        Overexpression of Zoysia ZjCIGR1 gene confers cold stress resistance to zoysiagrass

        Zoysia japonica Steud. is a warm-season lawn grass popular in Korea and elsewhere. They are cultivated in many places such as river banks, roadside, and play grounds. However, there still is a disadvantage of frequent mowing, and the grass grows poorly under the chilly conditions. To develop a grass variety that circumvents these drawbacks, we cloned the chitininducible gibberellins-responsive1 gene (CIGR1) from zoysiagrass. The full length of the ZjCIGR1 (Zj; Zoysia japonica Steud.) gene was obtained by 5′/3′ RACE PCR and the phylogenetic tree showed that it belonged to the CIGR1-subgroup in the PAT1-group of GRAS protein family. Expression of the ZjCIGR1 in wild-type plants was confirmed in roots, meristems, leaves, and flowers, especially high in the flowers. The transgenic zoysiagrass was confirmed by PCR using gene-specific primers, phosphinothricin-acetyl-transferase (PAT) strip test, and Southern blot analysis. ZjCIGR1-overexpressing plants acquired tolerance to cold stress displaying morphological phenotypes characteristic of stress resistance. In addition, in the transformants, expression of the ZjCIGR1 as well as cold-regulated (COR) gene was increased compared to the wild-type plants under cold stress condition. These results suggest that ZjCIGR1 gene is an important candidate for regulating cold stress resistance.

        난설헌의 캘리그래피와 아르 데코의 기하학적 문양을 이용한 커피 잔 패턴디자인

        관광을 테마로 하는 중소도시는 지역의 정체성 확립과 대외홍보를 위한 특화사업으로 그 지역만의 문화자원을 발굴하여 다양한 문화상품개발에 적극 활용하고 있다. 이제 문화상품은 그 지역이 갖고 있는 특정한 문화자원을 극대화시켜 지역 경제 활성화를 도모할 수 있는 유용한 수단이 되었다. 현대인들의 상품 선호성향은 상품의 품질 뿐만 아니라 상품 속에서 문화적 가치가 담긴 미적, 예술적 측면까지도 함께 즐기며 체험하고 있다. 따라서 본 연구는 강릉커피축제를 비롯해서 커피에 관련된 문화가 활성화 된 강릉의 지역적 특성을 고려하여 강릉의 역사인물 난설헌 허초희의 캘리그래피를 활용하여 잔(杯) 도자상품의 문양으로 활용할 수 있는 패턴디자인을 개발하고자 한다. 패턴으로 활용할 이미지는 난설헌의 친필서체 캘리그래피와 아르 데코 양식의 기하학적 문양을 접목시켜 창의적인 패턴으로 디자인하고 이를 잔 종류에 적용하여 문화상품에 활용 할 수 있음을 제시하고자 한다. 본 연구에 앞서 연구자는 선행논문으로 난설헌의 친필서체를 텍스타일디자인에 활용하는 패턴디자인을 연구하였다.(양순영,임혜숙2013) 선행논문에서 연구한 패턴디자인은 텍스타일디자인에 국한된 점이 있어 본 연구에서는 초서체의 특징인 곡선과 아르 데코의 기하학적 문양을 접목하여 개발한 패턴디자인을 잔(杯)에 적용시켜 지역의 역사 문화적 요소와 현대디자인의 기하학적 형태가 접목된 새로운 패턴으로 개발하고자 한다. 지역문화를 활용한 패턴디자인은 타 지역의 도자문화상품과의 차별화를 도모하고 독창적인 지역문화상품으로서 강릉의 이미지 구축과 홍보에 기여할 것으로 기대한다. Tourism-themed small local cities have explored their own cultural resources and developed diverse cultural products to establish and advertise their own local identity. Now, cultural products are a useful means to promote local economy by maximizing the strength of local cultural resources. In the modern times, people tend to see cultural values from both artistic and aesthetic aspects when they buy a product as well as the quality of the product. Therefore, this study attempted to develop pattern design which could be used as a cup pattern in ceramic ware using the handwriting of Nanseolheon Heo Cho-hui who was born in Gangneung considering local characteristics in which coffee-related culture is flourished including Gangneung Coffee Festival. For images to be used as patterns, Nanseolheon`s handwriting and Art Deco`s geometric patterns were mixed and designed in creative patterns. Then, they were applied to tea deigns to develop them into cultural products. Prior to this study, the pattern design in which Nanseolheon`s handwriting was used for textile design was studied. (Yang Soon-yeong, Im Hye-sook, 2013) In previous studies, pattern design was limited to textile design. Therefore, this study tried to develop new patterns which integrated local cultural factors and modern design after applying the developed pattern design to a cup. It appears that local culture-based pattern design would make Gangneung`s ceramic cultural products differ from those in other regions and make contribution to the development and spread of its own image as a unique local cultural product.

        신뢰 기반 경매 시스템에서의 퍼지를 이용한 신뢰도 측정과 신뢰도에 따른 거래 방식 추천

        The explosive growth of the internet-based transactions requires not only a secure payment system but also an appropriate trust measuring methodology and secure transaction protocols to guarantee the minimal risk for the transacting entities involved in specific transaction. Especially, in internet auction systems where either buyers or sellers or both can be more than one in one transaction, providing those system that make sure no one transacting entity takes a major risk becomes critical. In this paper, an improved trust measuring method using a relationship-based internet community for an auction system is proposed. The proposed system incorporates fuzzy set and calculation concepts to help build trust matrices and models, which is used to measure the level of risk involved in a specific auction trade concerned. Also, to optimize the auction trade process in terms of cost and time, the proposed system recommends a differentiated trade protocol according to the risk level involved in each auction trade. To test the appropriateness of the proposed trusted auction system, a prototype system has been developed under a Widows-NT environment.

        Styrene Oxide에서 2-Phenylethanol(PEA)를 생성하는 Pseudomonas putida Strain 2150-2의 분리 동정

        울산 유공 옥시케미칼 (주) PO/SM 공장 주변의 토양 샘플로부터 스티렌을 분해하여 성장하는 균주를 분리하였는데, 이 균주에 대한 생리·생화학적, 형태학적 특성을 조사한 결과와 DNA G+C 함량에서 Pseudomonas putida로 동정되었다. 이 균주의 스티렌 옥사이드에서 2-phenylethanol로의 대사경로가 연구되었고, 이 균주에 의해 스티렌 옥사이드가 페닐아세트알데하이드로, 다음에 2-phenylethanol(PEA)로, 그리고 페닐초산으로 대사되어 기존의 스티렌 자화 균주와는 다른 페닐초산으로 대사되어 기존의 스티렌 자화 균주와는 다른 대사경로를 가짐을 확인할 수 있었다. The strain which produces 2-phenylethanol (PEA) from syrene oxide was isolated from soil samples near Ulsan PO/SM plant. The isolated strain was identified as Pseudomonas putida through its morphological, physiological characteristics, and DNA G+C contents. Its metabolic pathway from styrene oxide to 2-phenylethanol was studied and it was found that styrene oxide was transformed to phenylacetaldehyde, to 2-phenylethanol (PEA), and then to phenylacetic acid by this strain.

      • 컴포넌트 커널 프레임워크를 이용한 커널의 동적 확장성

        기존의 커널 구조는 응용프로그램의 요구에 따라 커널의 기능을 고치거나 새로이 추가하는 것이 커널 자체의 복잡성으로 인하여 어려운 점이 많고 커널 내의 기능성들을 모듈화 함으로써 어려움을 극복하고자 하고 있다. 하지만 응용프로그램이 다양화되고 이러한 응용프로그램들의 요구에 따라 커널 내의 기능성이 변경되거나 추가됨으로써 이러한 모듈화는 깨어지고 있다. 즉 변경되거나 새로이 추가될 기능성을 미리 예측하는 것이 불가능하기 때문에 어떠한 모듈을 접근하는데 있어 고정된 1차원적인 인터페이스만으로는 모듈화를 유지하기가 힘들게 되고 특히 멀티미디어와 같은 새로운 응용프로그램들은 기존의 커널에 많은 새로운 기능들의 추가를 요구하고 있어서 상황은 더욱 악화되고 있다. 본 논문에서는 이러한 커널 복잡도를 낮추고 커널 구조를 보다 유연하게 만들기 위한 방안으로서 코어커널(CoreKernel)과 커널컴포넌트(KernelComponent)들의 집합으로서 커널을 구성하는 컴포넌트 커널 프레임워크(Component Kernel Framework)를 제시하고 이에 기반한 커널의 동적 확장 모델을 설계한다. 제시한 컴포넌트 기반 커널 확장 모델을 검증하고자 현재 개발중인 커널 프로토타입 소개하고 시뮬레이션을 통해 제안된 동적 확장 모델을 검증한다. Traditional kernels like UNIX have lots of difficulties in adding and modifying kernel functionalities due to inherent complexities of kernel architecture. They try to solve these problems through modularization. As there come to be a diversity of application programs and they require modification of existing kernel functionalities and addition of new ones, Modularity of kernel tends to be broken. In other words, because it is difficult to expect which functionality should be modified and what functionality should be newly added, To keep modularity of kernel is almost impossible if the modules are accessed only through fixed one-dimensional interface. This situation is likely to happen as multimedia applications that require new kernel functionalities like real-time scheduling and memory reservation become popular. Trying to decrease kernel complexities and to make kernel more modularized, this paper suggests Kernel Component Framework that construct kernel of Corekernel and a set of KernelComponents and then designs dynamic kernel extension model based on Kernel Component Framework. To verify suggested component-based kernel extension model, we introduce kernel prototype now being developed and simulate dynamic extension model.

        To know the effect of tube voltage in X-Ray Radiography. We made an experimental study on the tube voltage, on the exposure dose in taking general examination, and obtained the results as follows. A) In the Radiographic of the Chest P-A; The experimental value of tube voltage was obtained under the control of the applied voltage, the kind of some apparatus and thickness of body. 1) The average chest Radiographic condition was resulted as 65.5Kv, 16.6 mAs in Hospital around the chonnam area 2) It was resulted 21.93cm in average Thickness of the chest, exposure dose 20mR B) The thickness was 19.22cm in the abdomen and then appeared average tube voltage was 71.6Kv, 68.2 mAs C) Estimation could not in difference with because of the working dark room uniformly to hand-operated and automatic process.

        지식관리시스템을 위한 FAH 기반 전문가 검색 방법론

        In Knowledge Management Systems (KMS), tacit knowledge which is usually possessed as forms like know-how, experiences, and etc, is hard to be systemized while managing explicit knowledge is comparatively easy using information technology such as databases. Recent researches in knowledge management have shown that it is more applicable in many ways to provide expert search mechanism in KMS to pinpoint experts in the organizations with searched expertise so that users can contact them for help. In this paper, we propose an intelligent expert search framework to provide search capabilities for experts in similar or related fields according to the user's needs. In enabling intelligent expert searches, Fuzzy Abstraction Hierarchy (FAH) framework has been adopted, through which finding experts with similar or related expertise is possible according to the subject field hierarchy defined in the system. To test applicability and practicality of the proposed framework, the prototype system, knowledge Portal for Researchers in Science and Technology, was developed.

      • RAPD법을 이용한 양파품종(Allium cepa L.)간의 유전변이와 유연관계분석

        This study was carried out to investigate genetic variation and relationships among onion varieties, Allium cepa L., by RAPD-PCR analysis. In the test of 120 primers with 58 onion varieties collected from local and abroad, eight primers turned out to be useful for further analysis. These primers eventually provided a total of 71 bands, consisting of 68 polymorphic and three monomorphic ones. The polymorphic bands were subjected to phylogenetic analysis using NTSYS and PAUP computer programs. In the NTSYS analysis, similarity coefficient of the 58 onion varieties ranged from 0 to 0.9179 and that of 27 varieties with obvious origin ranged from 0.0306 to 0.9032, The dendrogram of 58 varieties obtained by NTSYS analysis resulted in 15 groups, but overall groupings were not consistent with bulb phenotypes (e.g., color, weight, and shape) or origin of the varieties either In the PAUP analysis, genetic divergence of the 58 onion varieties ranged from 0.042 to 0.532 and that of 27 varieties with obvious origin ranged from 0.042 to 0.394. Phylogenetic analysis using PAUP revealed 13 group with 58 varieties and nine group with 27 varieties, but these were not consistent with any bulb phenotypes or origin either, as seen in the NTSYS analysis.

      • 좁쌀약주 釀造를 위한 균주의 선발

        As the manufacturing techniques of foxtail millet wine, a traditional wine in Cheju island, has been transfered by person to person, basic studies on this are not carried out yet. There are many difficulties to industrialize it successfully without improvement of wine quality. The purposes of this study were to screen saccharifying mold strain and brewing yeast for foxtail millet wine-making. Optimum reaction conditions of crude enzyme of Aspergillus oryzae on saccharifying soluble starch were 55℃, pH 5.6, Saccharifying activity of crude enzyme was maintained 31.0~35.4% at 20~25℃, fermentation temperature, compared to optimum enzyme reaction conditions. Saccharomyces cerevisiae IAM 4274 was selected as brewing yeast strain for foxtail millet wine-making.
