SDS-PAGE, Crossed Immunoelectrophoresis 및 Immunoblotting을 이용한 Leptospira interrogans 혈청형간 항원 비교

The soluble antigen profiles and antigenic specificities of Leptospira interrogans serovars icterhaemorrhagiae, canicola, pomona and hardjo were examined by SDS-polyacrylamide gel electrophoresis, crossed immunoeletrophoresis and immunoblotting. The profiles of protein, glycoprotein and fraction containing N-acetylglucosamine of 4 serovars were compared. The protein profiles of 4 serovars were very similar except the range of 14,400 to 30,000 daltons. Molecular weight of glycoprotein of L, pomona was lower than other serovars. L canicola showed extra N-acetylglucosamine bands having molecular weight of 82,000 and 90,000 daltons. In crossed immunoelectrophoresis, a close antigenic relationship was found between L icterohaemorrhagiae and L canicola. In immunoblottings conducted with soluble antigens and rabbit antisera of 4 serovars, Leptospira interrogans serovars possessed cross-reactive antigens and serovar-specific antigens. The molecular weights of serovar-specific antigens were 45,000, 82,000 and 90,000, 31,000 and 24,000 daltons in L icterohaemorrhagiae, L canicola, L pomona, and L hardjo, respectively.

Background/Aims: Oxidative stress increases the risk of cardiovascular compli-cations of metabolic syndrome (MetS). This study was conducted to examine the difference in antioxidant capacity according to the presence of MetS, and to char-acterize the association between antioxidant capacity and MetS-related factors. Methods: We used the biological antioxidant potential (BAP) test to estimate antioxidant capacity. The BAP test has recently been used as an indicator of an-tioxidant capacity. We measured BAP levels in 45 patients with MetS (mean age, 44.6 ± 1.1 years) and 47 age- and sex-matched controls (mean age, 42.7 ± 1.1 years). To evaluate the association between antioxidant capacity and MetS, adiponectin, high-sensitivity C-reactive protein (hs-CRP), interleukin-6, tumor necrosis fac-tor- α, and homeostatic model assessment for insulin resistance (HOMA-IR), lin-ear regression and logistic analyses were performed. Results: The mean BAP of the MetS group (1,937.3 ± 36.5 μmol/L) was significantly lower than that of the non-MetS group (2,101.7 ± 29.5 μmol/L). Also, the mean BAP was low in persons having low high density lipoprotein and high triglyceride. Reduced antioxidant capacity was significantly associated with adiponectin, HO-MA- IR and hs-CRP after adjusting for age and sex. The odds ratios for MetS with BAP, log adiponectin, log HOMA-IR, and log hs-CRP were 0.63 (95% confidence interval [CI], 0.49 to 0.82), 0.22 (0.10 to 0.51), 14.24 (4.35 to 46.58), and 1.93 (1.36 to 2.75), respectively. Conclusions: Persons with MetS showed reduced antioxidant capacity. We identi- fied relationships between antioxidant capacity measured by BAP test and MetS, as well as MetS-related factors, such as insulin resistance, hs-CRP, and adiponectin.
Evaluation of Biological Performance in a Newly Developed Single-use Bioreactor, the CELBIC System

Since the SUB(Single-Use Bioreactor)s have several advantages compared with the conventional stainless steel-based bioreactors, such as flexibility in scale, low risk of contamination, reduced labors for cleaning/ sterilization, and validation/regulation issues, the usage of SUBs for cell culture has been increasing continuously, as the bioindustry of biopharmaceuticals (proteins, antibodies, cell therapies and gene therapies products) has expanded dramatically during recent decades. In this study, a newly developed cell culture system, the CELBIC system, driven by multi-directional rocking movement is introduced and evaluated its physical and biological performances as a cell culture device. Three recombinant cell lines, CHO DG44, CHO-S, and CHO-K1, producing proteins were used for experiments, and cell cultures were carried out in four different working volumes of 1 liter, 5 liters, 50 liters, and 250 liters. Those cell cultures as the biological evaluation were performed under the conditions chosen from the results of evaluation of physical performances of the CELBIC system. For comparison, Erlenmeyer flasks and STR bioreactors representing a conventional cell culture device were used as control groups. Productivity was analyzed through ELISA and HPLC analysis methods. In the CELBIC system, cell cultures could be carried out successfully. In addition, cell growth performances were equal or superior to those of the control groups. From the results of biological evaluation, it was found that the CELBIC system is a SUB system easy to scale-up and able to cultivate cells specialized in suspension culture for producing proteins. It is considered that the CELBIC system can be an appealing alternative as a culture device across the biopharmaceutical fields including research and industry.
