<P><B>Abstract</B></P> <P>Tuberculosis (TB), caused by infections of the <I>Mycobacterium tuberculosis</I> (MTB) complex, is the ninth leading cause of death worldwide, and several molecular approaches for MTB species identification and the detection of mutations associated with drug resistance have been developed to date. We previously developed a diagnostic assay for drug susceptibility testing that can detect mutations conferring resistance to anti-TB drugs using allele-specific primer extension on a microsphere-based platform for multiplex polymerase chain reaction. The aim of the present study was to optimize this diagnostic assay based on the evaluation of three methods for extracting mycobacterial DNA from clinical samples. Mycobacterial DNA of 81 samples was digested and decontaminated by <I>N</I>-acetyl-<SMALL>L</SMALL>-cysteine-2% NaOH and then extracted using three methods: “in-house” 5% Chelex-100 chelating resin, InstaGene Matrix, and MagPurix TB DNA Extraction Kit. The former two methods are manual extraction methods, whereas the MagPurix TB DNA Extraction Kit is an automated extraction method used with the MagPurix 12 s automated nucleic acid purification system. The extracted DNA was then subjected to our diagnostic assay, and the results were compared among methods. The magnetic bead method exhibited a higher extraction efficiency and resulted in greater diagnostic efficacy than the two resin-based methods with respect to both target gene detection and acid-fast bacilli smear grades. Therefore, the MagPurix TB DNA Extraction Kit is the optimal MTB DNA extraction method for our diagnostic assay of TB drug susceptibility testing.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Magnetic beads extract DNA of greater purity than that extracted by resin. </LI> <LI> Magnetic bead DNA extraction had higher efficiency in detection of TB genes. </LI> <LI> The automated extraction system is technically simple and completely closed. </LI> <LI> Magnetic bead extraction is optimal for anti-TB drug susceptibility gene testing. </LI> </UL> </P>
Seasonal Dependence of the Effect of Arctic Greening on Tropical Precipitation

<P> This paper examines the seasonal dependence of the effect of Arctic greening on tropical precipitation. In CAM3/CLM3 coupled to a mixed layer ocean, shrub and grasslands poleward of 60°N are replaced with boreal forests. With darker Arctic vegetation, the absorption of solar energy increases, but primarily in boreal spring and summer since little insolation reaches the Arctic in boreal winter. The net energy input into the northern extratropics is partly balanced by southward atmospheric energy transport across the equator by an anomalous Hadley circulation, resulting in a northward shift of the tropical precipitation. In contrast, in boreal fall, the slight increase in insolation over the Arctic is more than offset by increased outgoing longwave radiation and reduced surface turbulent fluxes in midlatitudes, from the warmer atmosphere. As a result, the Northern Hemisphere atmosphere loses energy, which is compensated by a northward cross-equatorial atmospheric energy transport, leading to a southward shift of the tropical precipitation in boreal fall. Thus, although Arctic vegetation is changed throughout the year, its effect on tropical precipitation exhibits substantial seasonal variations. </P>
Developing parametric design fashion products using 3D printing technology

This study created wearable fashion products with parametric design characteristics, using 3D printing technology. The goal of the study was to understand what parametric design features can be simulated with 3D modeling and printing technology, as well as to demonstrate what techniques can be used to produce fashion products using 3D printing technology. This study created two diferent parametric motifs using an FDM-type 3D printer with TPU and ABS as the printing materials. With those motifs, we produced three garments and two accessories. The limitations found during the process were modeling the exact measurement of the motifs that will merge with the apparel design seamlessly while maintaining the parametric features, as well as attaching the printed motifs to fabric without ruining the integrity of the textile. A signifcant implication of this study is that it recreates parametric designs on the human body and utilizes 3D printing technology for fashion products. This paper cast a light on a discussion about the technique can be applied on fashion design with full-sized body and encouraged designers to explore further with technological advancements in the future.
Ship collision avoidance route planning using CRI-based A<sup>*</sup> algorithm

This study presents a novel ship route planning algorithm that takes into account both operational economy and safety by integrating the A<sup>*</sup> algorithm with a collision avoidance algorithm that evaluates the Collision Risk Index (CRI) between the own ship and the target ship. The CRI-based A<sup>*</sup> algorithm defines a penalty zone, allowing the own ship to explore safe routes based on the International Regulations for Preventing Collisions at Sea 1972 (COLREGs) and performs an adaptive and effective node search on an extended local map grid according to various encounter situations. The proposed algorithm is validated through simulations of head-on, fine-broad crossing, converging crossing, and overtaking encounters, indicating an economical and safe optimum route compared to conventional ship domain-based route planning.

An accuracy of estimated distribution is significantly important to compute a performance of systems since the results of the performance is differed from the estimated function. There are some methods to evaluate the similarity between two distribution functions such as the Kullback-Leibler (KL), Jensen-Shannon (JS) divergence, and intersection area. KL is commonly used to measure the difference between two PDFs based on the entropy in the information theory, JS improved an asymmetric problem of KL, and IA (IA) quantifies an intersection area of two PDFs based on the area metric. Three methods have different characteristics and the results of evaluations are different. Therefore, in this study, the accuracies of statistical modeling which estimates a distribution from given data using three methods are compared through statistical simulation tests and case studies for real engineering data. Consequently, the IA can quantify an absolute accuracy and the most intuitive method.
