Roles of ErbB3-binding protein 1 (EBP1) in embryonic development and gene-silencing control

<P><B>Significance</B></P><P><I>Ebp1</I> deletion causes developmental defects with massive cell death and cessation of cell proliferation through dysregulation of epigenetic gene silencing unit, a Suv39H1/DNMT1 axis. Our study indicates that EBP1 regulates global gene expression via at least 2 mechnaisms. First, EBP1 acts as a transcriptional repressor for DNMT1 gene expression, allowing the escape of the repressive chromatin state. Second, EBP1 binds to the promoter region of the target gene inhibiting the association of DNMT1 with the downstream gene such as <I>Survivin</I>, regulating gene expression. Hence, these findings provide a molecular mechanism of how EBP1 functions in cell survival and transcriptional regulation by modulating epigenetic regulators during development.</P><P>ErbB3-binding protein 1 (EBP1) is implicated in diverse cellular functions, including apoptosis, cell proliferation, and differentiation. Here, by generating genetic inactivation of <I>Ebp1</I> mice, we identified the physiological roles of EBP1 in vivo. Loss of <I>Ebp1</I> in mice caused aberrant organogenesis, including brain malformation, and death between E13.5 and 15.5 owing to severe hemorrhages, with massive apoptosis and cessation of cell proliferation. Specific ablation of Ebp1 in neurons caused structural abnormalities of brain with neuron loss in [Nestin-Cre; <I>Ebp1</I><SUP><I>flox/flox</I></SUP>] mice. Notably, global methylation increased with high levels of the gene-silencing unit Suv39H1/DNMT1 in <I>Ebp1</I>-deficient mice. EBP1 repressed the transcription of <I>Dnmt1</I> by binding to its promoter region and interrupted DNMT1-mediated methylation at its target gene, <I>Survivin</I> promoter region. Reinstatement of EBP1 into embryo brain relived gene repression and rescued neuron death. Our findings uncover an essential role for EBP1 in embryonic development and implicate its function in transcriptional regulation.</P>
A case report of radiation therapy induced uterine rhabdomyosarcoma

In uterine sarcoma, leiomyosarcoma is the most common, with low grade endometrial stromal sarcoma and high grade endometrial stromal sarcoma. Among them, Rhabdomyosarcoma (RMS) is a very rare, aggressive mesenchymal tumor. Usually diagnosed in the pediatric period, it occurs commonly in the head and neck, urinary tract, reproductive organs, limbs and trunk. RMS is subclassified into embryonal, alveolar, anaplastic (previously called pleomorphic), and spindle cell / sclerosing variants. In adult patients, mostly, it occurs in soft tissues of limbs. RMS appears as a rhabdomyosarcomatous differentiation, usually with adenosarcoma, carcinosarcoma, and more rarely as a pure form of RMS. Clinicopathologic information is restricted compared with other gynecologic malignancies because the incidence is very rare. A 71-year-old patient visited the hospital to abdominal discomfort, anorexia and constipation. She was diagnosed as cervical cancer IIIB, squamous cell carcinoma in 2009, and underwent adjuvant concurrent chemoradiation therapy and chemotherapy of cisplatin and etoposide regimen. Followed by, Intracavitary radiotherapy (ICR) was performed with external beam radiation therapy (EBRT) 4,500cGy / 25fractions and parametrial booster 600cGy. The patient has been up until recently with no evidence of disease (NED). The patient underwent abdominopelvic CT and found a huge heterogeneous mass of 20 cm in size. The patient underwent surgery because of suspected recurrent cervical cancer or ovarian cancer. At the time of surgery, the tumor was suspected to be sarcoma rather than cervical cancer or ovarian cancer. However, before chemotherapy, she complained of speech disturbances and facial paralysis. She was referred to a neurologist and underwent brain MRI. Multiple masses on the occipital bone and parietal bone was detected and diagnosed as skull metastasis. PET / CT images were taken under the skull base level, so metastic diseases were not diagnosed preoperatively.

<P>The EMT (epithelial-mesenchymal transition) is involved in fibrosis and cancer, and is regulated by different signalling pathways mediated through soluble factors, actin reorganization and transcription factor actions. Because the tetraspan (also called tetraspanin) TM4SF5 (transmembrane 4 L6 family member 5) is highly expressed in hepatocellular carcinoma and induces EMT, understanding how TM4SF5 expression in hepatocytes is regulated is important. We explored the mechanisms that induce TM4SF5 expression and whether impaired signalling pathways for TM4SF5 expression inhibit the acquisition of mesenchymal cell features, using human and mouse normal hepatocytes. We found that TGFβ1 (transforming growth factor β1)-mediated Smad activation caused TM4SF5 expression and EMT, and activation of the EGFR [EGF (epidermal growth factor) receptor] pathway. Inhibition of EGFR activity following TGFβ1 treatment abolished acquisition of EMT, suggesting a link from Smads to EGFR for TM4SF5 expression. Further, TGFβ1-mediated EGFR activation and TM4SF5 expression were abolished by EGFR suppression or extracellular EGF depletion. Smad overexpression mediated EGFR activation and TM4SF5 expression in the absence of serum, and EGFR kinase inactivation or EGF depletion abolished Smad-overexpression-induced TM4SF5 and mesenchymal cell marker expression. Inhibition of Smad, EGFR or TM4SF5 using Smad7 or small compounds also blocked TM4SF5 expression and/or EMT. These results indicate that TGFβ1- and growth factor-mediated signalling activities mediate TM4SF5 expression leading to acquisition of mesenchymal cell features, suggesting that TM4SF5 induction may be involved in the development of liver pathologies.</P>

This study examines post-listing control systems for pharmaceutical pricing and suggests a reasonable improvement policy in Korea. There are four post-listing control systems for pharmaceutical pricing in Korean national health insurance. When each scheme was introduced, they had their own background and goals. But it is very difficult to understand and predict the timing and extent of the price cuts due to complex system by introducing or revising some schemes without clear policy objectives. At the same time, the social costs are often caused by various confusions, including distribution sites, due to frequent price cuts. For this reason, the current post-listing control system needs to be improved. We investigated the latest trends of the post-listing control system of the major countries that operate a similar system through literatures, website, reports of relevant institutions. From a mid- to long-term point of view, the system should be that can enhance the health of the population by comprehensively reflecting drug prescribing behavior and use status under clear target for drug expenditure management. It should also be developed as a system that can create a healthy industrial ecosystem for the development of the pharmaceutical industry.

커뮤니케이션 기술의 급격한 발달로 많은 사람들이 온라인 환경에서 대화할 수 있게 되었다. 특히 소셜미디어는 활발한 커뮤니케이션이 이루어지는 대표적인 온라인 채널 중 하나이다. 본 연구에서는 소셜 미디어상에서 성공적인 온라인 커뮤니케이션을 가능하게 하는 초기 댓글과 포스트의 역할에 대해 분석한다. 이를 위해, 대화형 소셜 미디어 중 하나인 Reddit 의 하위 커뮤니티들 중 뉴스와 관련된 커뮤니티에서 데이터를 수집하고 분석하였다. 다중회귀 분석 결과, 포스트와 초기 댓글의 선호 지수가 성공적인 온라인 커뮤니케이션에 유의미한 영향을 끼친다는 것을 확인하였다. 또한, 사람들은 포스트가 쉽고 직관적인 내용을 가질 때, 적극적으로 대화에 참여한다는 것을 발견할 수 있었다.
Inactivation of Norovirus by Lemongrass Essential Oil Using a Norovirus Surrogate System

<B>ABSTRACT</B><P>This study investigated the effect of lemongrass essential oil (LGEO) on the infectivity and viral replication of norovirus. Murine norovirus 1 (MNV-1), a surrogate of human norovirus, was preincubated with LGEO and then used to infect RAW 264.7 cells in a plaque reduction assay. LGEO exhibited a significant reduction in MNV-1 plaque formation in both time- and dose-dependent manners. The quantification of viral genome by quantitative real-time PCR showed similar results in line with those of the plaque reduction assay. It was revealed that citral, a single compound in LGEO, showed dramatic reduction in MNV-1 infectivity (−73.09% when using a treatment of 0.02%, v/v). The inhibitory activity of LGEO on viral replication was further investigated in HG23 cells that harbored a human norovirus replicon. LGEO treatment significantly reduced viral replication in HG23 cells, which suggests that LGEO may have dual inhibitory activities that inactivate viral coat proteins required for viral infection and suppress norovirus genome replication in host cells. In animal experiments, oral administration of murine norovirus preincubated with LGEO significantly suppressed virus infectivity in vivo. Collectively, these results suggest that LGEO, in particular the LGEO component citral, inactivates the norovirus and its subsequent replication in host cells. Thus, LGEO shows promise as a method of inhibiting norovirus within the food industry.</P>

Compartment syndrome is a clinical condition associated with decreased blood circulation that can lead to swelling of tissue in limited space. Several factors including lithotomy position, prolonged surgery, intermittent pneumatic compressor, and reperfusion after treatment of arterial thrombosis may contribute to compartment syndrome. However, compartment syndrome rarely occurs after gynecologic surgery. In this case, the patient was diagnosed as compartment syndrome due to reperfusion injury after treatment of arterial thrombosis, which occurred after laparoscopic radical hysterectomy and pelvic lymph node dissection for cervical cancer. Despite its rarity, prevention and identifying the risk factors of complication should be performed perioperatively; furthermore, gynecologist should be aware of the possibility of complications.
