2753. Using SYTO9 and PI stain, flow cytometric analysis could easily differentiate live bacteria and dead bacteria. The number of live bacteria determined by FCM was about fourfold higher than that counted by PCA. Therefore, the FCM method could be a practical tool for the quality control of Bifidobacterium longum BAA-2753 in the industrial production of BIFICO.

        Implementation and verification of a four-probe motion error measurement system for a large-scale roll lathe used in hybrid manufacturing

        <P>In this paper, a four-probe measurement system is implemented and verified for the carriage slide motion error measurement of a large-scale roll lathe used in hybrid manufacturing where a laser machining probe and a diamond cutting tool are placed on two sides of a roll workpiece for manufacturing. The motion error of the carriage slide of the roll lathe is composed of two straightness motion error components and two parallelism motion error components in the vertical and horizontal planes. Four displacement measurement probes, which are mounted on the carriage slide with respect to four opposing sides of the roll workpiece, are employed for the measurement. Firstly, based on the reversal technique, the four probes are moved by the carriage slide to scan the roll workpiece before and after a 180-degree rotation of the roll workpiece. Taking into consideration the fact that the machining accuracy of the lathe is influenced by not only the carriage slide motion error but also the gravity deformation of the large-scale roll workpiece due to its heavy weight, the vertical motion error is thus characterized relating to the deformed axis of the roll workpiece. The horizontal straightness motion error can also be synchronously obtained based on the reversal technique. In addition, based on an error separation algorithm, the vertical and horizontal parallelism motion error components are identified by scanning the rotating roll workpiece at the start and the end positions of the carriage slide, respectively. The feasibility and reliability of the proposed motion error measurement system are demonstrated by the experimental results and the measurement uncertainty analysis.</P>

      • Expression and Prognostic Implications of FOXO3a and Ki67 in Lung Adenocarcinomas

        To investigate the significance of FOXO3a and Ki67 in human lung adenocarcinomas. Envision immunohistochemical staining and Western blotting were used to examine the protein expression of FOXO3a in 127 cases of human lung adenocarcinoma specimens. The positive rate in lung adenocarcinoma (55.9%) was lower than that in normal tissues (80%). We found that the expression of FOXO3a was closely related with the degree of differentiation, TNM staging, lymph node metastasis and survival. In addition, significant differences in the different pathological types of lung adenocarcinoma cases (P<0.01). The FOXO3a positive rate of the acini as the main type (APA) (86.7%) and the lepidic as the main type (LPA) (82.4%) was higher than the solid as the main type (SPA) (50.0%), the papilla as the main type (PPA) (42.9%) and the micropapilla as the main type (MPA) (9.4%). Moreover, the expression of FOXO3a was negatively related with Ki67 expression. Our results suggested that the expression of FOXO3a is closely correlated with the aggressiveness of lung adenocarcinoma. It was indicated that disregulation of FOXO3a might play key roles in the occurrence and development of lung a denocarcinoma and joint detection of the two markers might play an important role in diagnosing tumors.

        Optimal feedrate planning on a five-axis parametric tool path with global geometric and kinematic constraints

        The optimal feedrate planning problem for the five-axis parametric tool path remains challenging due to the nonlinear relationships between the joint space and the Cartesian space. We present a novel and complete optimal feedrate planning method for a five-axis parametric tool path by constraining the velocity, acceleration, jerk in the joint space, and the chord error in the Cartesian space. Our method formulates the problem as an optimal control problem, and we propose an iteration control vector parametrization method to compute the optimal solution. Compared with the new development five-axis feedrate planning methods, our solution satisfies “bang-bang” optimal control, and each constraint is strictly under the limits globally. Examples and comparisons with two other methods are demonstrated to show the effectiveness of the algorithm.

        Effect of the Oxygen Flux Ratio on the Structural and the Optical Properties of Silver-oxide Films Deposited by Using the Direct-current Reactive Magnetron Sputtering Method

        Using values of the oxygen flux ratio (OFR = [O2]/[Ar]) ranging from 0 to 0.5, authors deposited a series of silver-oxide (Ag_xO) films on glass substrates by direct-current reactive magnetron sputtering (DC sputtering) at a substrate temperature of 150 ℃. The effect of the OFR on the film’s structural and optical properties was systematically investigated by using X-ray diffractometry, scanning electron microscopy and spectrophotometry. The Ag_xO films deposited clearly show an evolution of the film’s phase structure from the biphased (Ag + Ag_2O) structure to the biphased (AgO + Ag_2O) structure and then to the single-phased (Ag_2O) structure as value of the OFR increases. Accordingly, the film’s surface morphology, related to the film’s crystalline structure, clearly changes from a loose and porous surface structure to a compact surface structure and then to a pyramid-like surface structure with increasing value of the OFR. The novel porous structure may be attributed to the interruption of the silver’s growth course by the AgO on the film’s surface. Notably, a single-phased Ag_2O film is deposited by DC-sputtering at OFR = 0.5 due to the dual effects of thermal decomposition of the AgO phase and a combination reaction of AgO and Ag to Ag_2O. The oscillations both in the film’s reflectivity and transmissivity spectra are strengthened with increasing OFR, indicating an evolution from the metallic behavior of the biphased (Ag + Ag_2O) film to the dielectric behavior of the biphased (Ag_2O + AgO) film and the single-phased Ag2O film. The fitted optical absorption edges of the Ag_2O and the Ag_xO films deposited at values of the OFR of 0.5 and 0.33 are approximately 2.43 eV and 2.34 eV, respectively. The absorption edges are closely related to the direct interband transitions.
