      • Association of Six Susceptibility Loci with Prostate Cancer in Northern Chinese Men

        Background/Aim: Six prostate cancer (PCa) susceptibility loci were identified in a genome-wide association study (GWAS) in populations of European decent. However, the associations of these 6 single-nucleotide polymorphisms (SNPs) with PCa has remained tobe clarified in men in Northern China. This study aimed to explore the loci associated with PCa risk in a Northern Chinese population. Methods: Blood samples and clinical information of 289 PCa patients and 288 controls from Beijing and Tianjin were collected. All risk SNPs were genotyped using polymerase chain reaction (PCR)-high resolution melting curve technology and gene sequencing. Associations between PCa and clinical covariates (age at diagnosis, prostate-specific antigen [PSA], Gleason score, tumor stage, and level of aggressiveness) and frequencies of alleles and genotypes of these SNPs were analyzed using genetic statistics. Results: Among the candidate SNPs, 11p15 (rs7127900, A) was associated with PCa risk (P = 0.02, odds ratio [OR] = 1.64, 95% confidence interval [CI] = 1.09-2.46). Genotypes showed differences between cases and controls on 11p15 (rs7127900, A), 11q13 (rs7931342, T), and HNF1B (rs4430796, A) (P = 0.03, P = 0.01, and P = 0.04, respectively). The genotype TG on 11q13 (rs7931342, T) was positively associated with an increased Gleason score (P = 0.04, OR = 2.15, 95% CI = 1.02-4.55). Patients carrying TG on 17q24 (rs1859962, G) were negatively associated with an increased body mass index (BMI) (P = 0.03, OR = 0.44, 95% CI = 0.21-0.92) while those with AG on HNF1B (rs4430796, A) were more likely to have PSA increase (P = 0.002). Conclusion: Our study suggests that 11p15 (rs7127900, A) could be a susceptibility locus associated with PCa in Northern Chinese. Genotype TG on 11q13 (rs7931342, T) could be related to an increased Gleason score, AG on HNF1B (rs4430796, A) could be associated with PSA increase, and TG on 17q24 (rs1859962, G) could be negatively associated with an increased BMI in Chinese men with PCa.

        Discriminating Eggs from Two Local Breeds Based on Fatty Acid Profile and Flavor Characteristics Combined with Classification Algorithms

        This study discriminated fatty acid profile and flavor characteristics of Beijing You Chicken (BYC) as a precious local breed and Dwarf Beijing You Chicken (DBYC) eggs. Fatty acid profile and flavor characteristics were analyzed to identify differences between BYC and DBYC eggs. Four classification algorithms were used to build classification models. Arachidic acid, oleic acid (OA), eicosatrienoic acid, docosapentaenoic acid (DPA), hexadecenoic acid, monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), unsaturated fatty acids (UFA) and 35 volatile compounds had significant differences in fatty acids and volatile compounds by gas chromatographymass spectrometry (GC-MS) (p<0.05). For fatty acid data, k-nearest neighbor (KNN) and support vector machine (SVM) got 91.7% classification accuracy. SPME-GC-MS data failed in classification models. For electronic nose data, classification accuracy of KNN, linear discriminant analysis (LDA), SVM and decision tree was all 100%. The overall results indicated that BYC and DBYC eggs could be discriminated based on electronic nose with suitable classification algorithms. This research compared the differentiation of the fatty acid profile and volatile compounds of various egg yolks. The results could be applied to evaluate egg nutrition and distinguish avian eggs.

        Low Parameter Sensitivity Deadbeat Direct Torque Control for Surface Mounted Permanent Magnet Synchronous Motors

        In order to decrease the parameter sensitivity of deadbeat direct torque control (DB-DTC), an improved deadbeat direct torque control method for surface mounted permanent-magnet synchronous motor (SPMSM) drives is proposed. First, the track errors of the stator flux and torque that are caused by model parameter mismatch are analyzed. Then a sliding mode observer is designed, which is able to predict the d-q axis currents of the next control period for one-step delay compensation, and to simultaneously estimate the model parameter disturbance. The estimated disturbance of this observer is used to estimate the stator resistance offline. Then the estimated resistance is required to update the designed sliding-mode observer, which can be used to estimate the inductance and permanent-magnetic flux linkage online. In addition, the flux and torque estimation of the next control period, which is unaffected by the model parameter disturbance, is achieved by using predictive d-q axis currents and estimated parameters. Hence, a low parameter sensitivity DB-DTC method is developed. Simulation and experimental results show the validity of the proposed direct control method.

      • rs10505474 and rs7837328 at 8q24 Cumulatively Confer Risk of Prostate Cancer in Northern Han Chinese

        Aims: Genome-wide association studies (GWAS) have identified several risk variants for prostate cancer (pCa) mainly in Europeans, which need to be further verified in other racial groups. We selected six previously identified variants as candidates and to define the association with PCa in Northern Han Chinese. Methods: 749 subjects from Beijing and Tianjin in Northern China were included. Six variants (rs10505474, rs7837328, rs4242384, rs7813, rs486907 and rs1058205) were genotyped by high resolution melting (HRM) assays. The individual and cumulative contribution for of the risk of PCa and clinical covariates were analyzed. Results: Among the six candidate variants, onlyrs10505474, and rs7837328, both locating at 8q24 region, were associated with PCa in our population.rs10505474 (A) was associated with PCa ($OR_{recessive}=1.56$, p=0.006); and rs7837328 (A) was associated with PCa ($OR_{dominant}=1.38$, p=0.042/$OR_{recessive}=1.99$, p=0.003). Moreover, we observed a cumulative effects between them ($p_{trend}=2.58{\times}10^{-5}$). The joint population attributable risk showed the two variants might account for 71.85% of PCa risk. In addition, we found the homozygotes of rs10505474 (A) and rs7837328 (A) were associated with PCa clinical covariants (age at onset, tumor stage, respectively) ($p_{age}=0.046$, $P_{tumorstage}=0.048$). Conclusion: rs10505474 (A) and rs7387328 (A) at 8q24 are associated with PCa and cumulatively confer risk, suggesting the two variations could determine susceptibility to PCa in the Northern Chinese Han population.

        Reliable control of magnetic vortex chirality in asymmetrically optimized magnetic nanodisk

        Magnetic vortex has attracted attention in the field of information storage because their topological spin structures with chiral bistable states. If the vortex core polarity and vortex circulation sense can be controlled simultaneously in a nanodisk, which will be more beneficial to realize the multi-bit ultrahigh density storage. In this paper, a reliable control scheme for magnetic vortex chirality is proposed by optimizing the structure of Pac- Man-like nanodisk. The results show that the polarity and circulation of the vortex can be controlled simultaneously by changing the direction of the global magnetic field, and even the chiral states of the vortex can be determined by detecting the stray field distribution on the surface of the nanodisk. The optimized Pac-Man-like nanodisk provide an experimental method for the control and detection of magnetic vortex chirality, which will be beneficial to the realization of multi-bit magnetic storage or magnetic logic technology in the future.

        Bauschinger Effect or Kinematic Hardening: Bridging Microstructure and Continuum Mechanics

        As a strong and low-priced austenite stabilizer, nitrogen (0.085 wt%) was deliberately added into a hot-rolled Fe-0.2C-5Mn TRIP steel for optimizing the strength–ductility balance. Nitrogen addition can not only increase austenite fraction (> 50%), but also favor the improvement of austenite stability due to its partition behavior from ferrite to austenite during intercritical annealing. Besides, the low annealing temperature (635 °C) and nitrogen addition result in the simultaneous precipitation of cementite (size, 21–396 nm) and carbonitride (size, 7–193 nm) in ferrite and at grain boundaries, which can effectively increase the yield strength through precipitation strengthening (33–63 and 41–61 MPa, respectively). Furthermore, the sufficient interstitial solute originated from N addition as well as the discontinuous TRIP effect benefits the strong serrated flow behavior and multi-peak strain hardening behavior. As a result, the strong TRIP effect together with precipitation strengthening is believed to be responsible for the excellent strength–ductility balance of present steel annealed for 3 h, demonstrating a high tensile strength of 1043 MPa, outstanding total elongation of 43.3% and superior product of strength and elongation (PSE) of 44.8 GPa%.

        Melatonin Attenuates Noise Stress-induced Gastrointestinal Motility Disorder and Gastric Stress Ulcer: Role of Gastrointestinal Hormones and Oxidative Stress in Rats

        Background/Aims There are increasing evidences for gastrointestinal motility disorder (GIMD) and gastric stress ulcer induced by noise stress. The present study was to investigate the reversed effect of melatonin on GIMD and gastric stress ulcer induced by noise stress and potential mechanism. Methods Noise stress was induced on rats, and melatonin (15 mg/kg) was administered to rats by intraperitoneal injection. Differences were assessed in gastric residual rate (GRR), small intestine propulsion rate (SPR), Guth injury score, cortisol, gastrointestinal hormones (calcitonin-gene-related peptide and motilin) and oxidative stress markers (superoxide dismutase and malondialde hyde) in blood plasma as well as gastric mucosa homogenate with or without melatonin. The pathological examination of gastric mucosa was also performed. Results The GRR and SPR were improved by noise stress compared with control (P < 0.05). The pathological examination and Guth injury score revealed gastric stress ulcer. Moreover, the levels of cortisol, motilin and malondialdehyde in blood plasma and malondialdehyde in gastric mucosa homogenate were increased by noise stress (P < 0.05). CGRP and superoxide dismutase activity in both of blood plasma and gastric mucosa homogenate were significantly decreased (P< 0.05). Furthermore, melatonin reversed changes in GRR, SPR, pathological examination, Guth injury score, cortisol, motilin, CGRP, superoxide dismutase activity and malondialdehyde (P < 0.05). Conclusions Melatonin is effective in reversing the GIMD and gastric stress ulcer induced by noise stress. The underlying mechanism may be involved in oxidative stress and gastrointestinal hormones.
