        MiR-339 attenuates LPS-induced intestinal epithelial cells inflammatory responses and apoptosis by targeting TLR4

        Background Intestinal epithelial cells are important for defending against pathogen infection. LPS is an endotoxin that is highly antigenic and cytotoxic produced by bacteria. LPS disrupts the intestine epithelium integrity and induced the intestinal epithelial cell infammation and apoptosis. Our previous study has predicted the function of exosome miRNAs through bioinformatics analysis, and we found that miR-339 had a potential function in cell infammation response. To our knowledge, no published paper has demonstrated the miR-339 function in protecting the intestine epithelium against bacterial infection. Objective The objective of this study is to evaluate the miR-339 function in regulating intestinal epithelial cells to defend against bacterial infection through biological experiments and bioinformatics analyses. Methods Through the miR-339 transfection experiment and TLR4 interfering experiment, we evaluated the function of miR339 and TLR4 in the process of infammatory responses and apoptosis. Through Bioinformatics analyses and dual-luciferase reporter experiment, we identifed the target gene of miR-339. Results miR-339 attenuates LPS-induced intestinal epithelial cells infammatory responses through the TLR4/NF-κB signaling pathway and inhibited LPS-induced apoptosis through the P53 signaling pathway. TLR4 is the target gene of miR-339. TLR4 reduced LPS-induced proinfammatory responses and apoptosis. Conclusions In conclusion, miR-339 protected the intestine epithelial cells from LPS-induced cell infammation and apoptosis through targeting TLR4. This study expanded our understanding of how miRNAs and genes work collaboratively in regulating intestinal epithelial cells to defend against bacterial infection.

        Functional response of Wolbachia-infected and uninfected Trichogramma dendrolimi Matsumura (Hymenoptera: Trichogrammatidae) to Asian corn borer, Ostrinia furnacalis Guenée (Lepidoptera: Pyralidae) eggs

        Trichogramma dendrolimi Matsumura (Hymenoptera: Trichogrammatidae) are haplo-diploid egg parasitoid wasps widely used as biological control agents against the Asian corn borer, Ostrinia furnacalis Guenée (Lepidoptera: Pyralidae). Two reproductive modes have been found in this species. Typically, female wasps produce haploid male offspring from unfertilized eggs and diploid female offspring from fertilized eggs (bisexual). However, they can also produce only diploid females when they are infected with Wolbachia, though the eggs are unfertilized (thelytokokous). To investigate the potential of a thelytokous Wolbachia-infected (TdW+) and a bisexual uninfected (Td) T. dendrolimi strain as inundative biocontrol agents, the functional responses of both strains toward their natural enemy (Asian corn borer) were evaluated at three constant temperatures (20, 25 and 30 °C). The results revealed two types of functional response for both strains: type II and III. TdW+strain displayed type II at 30 °C (high temperature) and III at 20 °C (low temperature) and 25 °C, while Td strain displayed type II at 20 °C and III at 25 °C and 30 °C. The comparison of functional response between two strains indicated TdW+ and Td strain had no significant difference at 20 °C. However, the estimated host handling time of the TdW+ strain was significantly shorter than that of the Td strain at 25 °C, and TdW+ strain had significantly higher instantaneous search rate and shorter handling time than Td strain at 30 °C. Shorter host handling time and higher instantaneous search rate in thelytokous Wolbachia-infected T. dendrolimi showed that this strain might be more effective for controlling Asian corn borer.

        Five Forces Model of Computational Power: A Comprehensive Measure Method

        In this paper, a model is proposed to comprehensively evaluate the computational power. The five forces model of computational power solves the problem that the measurement units of different indexes are not unified in the process of computational power evaluation. It combines the bidirectional projection method with TOPSIS method. This model is more scientific and effective in evaluating the comprehensive situation of computational power. Lastly, an example shows the validity and practicability of the model.
