        Transcriptome analysis of the pheromone glands in Noorda blitealis reveals a novel AOX group of the superfamily Pyraloidea

        Noorda blitealis (Lepidoptera: Pyraloidea: Crambidae) is a major defoliating pest of Moringa trees. Focusing on its mating and reproduction, here we sequenced and analyzed the transcriptome of its pheromone glands (PGs) with a combination of Illumina sequencing, bioinformatics and phylogenetics approaches, coupled with a genomebased analysis. Transcriptome sequencing led to the yields of approximately 162 million clean reads, which were assembled into 60,578 unigenes and 121,692 transcripts, respectively. From the transcriptome, totally 117 genes encoding eight pheromone biosynthesis enzymes and one pheromone degradation enzyme were identified, 90 of which had complete open reading frames. A comparative analysis between PGs and bodies (removing PGs) revealed a large number of differentially expressed genes, including 79 pheromone biosynthesis and degradation related genes. Of the identified genes, NbliDES12 belonging to the △11 desaturase group was likely to a strong candidate for the desaturation of sex pheromones in N. blitealis, as implied by phylogenetic analyses and expression profiles. Finally and most notably, through genome and transcriptome analyses we discovered, for the first time, a novel aldehyde oxidase 6 (AOX6) group of the superfamily Pyraloidea that have been slightly expanded by gene duplications. Moreover, each orthologous AOX group shared highly conserved gene structure. Together, this current study has characterized the genes associated with sex pheromone biosynthesis and degradation from the PG transcriptome of N. blitealis, and more importantly, identifies a novel AOX group of the Pyraloidea.

      • Treatment Efficacy and Prognostic Factors for Huge HCC Based on Barcelona Clinic Liver Cancer Staging

        Objective: To explore the most appropriate treatment for patients with hepatocellular cancer (HCC) >10 cm by using the Barcelona Clinic Liver Cancer (BCLC) classification. Materials and Methods: A total of 124 HCC patients undergoing surgery were selected. Disease-free survival (DFS), overall survival (OS) and prognostic factors were respectively assessed. Results: This study showed that the cumulative 1-, 3-, 5-year survival rates were 79.7%, 59.8% and 41.6% in BCLC-A patients, 76.2%, 9.5% and 0% in BCLC-B patients and 44.9%, 0% and 0% in BCLC-C patients, respectively. The 1-, 3-, 5-year DFS rates were 49%, 24.5% and 9.1% in BCLC-A patients, 7.5%, 0% and 0% in BCLC-B patients, respectively. No BCLC-C patients survived 1 year after surgery. Multivariate analysis indicated that hepatitis B surface antigen (HBsAg), vascular invasion, intra-hepatic metastasis, curative resection, tumor rupture and pathologic differentiation were independent prognostic factors. Conclusions: Surgery is effective and safe for patients with HCC >10 cm with sufficient hepatic reserve.

        VEHICLE DYNAMIC CONTROL ALGORITHM AND ITS IMPLEMENTATION ON CONTROL PROTOTYPING SYSTEM

        A design of controller for vehicle dynamic control(VDC) and its implementation on the real vehicle were introduced. The controller has been designed using a three-degrees-of-freedom(3DOF) yaw plane vehicle, and the control algorithm was implemented on the vehicle by control prototyping system dSPACE. A hybrid control algorithm, which makes full use of the advantages of robust and fuzzy control, was adopted in the control system. Field test results show that the performance of the vehicle handling dynamics with hybrid controller is improved obviously compared to that without VDC and with simple robust controller on skiddy roads(friction coefficients lower than 0.3).

        Mathematical formulation and an improved moth–flame optimization algorithm for parallel two-sided disassembly line balancing based on fixed common stations

        Nowadays, rapid product iterations result in large quantities of end-of-life products. To meet the fast-growing demand for remanufacturing engineering, companies have quickened the standardization and industrialization of waste dissembling. Two-sided disassembly lines can effectively disassemble large-sized products on both sides of the lines, and parallel disassembly lines can disassemble multiple products simultaneously with fewer workstations and higher production efficiency. Combining the two types of disassembly can effectively increase the disassembly efficiency of large-sized products. However, the parallel two-sided disassembly line has not been fully investigated because of the essential complexity of the problem. Therefore, this research introduced the parallel two-sided disassembly line balancing problem with fixed common stations. First, a multi-objective mixed-integer programming model is established to solve the problem for the first time. The model is proved to be correct through small-scale numerical examples. Second, a multi-objective improved moth–flame optimization algorithm is implemented to solve the proposed large-scale problems. The proposed algorithm employs a two-phase decoding approach to design the scheme and a discrete moth for fire operation to search and replace new individuals, and then a restart strategy is introduced to reduce the probability of the population falling into a local optimum. Finally, the algorithm solved extensive disassembly line balancing problems with different layouts, including the straight-line, two-sided, and parallel two-sided, and case studies demonstrated the reliability and validity of the proposed method.

      • DNA Repair Capacity in Peripheral Blood Lymphocytes Predicts Efficacy of Platinum-based Chemotherapy in Patients with Gastric Cancer

        Objective: To investigate the correlation between ERCC1 expression levels in tumor tissue and peripheral blood lymphocytes (PBL) from patients with gastric cancer and assess the relationship between PBL DNA repair rate (DRR) and the efficacy of platinum chemotherapy. Methods: A total of 53 patients with gastric cancer receiving surgery and 20 controls were studied. ERCC1 protein expression in tumour tissue and PBL were determined by immunohistochemical staining. The PBL DRRs of 47 advanced patients and 20 controls were estimated by comet assay. Results: The positive expression rates of ERCC1 were 67. 9%, 56. 6% and 10.0% in tumour tissues, PBLs of gastric cancer patients, and PBLs of the control group. PBL ERCC1 expression correlated with that in tissue (${\chi}^2$=15. 463, p=0.000). Pearson contingency coefficient=0.475). DRRs of cancer patients by tail length (TL) (Z=4. 662, p=0.000) and tail moment (TM) (Z=3. 827, p=0.000) were significantly lower than that of control group. When TL was applied as an indicator, the correlation between DRR and chemotherapy efficacy was significant (Spearman rank correlation r=0.327, p=0.032). Patients with low levels of DRR in PBL presented better short-term efficacy of chemotherapy than those with high levels of DRR. Conclusions: The ERCC1 expression in PBLs may indirectly reflect ERCC1 expression in gastric cancer tissues. Compared with non-cancer populations, patients with gastric cancer may have lower DNA repair capacity. DRR in PBL may predict the short-term efficacy of platinum-based chemotherapy for patients with advanced gastric cancer.

      • MiRNA Synergistic Network Construction and Enrichment Analysis for Common Target Genes in Small-cell Lung Cancer

        Background: Small-cell lung cancer (also known as SCLC) is an aggressive form and untreated patients generally die within about 3 months. To obtain further insight into mechanism underlying malignancy with this cancer, an miRNA synergistic regulatory network was constructed and analyzed in the present study. Method: A miRNA microarray dataset was downloaded from the NCBI GEO database (GSE27435). A total of 546 miRNAs were identified to be expressed in SCLC cells. Then a miRNA synergistic network was constructed, and the included miRNAs mapped to the network. Topology analysis was also performed to analyze the properties of the synergistic network. Consequently, we could identified constitutive modules. Further, common target genes of each module were identified with CFinder. Finally, enrichment analysis was performed for target genes. Results: In this study, a miRNA synergistic network with 464 miRNAs and 2981 edges was constructed. According to the topology analysis, the topological properties between the networks constructed by LC related miRNAs and LC unrelated miRNAs were significantly different. Moreover, a module cilque0 could be identified in our network using CFinder. The module included three miRNAs (hsa-let-7c, hsa-let-7b and hsa-let-7d). In addition, several genes were found which were predicted to be common targets of cilque0. The enrichment analysis demonstrated that these target genes were enriched in MAPK signaling pathways. Conclusions: Although limitations exist in the current data, the results uncovered here are important for understanding the key roles of miRNAs in SCLC. However, further validation is required since our results were based on microarray data derived from a small sample size.

        The complete chloroplast genome of two Syzygium (Myrtaceae) species and comparative analysis with other related species

        Syzygium, a vital genus under Myrtaceae, is aesthetically pleasing and resistant and is used for urban greenery. However, with intensive research in recent years, an increasing number of Syzygium plants have been explored for their medicinal value. Analysing the Syzygium chloroplast (cp) genome can help assess its phylogenetic relationships and provide a theoretical basis for the development and application of Syzygium plants. In this study, we first sequenced the complete cp genomes of Syzygium grijsii and Syzygium rehderianum to investigate their phylogenetic relationships in Myrtaceae. In the copy region and reverse repeat region, 129 genes were detected, including 84 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Preference analysis showed that the eight Syzygium species preferred codons ending in A/U and contained abundant repetitive sequences and SSR sites. The ycf1, rpoC2, matK and ndhB genes had the most RNA editable sites and more genes with Ka/Ks < 1, indicating that species evolution was mainly subject to purifying selection. Comparative analysis of the cp genome showed that the eight species boundaries of Syzygium were similar, but the variability of the LSC and SSC regions was higher than that of the IR region, with 280 sites with Pi > 0.015, showing a high level of nucleotide diversity. Phylogenetic analysis showed that Syzygium grijsii was closely related to Syzygium jambos, and S. rehderianum was close to Plinia aureana. The cp genome and phylogeny of S. grijsii and S. rehderianum provide a theoretical basis for Syzygium species identification and exploitation.

        N6-Methyladenosine modification (m6 A) of circRNA-ZNF638 contributes to the induced activation of SHF stem cells through miR-361-5p/Wnt5a axis in cashmere goats

        Objective: The objective of this study was to investigate the effects of N6-Methyladenosine modification-circRNA-zinc finger protein 638 (m6A-circRNA-ZNF638) on the induced activation of secondary hair follicle (SHF) stem cells with its potential mechanisms in cashmere goats. Methods: The m6A modification of ZNF638 was analyzed using methylation immunoprecipitation with real-time quantitative polymerase chain reaction technique in SHF stem cells. The effects of circRNA-ZNF638 on the induced activation of SHF stem cells in m6A dependence were evaluated through the overexpression of circRNA-ZNF638/its m6Adeficient mutants in circRNA-ZNF638 knockdown SHF stem cells. The competitive binding of miR-361-5p to circRNA-ZNF638/Wnt5a 3′- untranslated region was analyzed through Dual-luciferase reporter assay. Results: The m6A-circRNA-ZNF638 had significantly higher transcription at anagen SHF bulge of cashmere goats compared with that at telogen, as well as it positively regulated the induced activation of SHF-stem cells in cashmere goats. Mechanismly, m6A-circRNA-ZNF638 sponged miR-361-5p to heighten the transcriptional expression of Wnt5a gene in SHFstem cells. We further demonstrated that the internal m6A modification within circRNAZNF638 is required for mediating the miR-361-5p/Wnt5a pathway to regulate the induced activation of SHF stem cells through an introducing of m6A-deficient mutant of circRNAZNF638. Conclusion: The circRNA-ZNF638 contributes the proper induced activation of SHF-stem cells in cashmere goats in m6A-dependent manner through miR-361-5p/Wnt5a axis.
