Over the last decade, there has been increased interest in the addictive potential of the Internet, and past studies have indicated that the addictive pattern of Internet use are associated with some psycho-social traits of the users and adolescents and young adults are especially vulnerable population. This study investigated the extent to which psycho-social factors were related to the extent of Internet usage among adolescents and to examine the potential influences of self-esteem, depression and impulsiveness on social relations of the adolescents with Internet addiction. A total of 2,495 adolescents ages between 15-17 responded to the questionnaire, which included Young's IAT, Rosenberg's Self-esteem instrument, Beck's BDI, BISII, Hudson's IFR and IPR. Results showed that 61.0% of the sample were considered Internet addicted(4.0% seriously pathological, 57.0% moderate), who suffer from higher level of depression, impulsiveness, and problems of social relations and low self-esteem than their peers. Regression analysis indicated that problems in social relations among adolescents with Internet addiction were predicted by low self-esteem and depression. No link was found between impulsiveness and social relations. Results suggest that preventive efforts for Internet addiction of adolescents are in imminent needs. Implications to help professionals are addressed.
Purification and Characterization of Aspartase from Hafnia alvei

Aspartase (EC 4.3.1.1) from Hafnia alvei was purified to homogeneity by a combination of DEAE-cellulose, Red A-agarose, and Sepharose 6B chromatography. The purified enzyme appeared homogeneous on denatured SDS-polyacrylamide gel electrophoresis. The purified enzyme was a tetrameric protein composed of identical subunits with a molecular weight of 55,000 daltons. The optimum pH for the enzymatic reaction was 8.5 and the optimum temperature for maximum activity was $45^{\circ}C$. The enzyme has an absolute requirement of divalent metal ions ($Mg^{2+}$, $Mn^{2+}$) at the alkaline pH. The enzyme, however, was inactivated in the presence of other divalent cations such as $Zn^{2+}$, $Ca^{2+}$. The helical content of the purified enzyme was estimated by CD spectropolarimetry to be 61%.
Synthesis of Peptides by Bovine Gastricsin

Bovine gastricsin catalyzes peptide synthesis over an optimum range of pH 4~5, resulting in satisfactory yields of methyl esters and p-nitroanilides of benzyloxycarbonyl tetra- to hexa-peptides, provided that hydrophobic amino acid residues form new peptide bonds. The effectiveness of the enzyme also depends on the nature of adjacent amino acid residues. An aspartic proteinase with a characteristic gastricsin specificity pattern would be useful for the synthesis of middle-length peptides.

<P>The two-dimensional nanocarbon material graphene (Gr) has been extensively studied due to its many potential biomedical applications including regenerative medicine, drug delivery, bioimaging, and biosensing. The effects of nitrogen-functionalisation on chemically driven Gr (CDG) cellular responses were studied by investigating the generation of reactive oxygen species (ROS) and mitochondrial morphology as well as focal adhesion, shape, proliferation and viability of HeLa cells grown on functionalised CDG (f-CDG) films. The drop casting of CDG nanosheets formed thin CDG films and the formation of nitrogen groups on the f-CDG thin films was mediated by N2plasma treatment without the formation of observable surface defects. N-containing functional groups on the CDG thin films contributed to an increase in hydrophilicity. The proliferation and viability of HeLa cells grown on the f-CDG thin films were enhanced compared to those grown on CDG films alone and control samples. N-functionalisation of CDG thin films effectively reduced the ROS generated from cells on the f-CDG films. These results indicate that N2plasma treatment of CDG is very useful in improving biocompatibility for the bio-application of graphene materials.</P>
독도의 자생식물 뿌리에서 분리한 내생진균의 다양성과 생장촉진활성

독도에 자생하고 있는 식물의 뿌리로부터 내생진균의 분리를 시도하여, 동도로 부터 참억새, 돌피, 쇠무릎과 서도에서 갯별꽃 및 율무쑥등을 연구재료로 사용하였다. 총 21종의 내생진균을 분리하였고, universal primers ITS-1과 ITS-4를 사용하여 ITS 영역을 PCR로 증폭하여 동정하였다. 염기서열 분석결과, 동도에 자생하는 참억새에서는 Penicillium 속 균주가 75%, Aspergillus 균주가 25%, 분리되었고, 돌피에서는 Penicillium 속 균주가 55%, Aspergillus 속 균주가 30%, Zygorhynchus 속 균주가 15%가 분리되었으며, 쇠무릎에서는 Penicillium 속 균주가 50%, Aspergillus 속 균주가 12%, Gibberella 속 균주가 13%, Talaromyces 속 균주가 9%, Umbelopsis 속 균주가 8% 분리되었다. 서도에 자생하는 갯별꽃에서는 Penicillium 속 균주가 76%, Pestalotiopsis 속 균주가 24% 분리되었고, 율무쑥에서는 Penicillium 속 균주가 81%, Mucor 속 균주가 19% 분리되었다. Bioassay결과, 돌피에서 분리된 Ec-3-1균주가 식물생장촉진활성을 나타내었다. 그리고 독도의 5종류 식물에서 Penicillium 속의 내생진균이 가장 많이 존재함을 알 수 있었다. Endophytic fungi were isolated from the roots of plants growing naturally on the island of Dokdo. Plant samples, such as Miscanthus sinensis, Achyranthus japonica and Echinochloa crusgali were isolated from Dongdo, and those such as Honkenya peploides and Artemsia koidzumii were isolated from Seodo. Twenty one strains of endophytic fungi were isolated from these plants. To identify the strains, PCR (polymerase chain reaction) amplification of the partial ITS (Internal Transcribed Spacer) regions was done with universal primers ITS-1 and ITS-4 to determine the nucleotide sequence of the ITS regions. Of the strains isolated from Miscanthus sinensis, 75% were Penicillium sp. and 25% were Aspergillus sp. Fifty five percent of strains isolated from Achyranthus japonica were Penicillium sp., 30% were Aspergillus sp. and 15% were Zygorhynchus sp. Strains isolated from Echinochloa crusgali were Penicillium sp. (50%), Aspergillus sp. (12%), Giberella sp. (13%), Talaromyces sp. (9%) and Umbelopsis sp. (8%). Of the strains isolated from Honkenya peploides, 76% were Penicillium sp. and 24% were Pestalotiopsis sp. Strains isolated from Artemisia koidzumii were Penicillium sp. (81%) and Mucor sp. (19%). As a result of bioassay, Ec-3-1 strain isolated from Echinochloa crusgalli showed plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp. was the most abundantly distributed fungal strain in all plants used in this study.

Direct Control of Stem Cell Behavior Using Biomaterials and Genetic Factors

<P>Stem cells have recently emerged as an important candidate for cell therapy. However, some major limitations still exist such as a small quantity of cell supply, senescence, and insufficient differentiation efficiency. Therefore, there is an unmet need to control stem cell behavior for better clinical performance. Since native microenvironment factors including stem cell niche, genetic factors, and growth factors direct stem cell fate cooperatively, user-specified <I>in vitro</I> settings are required to understand the regulatory roles and effects of each factor, thereby applying the factors for improved cell therapy. Among others, various types of biomaterials and transfection method have been employed as key tools for development of the <I>in vitro</I> settings. This review focuses on the current strategies to improve stemness maintenance, direct differentiation, and reprogramming using biomaterials and genetic factors without any aids from additional biochemicals and growth factors.</P>

Purpose: Mucopolysaccharidosis type II (MPS II or Hunter syndrome)is a rare lysosomal storage disorder caused by iduronate-2-sulfatase (IDS) deficiency. MPS II causes a wide phenotypic spectrum of symptoms ranging from mild to severe. IDS activity, which is measured in leukocyte pellets or fibroblasts, was reported to be related to clinical phenotype by Sukegawa-Hayasaka et al. Measurement of residual plasma IDS activity using a fluorometric assay is simpler than conventional measurements using skin fibroblasts or peripheral blood mononuclear cells. This is the first study to describe the relationship between plasma IDS activity and clinical phenotype of MPS II. Methods: We hypothesized that residual plasma IDS activity is related to clinical phenotype. We classified 43 Hunter syndrome patients as having attenuated or severe disease types based on clinical characteristics,especially intellectual and cognitive status. There were 27 patients with the severe type and 16 with the attenuated type. Plasma IDS activity was measured by a fluorometric enzyme assay using 4-methylumbelliferyl-α-iduronate 2-sulphate. Results: Plasma IDS activity in patients with the severe type was significantly lower than that in patients with the attenuated type (P=0.006). The optimal cut-off value of plasma IDS activity for distinguishing the severe type from the attenuated type was 0.63 nmol·4 hr-1·mL-1. This value had 88.2% sensitivity, 65.4% specificity, and an area under receiver-operator characteristics (ROC) curve of 0.768 (ROC curve analysis; P=0.003). Conclusion: These results show that the mild phenotype may be related to residual lysosomal enzyme activity.
