The direct effects of dipeptidyl peptidase-IV (DPP-IV) inhibitors on endoplasmic reticulum (ER) stress-induced apoptosis and inflammation in cardiomyocytes have not been elucidated. H9c2 cell viability, which was reduced by tunicamycin, was increased after DPP-IV inhibitor gemigliptin treatment. Gemigliptin significantly decreased the tunicamycin-mediated increase in glucose regulated protein 78 (GRP78) expression and ER stress-mediated signaling molecules such as protein kinase RNA-like endoplasmic reticulum kinase (PERK)/C-EBP homologous protein (CHOP) and inositol-requiring enzyme 1α (IRE1α)/c-Jun N-terminal kinase (JNK)-p38. Furthermore, gemigliptin effectively induced Akt phosphorylation in a dose-dependent manner. Using flow cytometry and Hoechst staining, we showed that treatment with Akt inhibitor significantly blocked the anti-apoptotic effects mediated by gemigliptin. The reduction in tunicamycin-induced GRP78 level and PERK/CHOP pathway activity by gemigliptin was reversed after treatment with Akt inhibitor. In conclusion, gemigliptin effectively inhibited ER stress-induced apoptosis and inflammation in cardiomyocytes via Akt/PERK/CHOP and IRE1α/JNK-p38 pathways, suggesting its direct protective role in cardiovascular diseases.

Three new MnII-benzoates coordination polymers containing various bipyridyl ligands (3,3'-dipicoylamine (3), 3-methylisoquinoline (4), and 4,4'-dithiopyridine (5)) and a [Mn6(O2CPh)10(μ4-OH)2(CH3OH)3(H2O)].1.5(C4H4N2) cluster (1) have prepared and their structures were determined. The bipyridyl ligands can act as bridging ligands to produce 1-D or 2-D polymeric compounds. The pyrazine produced a Mn6 cluster molecule, and 3-methylisoquinoline did a benzoate-bridged 1-D MnII compound. The Mn6 cluster (1) and 1-D MnII compounds (3) and (4) show antiferromagnetic property. The compounds 1, 3, and 4 have catalyzed efficiently the transesterification of a variety of esters, while 5 has displayed a very slow conversion. The thermal stabilities of these complexes were also examined.

Atomic-layer-deposited Al2O3 films were grown on ultrathin-body In0.53Ga0.47As substrates for III-V compound-semiconductor-based devices. Interface sulfur (S) passivation was performed with wet processing using ammonium sulfide ((NH4)2S) solution, and dry processing using post-deposition annealing (PDA) under a H2S atmosphere. The PDA under the H2S atmosphere resulted in a lower S concentration at the interface and a thicker interfacial layer than the case with (NH4)2S wet-treatment. The electrical properties of the device, including the interface property estimated through frequency dispersion in capacitance, were better for (NH4)2S wet-treatment than the PDA under a H2S atmosphere. They might be improved, however, by optimizing the process conditions of PDA. The PDA under a H2S atmosphere following (NH4)2S wet-treatment resulted in an increased S concentration at the interface, which improved the electrical properties of the devices.

Wall conditioning and impurity monitoring systems are developed in Versatile Experiment Spherical Torus (VEST). As a wall conditioning system, a baking system covering the vacuum vessel wall partially and a glow discharge cleaning (GDC) system using two electrodes with dc and 50kHz power supplies are installed. The GDC system operates with hydrogen and helium gases for both chemical and physical desorption. The impurity monitoring system with residual gas analyzer (RGA), operating at <10-5Torr with a differential pumping system, is installed along with the optical emission spectroscopy (OES) system to monitor the hydrogen and impurity radiation lines. Effects of these wall conditioning techniques are investigated with the impurity monitoring system for ohmic discharges of VEST. The partial baking and He GDC show limited effects on plasma characteristics but sufficient H2 GDC above 4h enables the longer plasma current duration up to ~15ms within 3h from the end of treatment.

The dependencies of the stack number N on perpendicular exchange-biasing (Hex) and coercivity Hc) in [Pd/Co]N and [Pd/Co (or CoFe)]N/FeMn multilayers were investigated. With the help of the careful designs of layer structures, a series of samples whose surface anisotropies have the linear function N was prepared with constant bulk anisotropies. From the experimental data obtained, it was found that Hex does not depend on the surface anisotropy, while Hc shows a strong dependence. Therefore, it is possible to tailor wide ranges of Hc (300-600 Oe) without varying Hex(∼200 Oe) through the single control parameter stack number N.

<P>Phosphatidylinositol 3-kinases (PI3Ks) have recently been implicated in apoptosis and ischemic cell death. We tested the efficacy of early intervention with a peptide PI3K activator in focal cerebral ischemia. After determining the most effective dose (24 mu g/kg) and time window (2 h after MCAO) of treatment, a total of 48 rats were subjected to middle cerebral artery occlusion (MCAO). Diffusion weighted MRI (DWI) was performed 1 h after MCAO and rats with lesion sizes within a predetermined range were randomized to either PI3K activator or vehicle treatment arms. Fluid attenuated inversion recovery (FLAIR) MRI, neurological function, western blots, and immunohistochemistry were blindly assessed. Initial DWI lesion volumes were nearly identical between two groups prior to treatment. However, FLAIR showed significantly smaller infarct volumes in the PI3K activator group compared with vehicle (146 +/- 81 mm(3) and 211 +/- 96 mm(3), p = 0.045) at 48 h. The PI3K activator group also had better neurological function for up to 2 weeks. In addition, PI3K activator decreased the number of TUNEL-positive cells in the peri-infarct region compared with the control group. Western blot and immunohistochemistry showed increased expression of phosphorylated Akt (Ser473) and GSK-3 beta (Ser9) and decreased expression of cleaved caspase-9 and caspase-3. Our results suggest a neuroprotective role of early activation of PI3K in ischemic stroke. The use of DWI in the randomization of experimental groups may reduce bias.</P>

Histones are DNA-binding proteins that assist in DNA packaging and protection. Here, we, for the first time, cloned a novel histone H1 cDNA (638bp) from the goldfish, Carassius auratus. Sequencing revealed that this histone H1 shared 68.1% amino acid identity and 73.9% similarity with that from the rainbow trout, Salmo gairdneri. We successfully expressed a full-length recombinant version (∼20kDa) and a truncated C-terminal fragment (∼6kDa; 61 amino acids) of this histone H1 as a partially soluble form using a maltose binding protein (MBP) fusion strategy in an Escherichia coli expression system. Our results revealed that both these recombinant histone H1 versions had DNA binding and protection functions, and MBP fusion system was an effective way to produce biological functional recombinant histone proteins in E. coli. This novel recombinant histone H1 protein and/or its C-terminal peptide could be used as potential mediators for efficacious gene delivery.
