Purpose: To investigate the association between dry eye syndrome (DE) and serum levels of interleukin (IL)-17 in patients with systemic immune-mediated diseases. Methods: IL-17 and IL-23 levels were measured in the sera of patients whose tear production was <5 mm on the Schirmer test. Subjects included patients with chronic graft-versus-host disease (GVHD), rheumatoid arthritis (RA), Sjogren`s syndrome (SS), systemic lupus erythematosus (SLE), and no systemic disease. Corneal/conjunctival fluorescein staining was scored and the correlation between the score and the IL-17 level was evaluated. Results: A strong correlation existed between IL-17 level and the type of systemic disease. IL-17 was significantly elevated in patients with chronic GVHD compared to those with RA and SS. IL-17 was not detectable in patients with SLE or in those without systemic disease. IL-23 was not detected in any of the subjects. IL-17 was significantly increased in patients with high fluorescein staining scores. Conclusions: Our data suggest that IL-17 is involved in the pathogenesis of DE in patients with systemic immune-mediated diseases.

Condyloma acuminatum, an infection caused by human papilloma virus, is one of the most common sexually transmitted diseases involving the anogenital area. Although it has been more common in adults, the incidence in children has been increasing recently. Although several treatment options are available for condyloma in adults, children`s treatment options are limited because non-painful alternatives should be considered as first-line therapy. Imiquimod cream is an immunomodulating agent that has been demonstrated to be a potent inducer of several cytokines promoting an antiviral cell-mediated immune response. Herein, we report two cases of condyloma acuminata in children which showed an excellent response to 5% imiquimod cream without any adverse reactions.

Abnormality in the central noradrenergic system may be related to the seizure prone state in the genetically epilepsy prone rats (GEPR). The present work deals with the characterization of the deficit in noradrenergic system if susceptitibility and intensity of seizure are dependent on central noradrenregic activities by comparing the activities of dopamine $\beta$-hydroxylase (DBH) which hydroxylates dopamine into noradrenaline. DBH activities were measured in 5 areas of brain of normal rats, native GEPR and severe GEPR. The results suggest that lower DBH activities in the midbrain of GEPRs may positively be coupled to the susceptibility to seizure, whereas the same characteristics of the native or severe GEPR are not neccessarily in parallel with the intensity of seizure.

Xenotransplantation 2010; 17: 140–146. © 2010 John Wiley & Sons A/S.</P><P>Abstract: Background: </P><P>We have demonstrated earlier that porcine corneal xenografts underwent an acute cell-mediated rejection in mice despite the absence of T cells. In the present study, we investigated the effect of complement depletion by cobra venom factor (CVF) on the corneal xenograft rejection in a pig-to-mouse model.</P><P>Methods: </P><P>Porcine corneas were orthotopically transplanted into C57BL/6 (B6) and severe combined immunodeficiency (SCID) mice. For complement depletion, 25 <I>&mgr;</I>g of CVF (1 g/kg bodyweight) was injected intraperitoneally on the day before and 1, 3, 5, and 7 days after transplantation. Graft survival was clinically assessed by slit lamp biomicroscopy and the median survival time (MST) was calculated. The grafts were histologically evaluated serially after transplantation using antibodies against CD4, CD8, NK1.1, and F4/80.</P><P>Results: </P><P>The CVF treatment significantly prolonged the porcine corneal xenograft survival in both B6 (MST 9.4 vs. 15.5 days; P = 0.0011) and SCID mice (MST 16.4 vs. 20.5 days; P = 0.0474). Histologically, whereas macrophages and CD4<SUP>+</SUP> T cells were progressively infiltrated into porcine corneal grafts in CVF-untreated B6 mice, the infiltration by both cells was markedly delayed and decreased in the xenografts in CVF-treated B6 mice. Likewise, macrophage infiltration, which was prominent in rejected porcine xenografts in SCID mice, was also reduced in CVF-treated SCID mice.</P><P>Conclusions: </P><P>Our results suggest that complement depletion by CVF delayed, although did not prevent, an acute cell-mediated rejection in a pig-to-mouse corneal xenotransplantation.</P>

<P>Intravenously administered mesenchymal stem/stromal cells (MSCs) engraft only transiently in recipients, but confer long-term therapeutic benefits in patients with immune disorders. This suggests that MSCs induce immune tolerance by long-lasting effects on the recipient immune regulatory system. Here, we demonstrate that i.v. infusion of MSCs preconditioned lung monocytes/macrophages toward an immune regulatory phenotype in a TNF-alpha-stimulated gene/protein (TSG)-6-dependent manner. As a result, mice were protected against subsequent immune challenge in two models of alloand autoimmune ocular inflammation: corneal allotransplantation and experimental autoimmune uveitis (EAU). The monocytes/macrophages primed by MSCs expressed high levels of MHC class II, B220, CD11b, and IL-10, and exhibited T-cell-suppressive activities independently of FoxP3(+) regulatory T cells. Adoptive transfer of MSC-induced B220(+) CD11b(+) monocytes/macrophages prevented corneal allograft rejection and EAU. Deletion of monocytes/macrophages abrogated the MSC-induced tolerance. However, MSCs with TSG-6 knockdown did not induce MHC II+ B220(+) CD11b(+) cells, and failed to attenuate EAU. Therefore, the results demonstrate a mechanism of the MSC-mediated immune modulation through induction of innate immune tolerance that involves monocytes/macrophages.</P>

A drug eruption is any adverse skin reaction caused by a drug used in a normal dose. Celecoxib(Celebrexn^ ), a cyclooxygenase(COX) 2 inhibitor, is a new generation non-steroidal antiinflammatory drug, recently introduced in Korea. It is increasingly used because of significantly lower rate of gastrointestinal injury than typically seen with most other NSAIDs. Cutaneous reactions related to celecoxib are rare and there have been no previous reports in Korea. We report a case of drug eruption probably due to this drug with positive skin tests and wish to underline the possible cutaneous reactions with this new drug.
Vascular rinsing and chilling carcasses improves meat quality and food safety: a review

Rinse & Chill® technology (RCT) entails rinsing the vasculature using a chilled isotonic solution (3℃; 98.5% water and a blend of dextrose, maltose, and sodium phosphates) to rinse out the residual blood from the carcass. Infusion of pre-chilled solutions into intact animal carcasses immediately upon exsanguination is advantageous in terms of lowering the internal muscle temperature and accelerating chilling. This technology is primarily used for purposes of effective blood removal, favorable pH decline, and efficient carcass chilling, all of which improve meat quality and safety. Although RCT solution contains some substrates, the pre-rigor muscle is still physiologically active at the time of early postmortem and vascular rinsing. Consequently, these substrates are fully metabolized by the muscle, leaving no detectable residues in meat. The technology has been commercially approved and in continuous use since 2000 in the United States and since 1997 in Australia. As of January 2022, 23 plants have implemented RCT among the 5 countries (Australia, US, Canada, New Zealand, and Japan) that have evaluated and approved RCT. All plants are operating under sound Sanitation Standard Operation Procedures (SSOP) and a sound Hazard Analysis Critical Control Point (HACCP) program. No food safety issues have been reported associated with the use of this technology. RCT has been adapted by the meat industry to improve product safety and meat quality while improving economic performance. Therefore, this review summarizes highlights of how RCT technically works on a variety of animal types (beef, bison, pork, and lamb).
