      • Sulfonylurea계 제초제를 이용한 올방개 방제 효과 증진 연구

        본 실험은 수도에서 문제 잡초로 대두되는 올방개의 효과적인 방제법을 구명하고자 sulfonylurea계 제초제를 각각의 약제간, 시기별로 구분하여 처리하였고, 주요 결과는 다음과 같다. 1 약제처리시기에 따른 올방개 방제효과는 이앙 후 10일 처리에서 Bensulfuron-methyl, Pyrazosulfurone-thyl, Azimsulfuron, Imazosulfuron, Halosulfuron, Cinosulfuron, Cyclosulfamuron 이상 7종이 올방개 방제효과가 우수했고, 이중 Bensulfuron-methyl, Pyrazosulfuron-ethyl, Azimsulfuron, Imazosulfuron은 이앙 후 20일 처리에서도 방제효과가 우수했다. 2 Pyrazosulfuron-ethyl + Mefenacet 합제를 시기별로 처리한 결과 이앙 후 14일과 21일 처리에서 90%이상의 방제가를 보여 우수한 방제효과를 보였던 반면, 이앙 후 7일과 28일 처리에서는 방제효과가 떨어졌다. 3. Befurasate는 Pyrazosulfuron-ethyl과 Azumsulfuron을 혼합사용하였을 때, Cyclosulfamuron은 Azimsulfuron과 혼합사용하였을 때 올방개의 방제가 증대되는 상승적 작용이 관찰되었다. 4. 이앙 동시처리제인 Imazosulfuron + Fentrazamude 합제, Imazosulfuron + Oxaziclo mefone + Dymron 합제, Bensulfuron-methyl + Fentrazamide합제의 경우 올방개의 방제가가 각각 68%, 76% 66%로, 이앙 후 10일에 처리한 Pyrazosulfuron-ethyl + Mefenacet 합제보다 방제가가 낮아, 올방개가 우점된 논에서의 이앙 동시처리제에 의한 올방개 방제는 어려운 것으로 판단되었다. Sulfonylureas are quite recent low dosage and wide spectrum herbicides, popularly used in paddy rice field. The acetolactate synthase inhibitors are effective for the control of broad leaf and perennial paddy weeds, however for the control of Eleocharis Kuroguwal Ohwi has been reported less than satisfactory. Therefore, present commercial sulfonylureas were evaluated for the control efficacy of Eleocharis Kuroguwal Ohwi and the time of application was studied for the better control. Among the tested sulfonylureas, Bensulfron-methyl, Pyrazosulfuron-ethyl, Azimsulfuron and Imazosulfuron exhibited 80% or better control efficacy at both treatments of 10 and 20 days after transplanting (DAT). While Halosulfuron, Cinosulfuron, and Cyclosulfamuron showed good weed control from 10 DAT treatment, but showed less than desirable results at 20 DAT treatment. Ethoxysulfuron showed low control efficacy at both of the time applied. Benfurasate, alone, appeared not effective for the control of Eleocharis kuroguwal Ohwi. Evaluation for the timing of the herbicide application showed that 14 to 21 DAT treatments appeared appropriate. When applied before the period, the weed regrowth became problematic. On the other hand, when applied the later, the weed outgrew to be controlled with the herbicide. For labor saving for herbicide application, herbicide application at transplanting was sought. However, Elevcharis kuroguwal Ohwi was difficult to control from the earlier application, the herbicide application at transplanting appeared impractical where Elevcharis kuroguwai Ohwi is dominant.

      • 인삼 Peroxidase(PgPrx3) 유전자의 분리 및 연초의 형질전환

        Peroxidase는 항산화에 관련된 효소이며 식물의 환경스트레스와 성장에 관련되는 중요한 유전자이다. 본 실험에서 사용된 Peroxidase Ⅲ의 cDNA는 Panax ginseng C.A. Meyer 잎에서 추출하였으며 PgPrx3라 명명하였다. 이 gene의 ORF영역은 1,065 bP이고 355개의 아미노산을 가지고 있다. BioEdit 프로그램을 이용하여 PgPrx3와 다른 식물의 peroxidase gene과 비교한 결과 상동성을 가지고 있었으며, Spinacia oleracea(70%), Vigna angulais(71%), Nicotiana tabacum(69%) and Linum usitatissimum(65%). 그중 Vigna angularis의 상동성이 가장 높았다. 담배에 본 유전자를 형질전환시켜 유전자의 특성을 검정하기 위하여 35S/35S/AMV/peroxidas/Tnos를 벡터 pRD400에 재조합 후 아그로박테리아를 이용하여 담배에 도입시켰다. 그리고 PCR를 이용하여 유전자 PgPrx3가 담배에 도입된 것을 확인하였으며, RT-PCR로 정상적으로 PgPrx3 유전자가 전사되어 RNA를 생성하고 있음을 확인하였다. A peroxidase (E.C. 1.11.1.7) is very important enzyme as antioxidants. The function of this gene is connected with growth and environmental stress of plant. A class Ⅲ peroxidase cDNA was isolated from the leaf of Panax ginseng C.A. Meyer and named as PgPrx3 which is consisted of an ORF (open reading frame) of 1,065 bp and an amino acid of 355 residues. BioEdit software was used to compare the PgPrx3 amino acid sequence with other plants that showed homologies with Spinacia oleracea (70%), Vigna angularis (71%), Nicotiana tabacum (69%) and Linum usitatissimum (65%). The peroxidase of Vigna angulatis was the most homologous with ginseng. The chimeric PgPrx3 gene, 35S/35S/AMV/peroxidas/ Tnos, was constructed in the binary vector pRD400. Agrobacterium tumefaciens strain LBA4404 harboring disarmed Ti-plasmid was used to transform Nicotiana tabacum L. using the leaf disc. Incorporation of the chimeric gene PgPrx3 into plants were confirmed by PCR analysis of genomic DNA and RT-PCR analysis of mRNA from transgenic tobacco. The PgPrx3 gene from ginseng was stably expressed in transgenic tobacco plants.

      • 더덕의 효율적인 재분화 및 뿌리발달

        더덕은 식용, 관상용, 생약재료로 쓰이는 방향성 식물이다. 본 연구는, 종자의 기내배양을 통하여 식물체 재분화와 뿌리발달을 실험했다. 종자의 발아는 GA_(3) 100 mg·L(-1)처리와 저온처리 시 매우 양호하였다. 더덕의 기내에서 shoot의 형성은 2 mg·L^(-1) BA, 2 mg·L(-1) NAA가 첨가된 MS 기본배지에서 가장 양호하였으며, 줄기의 절편체와 비교하여 잎 절편체에서 재분화가 잘되었다. 또한 뿌리의 발육은 계속적인 계대배양에 의해서 양호하였는데 shoot의 길이를 1 cm 정도 남긴 뿌리를 연속적으로 4회 계대배양에서 뿌리가 주근으로 발육하였으며 길이는 약 3 cm 및 직경은 0.5 cm의 정상적인 뿌리를 얻을 수 있었다. Codonopsis lanceolata is widely used as edibles, ornamental plant and crude drug material. This study was carried out to confirm the ratio of germination and efficient differentiation of shoot and root in vitro. The germination of Codonopsis lanceolata seed was excellent in the medium with GA_(3) 100 mg·L(-1) and chilling treatment. The shoot formation of Codonopsis lanceolata was good at the MS medium with 2 mg·L(-1) BA and 2 mg·L(-1) NAA. According to inoculum part, ratio of re-differentiation was higher from the leaf explant compared with the stem explant. For root development, successive subculture with 1 cm shoot of upper part from root was efficient. Roots of Codonopsis lanceolata cultured in vitro system was grown like normal root with length of 3 cm and thickness of 0.5 cm at the 4 successive subcultures.

        하나(일(一))와 여럿(다(多))의 상호전환 작용으로서 아리스토텔레스의 예술이론

        The core of an Aristotle`s theory on art is the concept of Mimesis as an imitation of `the universal`. In Aristotle, the art as the imitation after the nature means to imitate `the highest`, `the probabilities` and it must contain the vividness and verisimilitude, extracting the universals from the individuals before individuating it again. According to Aristotle, as `the universal` is contained in `the individual` art which imitates `the universal` must depict `the universal` of eidos that will come true in the future. As `the universal` of eidos is `entelekeia`, after all, art has to be concerned it. Therefore He asserts that the poem is more philosophical than the history. Because philosophy is concerns the universal truth, and the poem describes `the universal`, but the history utters the simple individual affairs. We can think that Mimesis intervenes in the poem not in the history. This mimesis is the work which conversions `the individual` into `the universal` and vice versa. In other words, excellent artist shall insight an abstract universal from `the individual` and he can turn over it into `theindividual` in real world again. And aristotle also points out another gift of a good poet. It is the ability of commend on metaphor in poem. The metaphor, according to Aristotle, is an ability which can find out the similarity among the difference for the nature of poetry and it is to translate the name of the one thing to the another. As the nature of the metaphor is an analogy, the metaphor means to see a thing in terms of the sight of the other and to be able to offer the cognitive extension by a new sight. Here, what is called a new sight means the very imagination. Because, essentially, the imagination is to look upon the image of one thing as another. Aristotle asserted that it is a sign of genius to be good at a metaphor, for the skillfulness of metaphor means to detect the likeness among the unlikeness in rapid. Now the writing of poem depends upon the ability to conversion `the individual` with `the universal` and to be good at the metaphor attributed to imagination. These Aristotle`s concept of mimesis and his theory on art has influenced the neo-classism afterwards. And his concept of mimesis is fixed as that of imitation on the universal thing in real world.

        Effects of Growth Retardants on Lodging-related Traits in Direct Seeded Rice on Flooded Paddy Surface

        This study examined the effects of growth retardants and midsummer drainage on lodging-related characters in direct seeding rice culture on flooded paddy surfaces. Treatments included in this study were continuous flooding, two time midsummer drainages (TTD), and inabenfide or trinexapac-ethyl application after TTD. Culm length was reduced most by 11.4cm in trinexapac-ethyl treatment after TTD, followed by 4.1cm in inabenfide treatment after TTD compared to TTD treatment. A significant reduction in culm length occurred at the 3rd internode in inabenfide treatment applied at 40 days before heading (DBH), while trinexapac-ethyl application at 8 DBH reduced the 3rd>2nd>1st internode. The culm diameter of the 4th internode was increased by the application of trinexapac-ethyl following TTD compared to TTD treatment. Thickness of culm wall was significantly increased by trinexapac-ethyl but not by inabenfide applications. Lodging index was the highest in continuous flooding and the lowest in trinexapac-ethyl treatment after TTD. Subsequently, field lodging did not occur in plots treated by trinexapac-ethyl. Rice plants subjected to midsummer drainage or growth retardants showed higher ripened grain ratio and 1000-grain weight, resulting in higher rice yield. These results indicate that trinexapac-ethyl, which can be applied at the relatively later growth stage after proper judgement of lodging occurrence, could be a useful tool for reducing lodging in direct seeding rice culture on flooded paddy fields

        Occurrence and Distribution of Weedy Rice in Kyonggi Region

        Distribution and occurrence of weedy rice in Kyonggi region were surveyed in 1996. Weedy rice was observed in 1368 fields (54.9%) of total 2490 fields. Almost two thirds of paddy fields in northern mountainous region were contaminated by weedy rice and more severe contamination, three forths of paddy fields, was observed in suburban regions. In those regions, occurrence of weedy rice was greater than those in north-eastern inland and south-western plain regions. The occurrence of weedy rice was higher in water seeding cultivation (66.7%) than other cultivation methods. The number of weedy rice per 10a was 756.7 plants in direct seeding on dry paddy and 379.4 plants in water seeding. The occurrence of weedy rice was higher in fields planted by farmer's seeds than that of paddy fields cultivated by certified seeds, and the longer the farmer's seeds being used, the more weedy rice occurred in paddy field

        Background : Currently, obesity and adult diseases as a result of sugar intake have been a consistent problem in Korea. Natural sweeteners as sugar substitutes have many advantages over sugar as a small quantity. Therefore, this study was conducted to investigate possibility of industrialization as food additives by using the nature sweetness components of stevia (Stevia rebaudiana Bertoni). Method and Results : Stevia was cultivated in a plastic house. The leaves and stems were harvested at October. They were ground into fine powder using a mill, and were extracted by high temperature and pression extraction method. The extracts were evaporated under vacuum and lyophilized. Three strawberry cultivars of ‘Seolhyang’, ‘Maehyang’, and ‘F22-196’ were cultivated in a plastic house and were harvested from March to May. The fruits harvested were stored at 50℃ until processing. In order to test the processing suitability of stevia extract, the characteristics of the three strawberry varieties (line) were investigated. As a result, the ‘F22-196’ line, which was bred as a processing strawberry, generally contained more antioxidant materials and activity than those of ‘Maehyang’ and ‘Seolhyang’ varieties. Comparing the sugar contents which affects the quality of strawberry jam, the average sugar contents of ‘F22-196’ line was higher than 'Maehyang' and 'Seolhyang' varieties. In the preparation of strawberry jam using ‘F22-196’, strawberry jam was prepared by adding only sugar or stevia extract powder, which was 1/100 of the amount of the sugar in only sugar strawberry jam, to the sensory test. As a result, we identified that sugar jam and stevia jam added stevia extract showed 50 : 50 at the sensory test and stevia jam does not make a difference to the marketing jam at the point of view of general consumers. Conclusion : ‘F22-196’ line represented the best quality for strawberry jam in test caltivars. Stevia powder is judged to be used as a sweetener of sugar substitutes in the production of strawberry jam and processing food.

        Clinical Features of Symptomatic Meckel's Diverticulum in Children: Comparison of Scintigraphic and Non-scintigraphic Diagnosis

        Purpose: Meckel's diverticulum (MD) has various clinical manifestations, and diagnosis or selectection of proper diagnostic tools is not easy. This study was conducted in order to assess the clinical differences of MD diagnosed by scintigraphic and non-scintigraphic methods and to find the proper diagnostic tools. Methods: We conducted a retrospective review ofthe clinical, surgical, radiologic, and pathologic findings of 34 children with symptomatic MD, who were admitted to Gachon University Gil Medical Center, Inha University Hospital, and The Catholic University of Korea, Incheon St. Mary's Hospital between January 2000 and December 2012. The patients were evaluated according to scintigraphic (12 cases; group 1) and non-scintigraphic (22 cases; group 2) diagnosis. Results: The male to female ratio was 7.5: 1. The most frequent chief complaint was lower gastrointestinal (GI) bleeding in group 1 and nonspecific abdominal pain in group 2, respectively. The most frequent pre-operative diagnosis was MD in both groups. Red blood cell (RBC) index was significantly lower in group 1. MD was located at 7 cm to 85 cm from the ileocecal valve. Four patients in group 1 had ectopic gastric tissues causing lower GI bleeding. The most frequent treatment modality was diverticulectomy in group 1 and ileal resection in group 2, respectively. Conclusion: To diagnose MD might be delayed unless proper diagnostic tools are considered. It is important to understand indications of scintigraphic and non-scintigraphic methods according to clinical and hematologic features of MD. Scintigraphy would be weighed in patients with anemia as well as GI symptoms.
