        Dynamic Path Planning Based on Fuzzy and Behavior Control Idea

        In order to solve dynamic path planning problem of multi-robot systems in unknown dynamic environment, a kind of method based on fuzzy and behavior control idea was proposed in this paper. This method designs two fuzzy controllers, which are danger degree fuzzy controller and velocity fuzzy controller. They took fully into account the position and the velocity of the obstacle and the azimuth of the target. Fuzzy rules were designed based on behavior control idea and which reflected move-to-goal behavior, avoid-obstacle behavior and follow-obstacle behavior. The experimental results show that the proposed method is feasible and valid and is suitable for the dynamic and complicated environment in particularly.

        Metabolic Engineering of Saccharomyces cerevisiae to Improve Glucan Biosynthesis

        β-Glucan is a chief structural polymer in the cell wall of yeast. β-Glucan has attracted intensive attention because of its wide applications in health protection and cosmetic areas. In the present study, the β-glucan biosynthesis pathway in S. Cerevisiae was engineered to enhance β-glucan accumulation. A newly identified bacterial β-1, 6-glucan synthase GsmA from Mycoplasma agalactiae was expressed, and increased β-glucan content by 43%. In addition, other pathway enzymes were investigated to direct more metabolic flux towards the building of β-glucan chains. We found that overexpression of Pgm2 (phosphoglucomutase) and Rho1 (a GTPase for activating glucan synthesis) significantly increased β-glucan accumulation. After further optimization of culture conditions, the β-glucan content was increased by 53.1%. This study provides a new approach to enhance β-glucan biosynthesis in Saccharomyces cerevisiae.

      • Lack of Association between the COMT rs4680 Polymorphism and Ovarian Cancer Risk: Evidence from a Meta-analysis of 3,940 Individuals

        Catechol-O-methyltransferase (COMT) is involved in estrogen metabolism and is vital to estrogen-induced carcinogenesis, including that of ovarian cancer. Although many recent epidemiologic studies have investigated associations between the COMT rs4680 polymorphism and ovarian cancer risk, the results remain inconclusive. We therefore performed a meta-analysis to derive a more precise estimate of associations. Systematic searches of the PubMed, Embase, Web of Science, Cochrane Library, Wanfang, China National Knowledge Infrastructure, and Chinese Biomedicine databases were undertaken to retrieve eligible studies. Odds ratios (ORs) with their corresponding 95% confidence intervals (CIs) were pooled to assess the strength of the association. In total, 8 case-control studies involving 1,293 cases and 2,647 controls were included in the meta-analysis. Overall, the results showed no evidence of significant association between the COMT rs4680 polymorphism and ovarian cancer risk in any of the assessed genetic models. Subgroup analyses by ethnicity also did not reveal any significant association in any genetic model (p>0.05). In conclusion, our findings suggest that the COMT rs4680 polymorphism may not contribute to the risk of ovarian cancer.

      • Blocking Bcl-2 Leads to Autophagy Activation and Cell Death of the HEPG2 Liver Cancer Cell Line

        Background: Apoptosis may be induced after Bcl-2 expression is inhibited in proliferative cancer cells. This study focused on the effect of autophagy activation by ABT737 on anti-tumor effects of epirubicin. Methods: Cytotoxic effects of ABT737 on the HepG2 liver cancer cell line were assessed by MTT assay and cell apoptosis through flow cytometry. Mitochondrial membrane potential was measured by fluorescence microscopy. Monodansylcadaverin (MDC) staining was used to detect activation of autophagy. Expression of p53, p62, LC3, and Beclin1, apoptotic or autophagy related proteins, was detected by Western blotting. Results: ABT737 and epirubicin induced growth inhibition in HepG2 cells in a dose- and time-dependent manner. Both ABT737 and epirubicin alone could induce cell apoptosis with a reduction in mitochondrial membrane potential as well as increased apoptotic protein expression. Further increase of apoptosis was detected when HepG2 cells were co-treated with ABT373 and epirubicin. Furthermore, our results demonstrated that ABT373 or epirubicin ccould activate cell autophagy with elevated autophagosome formation, increased expression of autophagy related proteins and LC3 fluorescent puncta. Conclusions: ABT737 influences cancer cells through both apoptotic and autophagic mechanisms, and ABT737 may enhance the effects of epirubicin on HepG2 cells by activating autophagy and inducing apoptosis.

        Improved 3D Semantic Segmentation Model Based on RGB Image and LiDAR Point Cloud Fusion for Automantic Driving

        LiDAR point cloud semantic segmentation algorithm is crucial to the environmental understanding of unmanned vehicles. At this stage, in autonomous vehicles, effectively integrating the complementary information of LiDAR and camera has become the focus of research. In this work, a network framework (called PI-Seg) for LiDAR point clouds semantic segmentation by fusing appearance features of RGB images is proposed. In this paper, the perspective projection module is introduced to align and synchronize point clouds with images to reduce appearance information loss. And an efficient and concise dual-flow feature extraction network is designed, a fusion module based on a continuous convolution structure is used for feature fusion, which effectively reduces the amount of parameters and runtime performance, and more suitable for autonomous driving scenarios. Finally, the fused features are added to the LiDAR point cloud features as the final output features, and the point cloud category label prediction is realized through the MLP network. The experimental results demonstrate that PI-Seg has a 5.3% higher mIoU score than SalsaNext, which is also a projection-based method, and still has a 1.4% performance improvement compared with the latest Cylinder3D algorithm, and in quantitative analyses the mAP value also has the best performance, showing that PI-Seg is better than other existing methods.

        Patterns and drivers of Vibrio isolates phylogenetic diversity in the Beibu Gulf, China

        Members of the genus Vibrio are ubiquitous in aquatic environments and can be found either in a culturable or a viable but nonculturable (VBNC) state. Despite widespread concerns as to how to define the occurrence and dynamics of Vibrio populations by culture-independent approaches, further physiological research and relevant biotechnological developments will require the isolation and cultivation of the microbes from various environments. The present work provides data and perspectives on our understanding of culturable Vibrio community structure and diversity in the Beibu Gulf. Finally, we isolated 1,037 strains of Vibrio from 45 samples and identified 18 different species. Vibrio alginolyticus, V. cyclitrophicus, V. tasmaniensis, V. brasiliensis, and V. splendidus were the dominant species that had regional distribution characteristics. The correlation between the quantitative distribution and community structure of culturable Vibrio and environmental factors varied with the Vibrio species and geographical locations. Among them, salinity, nitrogen, and phosphorus were the main factors affecting the diversity of culturable Vibrio. These results help to fill a knowledge gap on Vibrio diversity and provide data for predicting and controlling pathogenic Vibrio outbreaks in the Beibu Gulf.

        Tetrabromobisphenol A (TBBPA) generation and removal paths analysis in printed circuit board (PCB) industrial wastewater: Lab-scale investigations

        With the rapid development of printed circuit board (PCB) industry, the problem of the release of tetrabromobisphenol A (TBBPA) during PCB manufacturing processes was recognized and harmful to the receiving water body. In this study, TBBPA generation paths, fates in industrial wastewater in the PCB manufacturing processes were analyzed. Gas chromatograph-mass spectrometer (GC-MS) scanning results showed that the raw materials including solder mask ink, character ink and dry film mainly contained TBBPA. The PCB fabrication procedures, mainly including development, etching, stripping of inner layer manufacturing process, and pattern plating, tin-plating of outer layer manufacturing process, and development during the forming and surface treatment process would introduce TBBPA into wastewater. Flocculation and activated sludge process (ASP) that were commonly used for industrial wastewater treatment were selected to assess their removal capability for TBBPA in the wastewater, both which could remove more than 80% TBBPA. Low pH below 7, increased PAC and PAM dosages, and proper dosing during the slow-stirring stage could enhance the TBBPA removal of flocculation-sedimentation process. The ASP removes TBBPA mainly by the path of adsorption. High activated sludge concentration, low pH, medium-temperature and limited competitive adsorbates (NH4+, Cu2+) were beneficial to remove TBBPA by the ASP.

        Effects of Myogenin on Expression of Late Muscle Genes through MyoD-Dependent Chromatin Remodeling Ability of Myogenin

        MyoD and myogenin (Myog) recognize sets of distinct but overlapping target genes and play different roles in skeletal muscle differentiation. MyoD is sufficient for near-full expression of early targets, while Myog can only partially enhance expression of MyoD-initiated late muscle genes. However, the way in which Myog enhances the expression of MyoD-initiated late muscle genes remains unclear. Here, we examine the effects of Myog on chromatin remodeling at late muscle gene promoters and their activation within chromatin environment. Chromatin immunoprecipitation (ChIP) assay showed that Myog selectively bound to the regulatory sequences of late muscle genes. Overexpres-sion of Myog was found to overcome sodium butyrate-inhibited chromatin at late muscle genes in differ-entiating C2C12 myoblasts, shifting the transcriptional activation of these genes to an earlier time period. Furthermore, overexpression of Myog led to increased hyperacetylation of core histone H4 in differentiating C2C12 myoblasts but not NIH3T3 fibroblasts, and hyperacetylated H4 was associated directly with the late muscle genes in differentiating C2C12, indicating that Myog can induce chromatin remodeling in the presence of MyoD. In addition, co-immunopre-cipitation (CoIP) revealed that Myog was associated with the nuclear protein Brd4 in differentiating C2C12 myoblasts. Together, these results suggest that Myog enhances the expression of MyoD-initiated late muscle genes through MyoD-dependent ability of Myog to induce chromatin remodeling, in which Myog-Brd4 interaction may be involved.

        Tuning the Mechanical Properties of Silkworm Silk Fibres by Thermally Induced Modification of Crystalline Nanostructure

        Antheraea pernyi and Bombyx mori silk fibres, the most important and the subjects of several studies, have attractedwidespread attention due to their exceptional mechanical properties and promising applications. However, the determinantsbehind the variations of mechanical property between these two fibres still remains unknown. In this paper, the nanocrystallinestructures of these two silk fibres were tuned by thermal treatment in order to understand how the structure affects themechanical properties of fibres. It has been found that, along with an increase in temperature for thermal treatment, theincrease in the intramolecular β-sheet leads to progressive increase in longitudinal modulus and radial hardness, whilst thereduction in crystallinity results in great reduction in breaking tensile strength. The yield strength is proportional to theaverage crystallite size which increases along with the annealing temperature. Compared to B. mori, A. pernyi silk fibres havemore obvious temperature-dependant structural changes. These findings explain how size effects can be exploited to createbioinspired materials with tuneable mechanical properties.
