      • An Image Retrieval Method Based on Visual Dictionary and Saliency Region

        An image retrieval method based on the combination of visual dictionary and region of saliency was proposed in this paper, which aims to increase the accuracy of image retrieval. The image is divided into sampled blocks and the low-level features are extracted from these image blocks. Then a variety of features vectors are taken as the input vector for learning its corresponding visual dictionary respectively by non-negative sparse coding. Spatial information is added into the sparse representations of image by proposing the saliency polling method, and the similarity measure between sparse representation vectors is defined as SED (Squared Euclidean Distance), which considering the same nonzero entries and Euclidean distance of vectors at the same time. Results of experiment carried on Corel and Caltech datasets showed that this method can effectively improve the accuracy of image retrieval compared with the methods of single visual dictionary.

        Serum metabolomics in mice after paraquat posioning

        Backgrounds: Paraquat is toxic to humans and there is no antidote for paraquat poisoning. Moreover, the mechanisms of paraquat toxicity have not been clearly defined. Methods: In this study, we developed a serum metabolomic method using gas chromatography- mass spectrometry (GC-MS) to evaluate the effects of paraquat poisoning on mice. Pattern recognition analysis, including both principal component analysis and cross orthogonal partial least squares-discriminate analysis revealed that paraquat poisoning induced energy/redox cycle and metabolic perturbations. Results: Compared with the control group, the levels of creatinine, citric acid, succinic acid, fumaric acid and glycine in the sub acute paraquat poisoning group increased (P<0.05), while the levels of alpha-Tocopherol, 6-phosphogluconic acid and palmitoleic acid decreased (P<0.05). Conclusion: These findings provide an overview of systematic responses to paraquat exposure and metabolomic insight into the toxicological mechanism of paraquat. Our results demonstrate that the metabolomic method based on GC-MS can be a useful tool to elucidate the mechanism of PQ toxicity, and provide a new direction for its clinical diagnosis and treatment.

      • Expression and Detection of Retinol-Binding Protein-4 Gene of Pig in E. coli

        Retinol-Binding Protein-4 (RBP-4) is a low molecular weight lipocalin, which mainly functions as a carrier for vitamin A. Though liver is the main machinery for synthesis of this protein, it is also detectable in other extrahepatic tissues, for example, ovary, uterus, and placenta. Recent evidences have shown that RBP-4 plays important roles in animal reproduction, for example, promoting the development of uterus and embryo. To the best of our knowledge, our laboratory firstly reported that high level of RBP-4 existed in follicular fluid from follicular cysts in sows. Moreover, we have also found that RBP-4 could be secreted by granulosa cells, and RBP-4 receptor was detected in granulosa cells. However, there is no any evidence on the role of RBP-4 in regulating the follicular development. Therefore, cloning and expression of RBP-4 and preparation of polyclonal antibody could help us to explore the role of RBP-4 in follicular development. The aim of this work was to construct prokaryotic expression system of swine RBP-4 gene. The total RNA was extracted from swine’s normal ovarian tissue. The sequence including the whole length of RBP-4 was amplified by RT-PCR and inserted into pEASY-E1.Then transformed into E. coli BL21(DE3)pLysS after gene sequencing. Three hours later, adding IPTG with the final concentration of 1mmol/L and inducing five hours. After centrifugation, the supernatant was discarded. By adding Glucose to Luria-Bertani broth, the expressions of protein were increased. SDS-PAGE showed that the RBP-4 gene expressed in the form of inclusion body with a molecular weight of 21KD. Western-Blot results showed that the target protein could be specifically recognized by mouse anti-human monoclonal antibody. Prokaryotic expression vector of RBP-4 gene was successfully established, and the gene was successfully expressed n E. coli, which is ready for purification and RBP-4 polyclone antibody. Meanwhile, these results were beneficial to investigate the function of RBP-4 in follicular development.

      • Multi-objective Decision-making Method of Green Product in Manufacturing Based on Entropy-TOPSIS Method

        The green products in manufacture industry still differ from each other in terms of economics, technology, environmental friendliness, though they all reach the criteria of limitation. This paper expounds the necessity for product choice optimization and provides a model based on information entropy to judge different advantages of product groups. According to Entropy Weight Method and Technique for Order Preference by Similarity to Ideal Solution (TOPSIS), the basic theory of green product evaluation model is established. The index system includes selected evaluation indicators of green products, considering about the evaluation index of the critical values to construct a hierarchy criterion, which uses entropy to objectively determine the weight vector of each evaluation index. Then, combined with TOPSIS analysis approach and calculates the pros and cons of green products. It is concluded that the evaluation results based on the Entropy-TOPSIS evaluation model are consistent with the actual results, and the results are consistent with the evaluation results of fuzzy mathematics. The method comprehensively considered many influential factors of the green products, to avoid the limitation of single criterion, and the importance of various factors are analyzed and compared, the prediction result is more scientific, as the theoretical basis of green products, and provide a reliable guidance method for product improvement. This is objective to empower the law, which also fully embodies the idea of variable weights and overcome the past green products that exist in the evaluation process homogeneity empowering. That means fixed weights lead to the limitation of a lack of flexible evaluation.

        Adaptable Center Detection of a Laser Line with a Normalization Approach using Hessian-matrix Eigenvalues

        In vision measurement systems based on structured light, the key point of detection precision is to determine accurately the central position of the projected laser line in the image. The purpose of this research is to extract laser line centers based on a decision function generated to distinguish the real centers from candidate points with a high recognition rate. First, preprocessing of an image adopting a difference image method is conducted to realize image segmentation of the laser line. Second, the feature points in an integral pixel level are selected as the initiating light line centers by the eigenvalues of the Hessian matrix. Third, according to the light intensity distribution of a laser line obeying a Gaussian distribution in transverse section and a constant distribution in longitudinal section, a normalized model of Hessian matrix eigenvalues for the candidate centers of the laser line is presented to balance reasonably the two eigenvalues that indicate the variation tendencies of the second-order partial derivatives of the Gaussian function and constant function, respectively. The proposed model integrates a Gaussian recognition function and a sinusoidal recognition function. The Gaussian recognition function estimates the characteristic that one eigenvalue approaches zero, and enhances the sensitivity of the decision function to that characteristic, which corresponds to the longitudinal direction of the laser line. The sinusoidal recognition function evaluates the feature that the other eigenvalue is negative with a large absolute value, making the decision function more sensitive to that feature, which is related to the transverse direction of the laser line. In the proposed model the decision function is weighted for higher values to the real centers synthetically, considering the properties in the longitudinal and transverse directions of the laser line. Moreover, this method provides a decision value from 0 to 1 for arbitrary candidate centers, which yields a normalized measure for different laser lines in different images. The normalized results of pixels close to 1 are determined to be the real centers by progressive scanning of the image columns. Finally, the zero point of a second-order Taylor expansion in the eigenvector’s direction is employed to refine further the extraction results of the central points at the subpixel level. The experimental results show that the method based on this normalization model accurately extracts the coordinates of laser line centers and obtains a higher recognition rate in two group experiments.

        The induction of apoptosis by a newly synthesized diosgenyl saponin through the suppression of estrogen receptor-α in MCF-7 human breast cancer cells.

        <P>Estrogen receptor (ER)-α is an important therapeutic target in the clinical treatment of breast cancer. A potential down-regulator of ER-α, diosgenyl α-L-rhamnopyranosyl-(12)-[β-D-xylopyranosyl-(14)]-α-L-arabinopyranoside is a newly synthesized diosgenyl saponin named compound 22. This study evaluated the in vitro mechanism of compound 22 as an anticancer agent for breast cancer. Our results indicated that compound 22 selectively inhibited proliferation and induced apoptosis in ER-positive MCF-7 cells, compared with ER-negative MDA-MB-231 and MCF-10A cells. Western blot analysis showed that compound 22 decreased the expression of procaspase-3, procaspase-8, and survivin; and increased the expression of Fas ligand and cleaved PARP1 in MCF-7 cells, indicating that compound 22-induced apoptosis was mediated by the extrinsic pathway. This apoptosis was associated with the suppression of ER-α protein and mRNA expression and the inhibition of ER-DNA binding to the estrogen responsive element. Moreover, ER-α mediated gene expression such as c-Myc and cyclin D1 was reduced, and the activation of p38 and ERK 1/2 was significantly decreased after treatment with compound 22 in MCF-7 cells. Taken together, these results demonstrate that compound 22 down-regulates ER-α expression and induces apoptosis through the extrinsic pathway, suggesting that compound 22 may be effective in the treatment of ER-positive breast cancer.</P>

        HOXD Antisense Growth-Associated Long Noncoding RNA Promotes Triple-Negative Breast Cancer Progression by Activating Wnt Signaling Pathway

        Purpose: Triple-negative breast cancer (TNBC) is the most lethal subtype of breast cancer owing to high heterogeneity, aggressive nature, and lack of treatment options, which has a substantial deleterious effect on patients' lives. HOXD antisense growth-associated long noncoding RNA (lncRNA) (HAGLR) plays tumor-promoting roles in many cancers. In this study, we aimed to explore the role of HAGLR in TNBC. Methods: Quantitative real-time polymerase chain reaction assays were used to examine the expression of RNAs. Functional experiments were conducted to test the biological behavior of TNBC cells. Moreover, MS2-RNA immunoprecipitation, luciferase reporter, and RNA pull-down assays were conducted to verify the binding relationship between HAGLR, microRNA-143-5p (miR-143-5p), and serine- and arginine-rich splicing factor 1 (SRSF1). Results: HAGLR was found to be highly expressed in TNBC tissues and cells, and inhibiting HAGLR suppressed cell proliferation, migration, and invasion and promoted cell apoptosis in TNBC. Meanwhile, miR-93-5p was shown to bind to HAGLR and SRSF1. In addition, SRSF1 plays an oncogenic role in TNBC. Importantly, HAGLR could activate the Wnt signaling pathway by sponging miR-93-5p to upregulate SRSF1; thus, accelerating TNBC progression. Conclusion: HAGLR could promote the progression of TNBC through the miR-93-5p/SRSF1 axis to activate the Wnt signaling pathway.

        Extraction of polysaccharides under vacuum condition from Lentinus edodes stipe and their antioxidant activities in vitro

        To improve the efficiency of polysaccharideextraction, vacuum technology was used to breakdown thecell wall to extract polysaccharides from Lentinus edodesstipe, and the optimal parameters were optimized byresponse surface methodology. Then the antioxidantactivities in vitro of the polysaccharides were evaluated. Results showed that the optimal conditions for polysaccharideextraction were temperature of 62 C, vacuum of0.08 MPa, stirring speed of 1200 r/min, time of 25 min andmaterial/liquid ratio of 1:26 (g/mL). Under these conditions,the yield of polysaccharide was 4.28%. The decolorizationrate and deproteinization rate of polysaccharidestreated by kaolin and hydrogen peroxide were higher thanthat of activated carbon. However, there was no significantinfluence on the polysaccharide retention rate, that wasabout 80%. Results also indicated that polysaccharidetreated with activated carbon could obtain higher antioxidantactivity.

        M2-like macrophages transplantation protects against the doxorubicin-induced heart failure via mitochondrial transfer

        Aims: The alternatively activated macrophages have shown a cardioprotective effect in heart failure. However, the effect of M2 adoptive transfer in non-ischemic heart failure is unknown. In this study, we evaluated the efficacy of M-CSF plus IL-4 induced M2-like macrophages transplantation in doxorubicin-induced cardiotoxicity. Methods: Bone marrow mononuclear cells were polarized as CCR2+CD206+ M2-like macrophages by a combination of M-CSF plus IL-4 treatment. C57BL/6 mice received a single intraperitoneal njection of doxorubicin (15 mg/kg). The treatment group were treated with M2-like macrophages (1 × 10^6 cells per mouse; i.v.) once a week for 2 weeks. After 3 weeks, we examined the percentage of resident cells and cardiac function. Furthermore, we evaluated cardiac fibrosis, cardiomyocyte apoptosis and circulating inflammatory factors. Finally, we investigated the mitochondria transfer in vitro in a direct and indirect co-culture conditions. Results: Cardiac function was significantly improved in doxorubicin-induced heart failure by adoptive transfer of M2-like macrophages. Besides, M2-like macrophages treatment attenuated cardiac fibrosis and cardiomyocyte apoptosis, as well as increased the level of circulating IL-4 and Th2 response. In vitro, M2-like macrophages could transfer mitochondria to injured cardiomyocytes in a direct and indirect way. Conclusions: In our study, adoptive transfer of M2-like macrophages could protect against the doxorubicin-induced cardiotoxicity, which may be partly attributed to mitochondria transfer. And M2-like macrophages transplantation could become a treatment for non-ischemic heart failure in the clinical practice.
