      • Effect of Trichostatin A on CNE2 Nasopharyngeal Carcinoma Cells - Genome-wide DNA Methylation Alteration

        Trichostatin A (TSA) is a histone deacetylase (HDAC) inhibitor. We here investigated its effects on proliferation and apoptosis of the CNE2 carcinoma cell line, and attempted to establish genome-wide DNA methylation alteration due to differentially histone acetylation status. After cells were treated by TSA, the inhibitory rate of cell proliferation was examined with a CCK8 kit, and cell apoptosis was determined by flow cytometry. Compared to control, TSA inhibited CNE2 cell growth and induced apoptosis. Furthermore, TSA was found to induce genome-wide methylation alteration as assessed by genome-wide methylation array. Overall DNA methylation level of cells treated with TSA was higher than in controls. Function and pathway analysis revealed that many genes with methylation alteration were involved in key biological roles, such as apoptosis and cell proliferation. Three genes (DAP3, HSPB1 and CLDN) were independently confirmed by quantitative real-time PCR. Finally, we conclude that TSA inhibits CNE2 cell growth and induces apoptosis in vitro involving genome-wide DNA methylation alteration, so that it has promising application prospects in treatment of NPC in vivo. Although many unreported hypermethylated/hypomethylated genes should be further analyzed and validated, the pointers to new biomarkers and therapeutic strategies in the treatment of NPC should be stressed.

        Transcriptome profiling of PeCRY1 transgenic Populus tomentosa

        Cryptochromes, a class of blue light photoreceptors, play vital roles in regulating growth and development in higher plants. Despite their control over various important traits, there have been few studies focusing on cryptochromes in forest trees to date. In this study, the Euphrates poplar (Populus euphratica) gene PeCRY1 (cryptochrome 1 of Populus euphratica) was isolated and heterologously expressed in Populus tomentosa. Three biological replicates of each of the PeCRY1 transgenic P. tomentosa (CRY1) and wild-type (WT) plants were processed for transcriptome profiling. We found 34792 commonly expressed transcripts among the 93868 detected unigenes. Using R package DESeq, we identified 357 differentially expressed genes (DEGs), including 132 upregulated and 225 downregulated genes. Gene ontology (GO) enrichment and KEGG pathway enrichment were used to better understand the functions of detected DEGs. Biosynthetic processes, such as starch and sucrose metabolism, which are closely related to growth and development, were highly enriched. Organic cyclic compound biosynthesis was downregulated, whereas carbohydrate metabolism was upregulated. Through KEGG pathway enrichment, we observed that the pentose phosphate pathway, photosynthesis, and circadian rhythm were significantly enriched. Another method of expression analysis based on quantitative reverse transcription polymerase chain reaction (qRT-PCR) validated our RNA sequencing (RNA-seq) results.

        Effects of maternal dietary energy restriction on breast muscle fibre development in the offspring of broiler breeders

        Objective: The effects of maternal dietary energy levels on breast muscle fibre development in offspring of broiler breeders were investigated. Methods: A total of 480 20-week-old Arbor Acres (AA) healthy female broiler breeders, with an average body weight of 2.33±0.01 kg, were randomly divided into 4 treatment groups with 6 replicates and 20 broiler breeders for each replicate and fed a corn and soybean meal diet with 100%, 80%, 70%, and 50% energy levels, respectively. Approximately 300 eggs per treatment were collected for incubation for 6 days. Then, 120 0-day-old female broilers at each energy level were randomly selected and divided into 6 replicates with 20 broilers for each replicate, with this experimental phase with the offspring lasting for 49 days. Results: Compared with the 100% energy group, the breast muscle fibre diameter at embryonic day 21 in the 80% energy group was significantly reduced (p<0.05). In the 80% energy group, the muscle fibre density of the breast increased significantly (p<0.05) at embryonic days 15 and 21. The breast muscle fibre diameter of the offspring in each group was significantly decreased (p<0.05) on the 1st day. The breast muscle sarcomere length of the embryos in the 80% energy group was significantly higher (p<0.05) than those in the 70% and 50% energy groups. Compared with the 100% energy group, the expression of the myostatin gene in the offspring was significantly decreased (p<0.05). Conclusion: In conclusion, the effects of a maternal dietary energy level of 80% in this study were found to be optimal for breast muscle fibre development in offspring, which indicated that the metabolic energy level of AA broilers of 9.36 MJ/kg for the mid-term diet for laying eggs has a more practical significance.

        SMAD4 Suppresses AURKA-Induced Metastatic Phenotypes via Degradation of AURKA in a TGFβ-Independent Manner

        <P>SMAD4 has been suggested to inhibit the activity of the WNT/β-catenin signaling pathway in cancer. However, the mechanism by which SMAD4 antagonizes WNT/β-catenin signaling in cancer remains largely unknown. Aurora A kinase (AURKA), which is frequently overexpressed in cancer, increases the transcriptional activity of β-catenin/T-cell factor (TCF) complex by stabilizing β-catenin through the inhibition of GSK-3β. Here, SMAD4 modulated AURKA in a TGFβ-independent manner. Overexpression of SMAD4 significantly suppressed AURKA function, including colony formation, migration, and invasion of cell lines. In addition, SMAD4 bound to AURKA induced degradation of AURKA by the proteasome. A luciferase activity assay revealed that the transcriptional activity of the β-catenin/TCF complex was elevated by AURKA, but decreased by SMAD4 overexpression. Moreover, target gene analysis showed that SMAD4 abrogated the AURKA-mediated increase of β-catenin target genes. However, this inhibitory effect of SMAD4 was abolished by overexpression of AURKA or silencing of AURKA in SMAD4-overexpressed cells. Meanwhile, the SMAD4-mediated repression of AURKA and β-catenin was independent of TGFβ signaling because blockage of TGFβR1 or restoration of TGFβ signaling did not prevent suppression of AURKA and β-catenin signaling by SMAD4. These results indicate that the tumor-suppressive function of SMAD4 is mediated by downregulation of β-catenin transcriptional activity via AURKA degradation in a TGFβ-independent manner.</P><P><B>Implications:</B> SMAD4 interacts with AURKA and antagonizes its tumor-promoting potential, thus demonstrating a novel mechanism of tumor suppression. <I>Mol Cancer Res; 12(12); 1779–95. ©2014 AACR</I>.</P>

        Fault Diagnosis and Fault Tolerant Control for Manipulator with Actuator Multiplicative Fault

        In this paper, a new fault diagnosis and fault tolerant control algorithm for manipulators with actuator multiplicative fault is proposed. The dynamic model of the manipulator with disturbance is taken as the research object. When faults occur in the actuator, a nonlinear observer based on radial basis function (RBF) neural network is used to estimate the fault information. After the fault information is obtained, an adaptive back-stepping sliding mode controller is used to control the manipulator to reach the desired trajectory. At last, an illustrated example is given to demonstrate the efficiency of the proposed algorithm, and satisfactory results have been obtained.

        Five Forces Model of Computational Power: A Comprehensive Measure Method

        In this paper, a model is proposed to comprehensively evaluate the computational power. The five forces model of computational power solves the problem that the measurement units of different indexes are not unified in the process of computational power evaluation. It combines the bidirectional projection method with TOPSIS method. This model is more scientific and effective in evaluating the comprehensive situation of computational power. Lastly, an example shows the validity and practicability of the model.

      • Cathepsin K Activity Controls Injury-Related Vascular Repair in Mice

        <P>Cathepsin K (CatK) is one of the most potent mammalian collagenases. We showed previously the increased expression of CatK in human and animal atherosclerotic lesions. Here, we hypothesized that ablation of CatK mitigates injury-induced neointimal hyperplasia. Male wild-type (CatK<SUP>+/+</SUP>) and CatK-deficient (CatK<SUP>−/−</SUP>) mice underwent ligation or a combination of ligation and polyethylene cuff-replacement injuries to the right common carotid artery just proximal to its bifurcation, and they were then processed for morphological and biochemical studies at specific time points. On operative day 28, CatK<SUP>−/−</SUP> significantly reduced neointimal formation and neovessel formation in both single- and combination-injured arteries compared with the Cat K<SUP>+/+</SUP> mice. At early time points, CatK<SUP>−/−</SUP> reduced the lesion macrophage contents and medial smooth muscle cell proliferation, the mRNA levels of monocyte chemoattractant protein-1, toll-like receptor-2, toll-like receptor-4, chemokine ligand-12, and the gelatinolytic activity related to matrix metalloproteinase-2/-9. An aorta-explant assay revealed that smooth muscle cell movement was impaired in the CatK<SUP>−/−</SUP> mice compared with the CatK<SUP>+/+</SUP> mice. In addition, the smooth muscle cells and macrophages from CatK<SUP>−/−</SUP> mice had less invasive ability through a reconstituted basement membrane barrier. This vasculoprotective effect was mimicked by Cat inhibition with <I>trans</I>-epoxysuccinyl-L-leucylamido-{4-guanidino} butane (E64<I>d</I>). These results demonstrate an essential role of CatK in neointimal lesion formation in response to injury, possibly via the reduction of toll-like receptor-2/-4–mediated inflammation and smooth muscle cell proliferation, suggesting a novel therapeutic strategy for the control of endovascular treatment–related restenosis by regulating CatK activity.</P>

        Sex Comb on Midleg Like-2 Accelerates Hepatocellular Carcinoma Cell Proliferation and Metastasis by Activating Wnt/β-Catenin/EMT Signaling

        Purpose: The purpose of this study was to investigate the influences of sex comb on midleg like-2 (SCML2) on hepatocellular carcinoma (HCC) and potentially related mechanisms. Materials and Methods: SCML2 expression in tumor tissues and cells was analyzed using the TCGA database and/or qRT-PCR. The proliferation of HCC cells was detected by CCK-8, colony formation, and EdU assays. The migration and invasion of HCC cells were detected by transwell and wound healing assays. Apoptosis of HCC cells was determined by flow cytometry. Additionally, qRT-PCR and Western blot were used to detect the expression of SCML2 and Wnt/β-catenin/epithelial–mesenchymal transition (EMT) signaling. A xenograft model in mice was established to verify the in vitro findings. Results: We found that SCML2 was highly expressed in HCC tissues and cells and that high expression of SCML2 was correlated with poor prognosis in HCC patients. SCML2 overexpression promoted proliferation, invasion, and migration and repressed apoptosis of HCC cells. The reverse results were obtained in SCML2-silenced cells. Further, we found that SCML2 activated the Wnt/β-catenin/EMT pathway. SCML2 silencing reduced the protein levels of Wnt3a, β-catenin, N-cadherin, Vimentin, and Snail and enhanced E-cadherin protein expression both in vivo and in vitro. Conclusion: SCML2 silencing inhibits the proliferation, migration, and invasion of HCC cells by regulating the Wnt/β-catenin/EMT pathway.

        Robust Actuator Fault Reconstruction for Takagi-Sugeno Fuzzy Systems with Time-varying Delays via a Synthesized Learning and Luenberger Observer

        This paper addresses the problem of robust actuator fault reconstruction for Takagi-Sugeno (T-S) fuzzy systems subjects to actuator faults, unknown inputs and time-varying delays via a Fuzzy Synthesized Learning and Luenberger Observer (FSL2O). Through a coordinate transformation, the original T-S fuzzy system is decomposed into two subsystems: subsystem-1 effected by actuator faults and subsystem-2 effected by unknown inputs. In the presented FSL2O methodology, a Reduced-Order Fuzzy Learning Observer (ROFLO) is explored for the subsystem1 to reconstruct actuator faults accurately while a Reduced-Order Fuzzy State (Luenberger) Observer (ROFSO) is designed for the subsystem-2 based on the H∞ control technique such that it has strong robustness against the unknown inputs. The synthesized design of the ROFLO and the ROFSO is formulated in a unified manner in terms of Linear Matrix Inequalities (LMIs) that can be conveniently solved using LMI optimization technique. In addition, as a comparison, a full-order FLO is suggested for robust actuator fault reconstruction. Finally, a numerical exampleand simulation are provided to demonstrate the effectiveness of the proposed approaches.

        Synergistic Effect of Copper and Cobalt in Cu-Co-O Composite Nanocatalyst for Catalytic Ozonation

        A novel Cu-Co-O composite nanocatalyst was designed and prepared for the ozonation of phenol. A synergistic effect of copper and cobalt was observed over the Cu-Co-O composite nanocatalyst, which showed higher activity than either copper or cobalt oxide alone. In addition, the Cu-Co-O composite revealed good activity in a wide initial pH range (4.11-8.05) of water. The fine dispersion of cobalt on the surface of copper oxide boosted the interaction between catalyst and ozone, and the surface Lewis acid sites on the Cu-Co-O composite were determined as the active sites. The Raman spectroscopy also proved that the Cu-Co-O composite was quite sensitive to the ozone. The trivalent cobalt in the Cu-Co-O composite was proposed as the valid state.
