        Mechanism of the anti-liver fibrosis effect of Periplaneta americana extracts that promote apoptosis of HSC-T6 cells through the Bcl-2/Bax signaling pathway

        Periplaneta americana L. (Blattidae) extract (PA-B) possesses anti-oxidant, anti-inflammatory, and hep atoprotective properties; however, its mechanism of action in liver fibrosis remains unclear. Herein, we inves tigated the hepatoprotective effect and mechanism of action of PA-B. HSC-T6 cells treated with PA-B were stimulated with TGF-β1, and immunofluorescence, western blotting, Hoechst 33,342 staining, confocal laser scanning microscopy, and flow cytometry were performed. Furthermore, the effects of PA-B were verified in a rat model of hepatic fibrosis established using 40 % carbon tetrachloride (CCl 4 ). PA-B induced apoptosis of HSC-T6 cells by inhibiting proliferation (maximum inhibition ratio 99.08 %), reducing mitochondrial membrane po tential, and ameliorating apoptosis-related proteins (P < 0.01). PA-B (120 mg/kg) significantly alleviated CCl 4 -induced hepatic histopathological fibrosis and reduced the levels of aspartate aminotransferase (from 27.68 to 20.58 U/L) and alanine aminotransferase (from 16.88 to 5.93 U/L; P < 0.05 or P < 0.01). Immunohistochemistry and western blotting showed that α-SMA levels were reduced (from 0.49 to 0.23) and that the Bcl/Bax signaling pathway was significantly inhibited by PA-B (120 mg/kg; P < 0.05 or P < 0.01). PA-B exhibits anti-fibrotic effects by mediating the Bcl-2/Bax pathway and promoting HSC-T6 cell apoptosis, providing a basis for its development as a drug to treat liver fibrosis.

        Cinnamaldehyde Derivatives Inhibit Coxsackievirus B3-Induced Viral Myocarditis

        The chemical property of cinnamaldehyde is unstable in vivo, although early experiments have shown its obvious therapeutic effects on viral myocarditis (VMC). To overcome this problem, we used cinnamaldehyde as a leading compound to synthesize derivatives. Five derivatives of cinnamaldehyde were synthesized: 4-methylcinnamaldehyde (1), 4-chlorocinnamaldehyde (2), 4-methoxycinnamaldehyde (3), α-bromo-4-methylcinnamaldehyde (4), and α-bromo-4-chlorocinnamaldehyde (5). Neonatal rat cardiomyocytes and HeLa cells infected by coxsackievirus B3 (CVB3) were used to evaluate their antiviral and cytotoxic effects. In vivo BALB/c mice were infected with CVB3 for establishing VMC models. Among the derivatives, compound 4 and 5 inhibited the CVB3 in HeLa cells with the half-maximal inhibitory concentrations values of 11.38 ± 2.22 μM and 2.12 ± 0.37 μM, respectively. The 50% toxic concentrations of compound 4 and 5-treated cells were 39-fold and 87-fold higher than in the cinnamaldehyde group. Compound 4 and 5 effectively reduced the viral titers and cardiac pathological changes in a dose-dependent manner. In addition, compound 4 and 5 significantly inhibited the secretion, mRNA and protein expressions of inflammatory cytokines TNF-α, IL-1β and IL-6 in CVB3-infected cardiomyocytes, indicating that brominated cinnamaldehyde not only improved the anti-vital activities for VMC, but also had potent anti-inflammatory effects in cardiomyocytes induced by CVB3.

      • IGF-1 from Adipose-Derived Mesenchymal Stem Cells Promotes Radioresistance of Breast Cancer Cells

        Purpose: The aim of this study was to investigate effects of adipose-derived mesenchymal stem cells (AMSCs) on radioresistance of breast cancer cells. Materials and Methods: MTT assays were used to detect any influence of AMSC supernatants on proliferation of breast cancer cells; cell migration assays were used to determine the effect of breast cancer cells on the recruitment of AMSCs; the cell survival fraction post-irradiation was assessed by clonogenic survival assay; ${\gamma}$-H2AX foci number post-irradiation was determined via fluorescence microscopy; and expression of IGF-1R was detected by Western blotting. Results: AMSC supernatants promoted proliferation and radioresistance of breast cancer cells. Breast cancer cells could recruit AMSCs, especially after irradiation. IGF-1 derived from AMSCs might be responsible for the radioresistance of breast cancer cells. Conclusions: Our results suggest that AMSCs in the tumor microenvironment may affect the outcome of radiotherapy for breast cancer in vitro.

      • Relationship Between the SER Treatment Period and Prognosis of Patients with Small Cell Lung Cancer

        Purpose: To explore the relationship between SER (time between the start of any treatment and the end of radiation therapy) and the survival of patients with limited-stage small cell lung cancer. Materials and Methods: Between 2008 and 2013, 135 cases of limited-stage small cell lung cancer (LS-SCLC) treated with consecutively curative chemoradiotherapy were included in this retrospective analysis. In terms of SER, patients were divided into early radiotherapy group (SER<30 days, n=76) and late radiotherapy group ($SER{\geq}30$ days, n=59) with a cut-off of SER 30 days. Outcomes of the two groups were compared for overall survival. Results: For all analyzable patients, median follow-up time was 23.8 months and median overall survival time was 16.8 months. Although there was no significant differences in distant metastasis free survival between the two groups, patients in early radiotherapy group had a significantly better PFS (p=0.003) and OS (p=0.000). Conclusions: A short SER may be a good prognostic factor for LD-SCLC patients treated with concurrent chemoradiotherapy.

        Canonical transient receptor potential channels and their modulators: biology, pharmacology and therapeutic potentials

        Canonical transient receptor potential channels(TRPCs) are nonselective, high calcium permeability cationicchannels. The TRPCs family includes TRPC1, TRPC2,TRPC3, TRPC4, TRPC5, TRPC6, and TRPC7. These channelsare widely expressed in the cardiovascular and nervoussystems and exist in many other human tissues and celltypes, playing several crucial roles in the human physiologicaland pathological processes. Hence, the emergenceof TRPCs modulators can help investigate these channels’applications in health and disease. It is worth noting that theTRPCs subfamilies have structural and functional similarities,which presents a signifi cant diffi culty in screening anddiscovering of TRPCs modulators. In the past few years,only a limited number of selective modulators of TRPCswere detected; thus, additional research on more potent andmore selective TRPCs modulators is needed. The presentreview focuses on the striking desired therapeutic eff ectsof TRPCs modulators, which provides intel on the structuralmodifi cation of TRPCs modulators and further pharmacologicalresearch. Importantly, TRPCs modulators cansignifi cantly facilitate future studies of TRPCs and TRPCsrelated diseases.

        Stereo-digital image correlation in the behavior investigation of CFRP-steel composite members

        The application of carbon fiber reinforced polymer (CFRP) in steel structures primarily includes two categories, i.e., the bond-critical application and the contact-critical application. Debonding failure and buckling failure are the main failure modes for these two applications. Conventional electrometric techniques may not provide precise results because of the limitations associated with single-point contact measurements. A nondestructive full-field measurement technique is a valuable alternative to conventional methods. In this study, the digital image correlation (DIC) technique was adopted to investigate the bond behavior and buckling behavior of CFRP-steel composite members. The CFRP-to-steel bonded joint and the CFRPstrengthened square hollow section (SHS) steel column were tested to verify the suitability of the DIC technique. The stereo- DIC technique was utilized to measure continuous deformation. The bond-slip relationship of the CFRP-to-steel interface was derived using the DIC data. Additionally, a multi-camera DIC system consisting of four stereo-DIC subsystems was proposed and applied to the compressive test of CFRP-strengthened SHS steel column. The precise buckling location and CFRP delamination of the CFRP-strengthened SHS steel column were identified. The experimental results confirm that the stereo-DIC technique can provide effective measurements for investigating the behaviors of CFRP-steel composite members.

      • Liposome-mediated Induction of Apoptosis of Human Hepatoma Cells by C-Myc Antisense Phosphorothioate Oligodeoxynucleotide and 5-Fluorouracil

        Background: The aim of this study was to investigate the effect of a c-myc antisense oligodeoxynucleotide and 5-fluorouracil on the expression of c-myc, invasion and proliferation of HEPG-2 liver cancer cells. Materials and Methods: HEPG-2 cells were treated with lipiosome-mediated c-myc ADSON and 5-fluorouracil. The proliferation inhibition rate and invasion were measured by MTT and invasion assay, respectively. Cell apoptosis was detected by flow cytometry and expression of c-myc by RT-PCR and immunohistochemistry. Results: The proliferation inhibition rate was significantly higher in the antisense oligodeoxynucleotide added-5-fluorouracil group than single antisense oligodeoxynucleotide or 5-fluorouracil group (p<0.05). G0/G1 cells in the antisense oligodeoxynucleotide group and S cells in the 5-fluorouracil groups were significantly increased than that in the control group, respectively (P<0.01). The amplification strips of PCR products in 5-FU, ASODN and combination groups were significantly weaker than that in the control group (P<0.01). The percentage of c-myc-protein-positive cells were significantly lower in antisense oligodeoxynucleotide, 5-fluorouracil and combination groups than that in the control group (P<0.01). Conclusions: A liposome-mediated c-myc antisense oligodeoxynucleotide and 5-fluorouracil can inhibit the proliferation and invasion of liver cancer cells by reducing the expression of c-myc. A c-myc antisense oligodeoxynucleotide can increase the sensitivity of liver cancer cells to 5-fluorouracil and decrease the dosage of the agent necessary for efficacy, providing an experimental basis for the clinical therapy of liver cancer.

        Voice activity detection (VAD) is the key of voice recognition, voice synthesis and speech-sound enhancement.For the sake of improve the accuracy and robustness of speech endpoint detection system. Combining the advantages of adaptive genetic algorithm (AGA) and improved radial basis function network (RBF) defects in existing learning methods. This paper presents a comprehensive detection method-- Adaptive genetic algorithm radial basis function network. This method uses adaptive genetic algorithm to simultaneously optimize the center, the width and the structure of RBF network. The method using wavelet analysis to extract the characteristics of the speech signal, use them as an input amount to the radial basis function networks. Establish voice detection system model, this method enhance the accuracy of the detection system and has better robustness.

        Expression profiles of circular RNAs in sheep skeletal muscle

        Objective: Circular RNAs (circRNAs) are a newfound class of non-coding RNA in animals and plants. Recent studies have revealed that circRNAs play important roles in cell proliferation, differentiation, autophagy and apoptosis during development. However, there are few reports about muscle development-related circRNAs in livestock. Methods: RNA sequencing analysis was employed to identify and annotate circRNAs from longissimus dorsi of sheep. Reverse transcription followed by real-time quantitative (q) polymerase chain reaction (PCR) analysis verified the presence of these circRNAs. Targetscan7.0 and miRanda were used to analyse the interaction of circRNA-microRNA (miRNA). To investigate the function of circRNAs, an experiment was conducted to perform enrichment analysis hosting genes of circRNAs using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) pathways. Results: About 75.5 million sequences were obtained from RNA libraries of sheep skeletal muscle. These sequences were mapped to 729 genes in the sheep reference genome. We identified 886 circRNAs, including numerous circular intronic RNAs and exonic circRNAs. Reverse transcription PCR (RT-PCR) and DNA sequencing analysis confirmed the presence of several circRNAs. Real-Time RT-PCR analysis exhibited resistance of sheep circRNAs to RNase R digestion. We found that many circRNAs interacted with muscle-specific miRNAs involved in growth and development of muscle, especially circ776. The GO and KEGG enrichment analysis showed that hosting genes of circRNAs was involved in muscle cell development and signaling pathway. Conclusion: The study provides comprehensive expression profiles of circRNAs in sheep skeletal muscle. Our study offers a large number of circRNAs to facilitate a better understanding of their roles in muscle growth. Meanwhile, we suggested that circ776 could be analyzed in future study.
