        Hsa-miRNA-143-3p Reverses Multidrug Resistance of Triple-Negative Breast Cancer by Inhibiting the Expression of Its Target Protein Cytokine-Induced Apoptosis Inhibitor 1 In Vivo

        Purpose: Multidrug resistance (MDR) remains a major obstacle in the treatment of triple-negative breast cancer (TNBC) with conventional chemotherapeutic agents. A previous study demonstrated that hsa-miRNA-143-3p plays a vital role in drug resistance of TNBC. Downregulation of hsa-miRNA-143-3p upregulated the expression of its target protein cytokine-induced apoptosis inhibitor 1 (CIAPIN1) in order to activate MDR, while upregulation of hsa-miRNA-143-3p effectively enhances the sensitivity of drug-resistant TNBC cells to chemotherapeutics. The present study aimed to further verify these findings in vivo. Methods: We established a hypodermic tumor nude mice model using paclitaxel- resistant TNBC cells. We expressed ectopic hsa-miRNA- 143-3p under the control of a breast cancer-specific human mammaglobin promoter that guided the efficient expression of exogenous hsa-miRNA-143-3p only in breast cancer cells. Thereafter, we overexpressed hsa-miRNA-143-3p in xenografts using a recombinant virus system and quantified the expression of hsa-miRNA-143-3p, CIAPIN1 protein, and proteins encoded by related functional genes by western blot. Results: We successfully completed the prospective exploration of the intravenous virus injection pattern from extensive expression to targeted expression. The overexpression of hsa-miRNA-143-3p significantly alleviated chemoresistance of TNBC by inhibiting viability. In addition, we observed that the expression of CIAPIN1 as a hsa-miRNA-143-3p target protein was remarkably decreased. Conclusion: We partly illustrated the mechanism underlying the hsa-miRNA-143-3p/CIAPIN1 drug resistance pathway. HsamiRNA- 143-3p as a tumor suppressive microRNA may be a novel target to effectively reverse MDR of TNBC in vivo.

        Effect of Glucagon-like Peptide 2 on Tight Junction in Jejunal Epithelium of Weaned Pigs though MAPK Signaling Pathway

        The glucagon-like peptide 2 (GLP-2) that is expressed in intestine epithelial cells of mammals, is important for intestinal barrier function and regulation of tight junction (TJ) proteins. However, there is little known about the intracellular mechanisms of GLP-2 in the regulation of TJ proteins in piglets' intestinal epithelial cells. The purpose of this study is to test the hypothesis that GLP-2 regulates the expressions of TJ proteins in the mitogen-activated protein kinase (MAPK) signaling pathway in piglets' intestinal epithelial cells. The jejunal tissues were cultured in a Dulbecco's modified Eagle's medium/high glucose medium containing supplemental 0 to 100 nmol/L GLP-2. At 72 h after the treatment with the appropriate concentrations of GLP-2, the mRNA and protein expressions of zonula occludens-1 (ZO-1), occludin and claudin-1 were increased (p<0.05). U0126, an MAPK kinase inhibitor, prevented the mRNA and protein expressions of ZO-1, occludin, claudin-1 increase induced by GLP-2 (p<0.05). In conclusion, these results indicated that GLP-2 could improve the expression of TJ proteins in weaned pigs' jejunal epithelium, and the underlying mechanism may due to the MAPK signaling pathway.

      • Investment and Determinants of Investment Cash Flow Sensitivity : Evidence from Privately Listed Chinese Firms

        This study examines firm-level capital investment efficiency and the determinants of investment cash flow sensitivity (CFS). Using financial and ultimate ownership structure data from China, we show that privately listed Chinese firms with high accounting quality tend to increase investment expenditures and have underinvestment problems. Previous studies have claimed that a positive relation exists between investment and cash flow in China, but our results do not support high investment-CFS caused by agency costs. By contrast, our results suggest that firms with high investment-CFS may be highly constrained financially. We also find that a negative relation exists between investment opportunity and investment-CFS. In addition, younger firms face higher investment-CFS.

      • Roles of E-Cadherin (CDH1) Genetic Variations in Cancer Risk: a Meta-analysis

        E-Cadherin (CDH1) genetic variations may be involved in invasion and metastasis of various cancers by altering gene transcriptional activity of epithelial cells. However, published studies on the association of CDH1 gene polymorphisms and cancer risk remain contradictory, owing to differences in living habits and genetic backgrounds. To derive a more better and comprehensive conclusion, the present meta-analysis was performed including 57 eligible studies of the association between polymorphisms of CDH1 gene promoter -160 C>A, -347 G>GA and 3'-UTR +54 C>T and cancer risk. Results showed that these three polymorphisms of CDH1 were significantly associated with cancer risk. For -160 C>A polymorphism, -160A allele carriers (CA and CA+AA) had an increased risk of cancer compared with the homozygotes (CC), and the similar result was discovered for the -160A allele in the overall analyses. In the subgroup analyses, obvious elevated risk was found with -160A allele carriers (AA, CA, CA+AA and A allele) for prostate cancer, while a decreased colorectal cancer risk was shown with the AA genotype. For the -347 G>GA polymorphism, the GAGA genotype was associated with increased cancer risk in the overall analysis with homozygous and recessive models. In addition, results of subgroup analysis indicated that the elevated risks were observed in colorectal cancer and Asian descendants. For +54 C>T polymorphism, a decreased risk of cancer was found in heterozygous, dominant and allele models. Moreover, +54T allele carriers (CT, CT+TT genotype and T allele) showed a potential protective factor in gastric cancer and Asian descendants.

        Dimethyl Sulfoxide Suppresses Mouse 4T1 Breast Cancer Growth by Modulating Tumor-Associated Macrophage Differentiation

        Purpose: The universal organic solvent dimethyl sulfoxide (DMSO)can be used as a differentiation inducer of many cancer cells andhas been widely used as a solvent in laboratories. However, itseffects on breast cancer cells are not well understood. The aimof this study is to investigate the effect and associated mechanismsof DMSO on mouse breast cancer. Methods: We appliedDMSO to observe the effect on tumors in a mouse breast cancermodel. Tumor-associated macrophages (TAMs) were tested byflow cytometry. Ex vivo tumor microenvironment was imitated by4T1 cultured cell conditioned medium. Enzyme-linked immunosorbentassays were performed to detect interleukin (IL)-10 andIL-12 expression in medium. To investigate the cytotoxicity ofDMSO on TAMs, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide (MTT) assays were performed. Results: We foundthat DMSO produced tumor retardation when injected into mouseperitoneal cavities in a certain concentration range (0.5–1.0 mg/g). Furthermore, as detected by flow cytometry, TAM subtypeswere found to be transformed. We further imitated a tumor microenvironmentin vitro by using 4T1 cultured cell conditionedmedium. Similarly, by using low concentration DMSO (1.0%–2.0% v/v), TAMs were induced to polarize to the classically activatedmacrophage (M1-type) and inhibited from polarizing intothe alternatively activated macrophage (M2-type) in the conditionedmedium. IL-10 expression in tumors was reduced, whileIL-12 was increased compared with the control. Furthermore, wereported that 2.0% (v/v) DMSO could lead to cytotoxicity in peritonealmacrophages after 48 hours in MTT assays. Conclusion:Our findings suggest that DMSO could exert antitumor effects in4T1 cancer-bearing mice by reversing TAM orientation and polarizationfrom M2- to M1-type TAMs. These data may providenovel insight into studying breast cancer immunotherapy.

      • The Brainwave Observation of Synchronization Spectrum Effect under Stimulation of Homemade Binaural Beats Piano Music

        With the pressure of society and the large population of insomnia, the sleep problem is becoming more and more serious as the population ages. This study hopes to replace medicines with music and enable patients to relax and enter sleep more quickly. According to clinical experiments, hemi-sync can relax the body and mind and improve the difficulty of sleep. In this experiment, 3.4Hz binaural beats with three different styles of our own sleeping piano music were tested on college students. Emotiv 14-channel wireless EEG device was used to measure and record, analyze the experimental data and present as the spectrogram. For each music beats style, 30 students were selected to perform listening experiments. Calculated the success rate of hemi-sync generated by EEG device, and compared the degree of influence of the three music pieces on the listeners and the differences of frequency band in EEG. The experiment results showed that the hemi-sync appear, with a success rate of more than 40%. Different styles of binaural beats piano music and hemi-sync provide a variety of styles of self-created beats piano music for the public to choose, to shorten the time required to get into sleep and solve the problem of insomnia. In addition, we must observe the feasibility of hemi-sync to the subjects. Using the machine learning algorithm, it was found that in the F7 of the brain area, the accuracy of using the RF algorithm can reach 70.1%, while using XGB it reaches 70.4%, which means that the detection of correlation of hemi-sync and F7 in EEG can improve the verification of the influence of binaural beats on EEG. It shows a clear picture in the future that the beats music can influence the brain, so that people with insomnia can be treated with the most suitable music.

        The Physiological Functions and Polymorphisms of Type II Deiodinase

        Type II deiodinase (DIO2) is thought to provide triiodothyronine (T3) to the nucleus to meet intracellular needs by deiodinating theprohormone thyroxine. DIO2 is expressed widely in many tissues and plays an important role in a variety of physiological processes,such as controlling T3 content in developing tissues (e.g., bone, muscles, and skin) and the adult brain, and regulating adaptive thermogenesis in brown adipose tissue (BAT). However, the identification and cloning of DIO2 have been challenging. In recent years,several clinical investigations have focused on the Thr92Ala polymorphism, which is closely correlated with clinical syndromessuch as type 2 diabetes, obesity, hypertension, and osteoarthritis. Thr92Ala-DIO2 was also found to be related to bone and neurodegenerative diseases and tumors. However, relatively few reviews have synthesized research on individual deiodinases, especiallyDIO2, in the past 5 years. This review summarizes current knowledge regarding the physiological functions of DIO2 in thyroid hormone signaling and adaptive thermogenesis in BAT and the brain, as well as the associations between Thr92Ala-DIO2 and bone andneurodegenerative diseases and tumors. This discussion is expected to provide insights into the physiological functions of DIO2 andthe clinical syndromes associated with Thr92Ala-DIO2.

        Carbon Paper-Supported NiCo/C–N Catalysts Synthesized by Directly Pyrolyzing NiCo-Doped Polyaniline for Oxygen Reduction Reaction

        In this study, we report the findings that the C–N composites containing Ni and Co (Ni1Co1/C–N, Ni3Co1/C–N, Ni6Co1/C–N, Ni9Co1/C–N, Ni10Co0/C–N and Ni0Co10/C–N) can be produced by direct pyrolysis of the NiCo-doped polyaniline (PANI) precursors in N2 atmosphere at 800 ℃ and show efficient electroactivity for oxygen reduction reaction (ORR) in alkaline media. Distribution and compositions of the catalysts were characterized by SEM, TEM, EDS and XRD techniques. The catalysts were loaded on carbon paper to prepare gas diffusion electrodes, in which electrocatalytic activity for ORR in alkaline media was investigated by voltammetric techniques. The ORR current density on these carbon paper-supported NiCo/C–N catalysts exhibits a linear increase with the negative shift of ORR potential. The ORR onset potential is around 0.2 V (versus Ag/AgCl) in alkaline media. Among the prepared catalysts, the catalyst Ni6Co1/C–N presents the largest ORR current density, which is 68.5 mA cm -2@-0.8 V (versus Ag/AgCl) in alkaline media. Moreover, Ni6Co1/C–N catalyst also presents good electrocatalytic activity stability for ORR.

        Short-term, Multi-center Prospective Clinical Study of Short Implants Measuring Less Than 7mm

        Objective : This prospective study sought to verify the stability of three types of short implants measuring 7mm or less. Materials and methods : Implants measuring 7mm or less were placed in patients at multicenter dental clinics in Korea, China, Taiwan, and Singapore. Initial stability, intraoperative and postoperative complications, crestal bone loss, and survival rate of the implant were prospectively evaluated. Results : The primary stability of a 6-mm implant was lower than that of a 7-mm implant. The marginal bone loss of short implants measuring less than 7mm was minimal. Complications such as wound dehiscence, implant mobility, and peri-implant mucositis developed, and these were associated with initial implant failure. The short-term survival rate of 6-mm implant was 93.7%, and that of 7-mm implant, 96.6%. Conclusion : Short implant for the mandible with insufficient height for the residual ridge can be selectively used. Poor primary stability and wound dehiscence can cause osseointegration failure and alveolar bone loss.

        Moth-Flame Optimization-Based Maximum Power Point Tracking for Photovoltaic Systems Under Partial Shading Conditions

        This paper presents a moth-flame optimization (MFO)-based maximum power point tracking (MPPT) method for photovoltaic (PV) systems. The MFO algorithm is a new optimization method that exhibits satisfactory performance in terms of exploration, exploitation, local optima avoidance, and convergence. Therefore, the MFO algorithm is quite suitable for solving multiple peaks of PV systems under partial shading conditions (PSCs). The proposed MFO-MPPT is compared with four MPPT algorithms, namely the perturb and observe (P&O)-MPPT, incremental conductance (INC)-MPPT, particle swarm optimization (PSO)-MPPT and whale optimization algorithm (WOA)-MPPT. Simulation and experiment results demonstrate that the proposed algorithm can extract the global maximum power point (MPP) with greater tracking speed and accuracy under various conditions.
